MORPHOMETRICAL AND STEREOLOGICAL ANALYSIS OF THE SUPERIOR CERVICAL GANGLION OF Rattus norvegicus SUBMITTED TO CHRONIC TREATMENT WITH CORTISOL International Journal of Morphology
Cortez,C. M.; Costa,W. S.; Babinski,M. A.; Chagas,M. A..
The present work investigates the corticoid action on the growth of the superior cervical ganglion of the rat and describes the cortisol effect during early stages of development. The study was based on morphometric and stereological analysis of the perikarya. Eight rats were treated intraperitoneally with cortisol (1mg/Kg/day) during 36 days. Treatment was initiated in the 8th day after birth and was withdrawn one day before the sacrifice. There was a significant difference (<=P0,05) for the neural mean diameter between the control group (16.78 ± 1.11mm) and treated animals (15.84 ±0.99mm). The decrease of perikarya neuronal diameter was also demonstrated by stereological methods. Morphometrical findings may suggest alterations in superior cervical...

Vitamin C and endogenous cortisol in foreign-body inflammatory response in pacus PAB
Belo,Marco Antonio de Andrade; Moraes,Julieta Rodini Engrácia de; Soares,Vando Edésio; Martins,Maurício Laterça; Brum,Camila Dantas; Moraes,Flávio Ruas de.
The objective of this work was to evaluate the effect of food supplementation with vitamin C on macrophage and multinucleated giant cell (MGC) activities of pacus at two stocking densities. The experiment was carried out in a 2x2x3 split-plot factorial arrangement with: 0 and 500 mg kg-1 vitamin C; 5 and 20 kg m-3 stocking densities; and evaluation times at 3, 6, and 12 days after the subcutaneous implantation of glass coverslips (DPI). The number of macrophages and MGC, as well as cortisol and glucose plasma levels were determined. The number of macrophages and MGC with two to five nuclei was significantly greater in fish supplemented with vitamin C at 5 kg m-3 stocking density at 3 DPI in comparison to nonsupplemented ones. The macrophage and MGC counts...

Quantitative computed tomography of the liver in dogs submitted to prednisone therapy Arq. Bras. Med. Vet. Zootec.
Costa,L.A.V.S.; Oliveira,D.C.; Lopes,B.F.; Lanis,A.B.; Teixeira,M.W.; Costa,F.S..
The aim of this study is to evaluate the possible alterations in radiodensity of the hepatic parenchyma and spleen resulting from the corticotherapy with prednisone, correlating these findings with serum biochemical hepatic transaminase activity. A clinical trial was performed on eight healthy dogs with the intent of evaluating possible hepatic alterations after prednisone therapy using quantitative computed tomography. All animals received prednisone orally at a dose of 2mg/kg of weight for 30 days. The radiodensity of the liver and spleen was determined before and after the period that the medication was administered. An increase in the radiodensity of the hepatic parenchyma between the initial and final experimental stages was observed. In the present...

MDCK cells express serotonin-regulable 11ß-hydroxysteroid dehydrogenase type 2 Biocell
Zallocchi,Marisa L.; Damasco,Maria C.; Calvo,Juan C.; Lantos,Carlos P.; Matkovic',Laura B..
Prior to this work, we found that adrenal as well as extra-adrenal factors activate the response of renal 11ß-hydroxysteroid dehydrogenase 2 to stressful situations. These results -showing ways through which the organism hinders the pathological occupation of mineralocorticoid receptors by glucocorticoids leading to sodium retention and hypertension- prompted the present study on the nature of the above-mentioned extraadrenal factors. Serotonin was chosen because of its properties as a widely distributed neurohormone, known to interact with glucocorticoids at many sites, also exhibiting increased levels and effects under stressful situations. We studied serotonin effects on 11b-hydroxysteroid dehydrogenase 2 activity in a cell line derived from distal...

Insulin signaling proteins in pancreatic islets of insulin-resistant rats induced by glucocorticoid Biol. Res.
De Paula,Flávia MM; Boschero,Antonio C; Carneiro,Everardo M; Bosqueiro,José R; Rafacho,Alex.
Chronic administration of glucocorticoids induces insulin resistance that is compensated by an increase in p-cell function and mass. Since insulin signaling is involved in the control of p-cell function and mass, we investigated the content of insulin pathway proteins in pancreatic islets. Rats were made insulin resistant by daily administration of dexamethasone (1mg/kg, b.w., i.p.) for 5 consecutive days (DEX), whilst control rats received saline (CTL). Circulating insulin and insulin released from isolated islets were measured by radioimmunoassay whereas the content of proteins was analyzed by Western blotting. DEX rats were hyperinsulinemic and exhibited augmented insulin secretion in response to glucose (P < 0.01). The IRa-subunit, IRS-1, Shc, AKT,...

Perinatal development and adult blood pressure BJMBR
Ashton,N..
A growing body of evidence supports the concept of fetal programming in cardiovascular disease in man, which asserts that an insult experienced in utero exerts a long-term influence on cardiovascular function, leading to disease in adulthood. However, this hypothesis is not universally accepted, hence animal models may be of value in determining potential physiological mechanisms which could explain how fetal undernutrition results in cardiovascular disease in later life. This review describes two major animal models of cardiovascular programming, the in utero protein-restricted rat and the cross-fostered spontaneously hypertensive rat. In the former model, moderate maternal protein restriction during pregnancy induces an increase in offspring blood...
