Fibrinolytic protease production by new Streptomyces sp: DPUA 1576 from Amazon lichens Electron. J. Biotechnol.

Background Streptomyces sp. DPUA 1576 from Amazon lichens was studied to protease and fibrinolytic production. A 2² factorial experimental design was applied to optimize its protease enzyme production using two independent variables, namely soybean flour and glucose concentrations. Results The optimal conditions to obtain high protease production (83.42 U/mL) were 1.26% soybean flour and 1.23% glucose concentration. A polynomial model was fitted to correlate the relationship between the two variables and protease activity. In relation to fibrinolytic activity, the highest activity of 706.5 mm² was obtained at 1.7% soybean flour and 1.0% glucose concentration, which was 33% higher than plasmin. Production and stability studies of the bioemulsifier obtained from a new strain of Candida glabrata UCP 1002 Electron. J. Biotechnol.

Evaluation of both tenso-active and emulsifying activities indicated that a biosurfactant was produced by the newly isolated and promising strain Candida glabrata isolated from mangrove sediments. The extracellular water-soluble emulsifying agent was isolated and identified as a heteropolymer. The maximum of bioemulsifier production was observed when the strain was grown on soluble and insoluble substrates cotton seed oil plus glucose, reaching values of 10.0 g/l after 144 hrs at 200 rpm. The cell-free culture broth containing the examined agent lowered the surface tension of the medium to 31 mN/m. В экспериментальных условиях исследовано влияние голодания на направленность метаболических процессов в тканях двустворчатого моллюска-вселенца Anadara inaequivalvis (Bruguiere, 1789). Длительность эксперимента – 18 сут. Показано, что на начальных этапах голодания (6 сут.) анадара использует ресурс тканевого лактата в направлении реакций окислительного декарбоксилирования. Об этом свидетельствует значительный рост активности лактатдегидрогеназы на фоне снижения содержания лактата и повышения уровня пирувата в тканях. Процесс адаптации анадары к голоданию идет по пути использования резерва аминокислот в процессах биосинтеза белка. The goal of this study was to evaluate the effects of seasonal variations on energy metabolism in different tissues of the freshwater crayfish Parastacus brasiliensis (von Martens, 1869). Crayfish were collected monthly from January 2001 to January 2003 in São Francisco de Paula, Rio Grande do Sul, Brazil, in a stream and in a culture tank. Haemolymph samples were collected from each crayfish in the field with a syringe, by puncturing the membrane at the base of the chelipeds. Combined hydrothermal process of auto-hydrolysis and enzymatic hydrolysis as pretreatment for release of glucose from malt bagasse. The auto-hydrolysis pretreatment removes the hemicellulose and chemically modifies the lignin, facilitating the access of enzymes to the cellulose, which will be converted into glucose.

Biocatalyst consisting of immobilized cellulases for hydrolysis of pretreated sugarcane bagasse by organosolv process. The combination with easily hydrolyzable cellulose allows obtaining highly pure glucose and the reuse of enzymes. The immobilized enzyme represents most of the solid recovered at the end of the hydrolysis because almost all the solid substrate added to the reactor is solubilized.

Alkaline pretreatment process followed by enzymatic hydrolysis for release of glucose from malt bagasse. The alkaline pretreatment promotes biomass delignification, exposing the cellulose fibers, making them accessible to enzymatic action. A maximum conversion of cellulose into glucose (74%) was achieved when sodium hydroxide was used at a concentration of 4% (w/v).

Effect of diets containing different levels of highly unsaturated fatty acids on physiological and immune responses in Pacific whiteleg shrimp Litopenaeus vannamei (Boone) exposed to handling stress ArchiMer

Juveniles fed a diet containing a low or a high level of highly unsaturated fatty acids (HUFA) for 38 days were exposed to handling stress. In a first experiment, stress was applied daily for 30 days, after which the physiological and immunological variables were measured, whereas in a second experiment, stress was applied once and samples were obtained 1 and 24 h after the stressor event. Shrimp that were stressed for 30 days showed significantly lower survival, final weight and feed consumption compared with unstressed shrimp. The concentration of the high-density lipoprotein beta-glucan-binding protein was significantly higher in shrimp fed the high-HUFA diet. Phosphoenolpyruvate carboxykinase (PEPCK) is a rate-limiting enzyme in hepatic gluconeogenesis and therefore plays a central role in glucose homeostasis. The aim of this study was to analyse the nutritional regulation of PEPCK gene expression in rainbow trout (Oncorhynchus mykiss), which are known to use dietary carbohydrates poorly. A full-length hepatic PEPCK cDNA (2637 base pairs with one open reading frame putatively encoding a 635-residue protein) was cloned and found to be highly homologous to mammalian PEPCKs. In crayfish, the hemolymph glucose level shows an endogenous circadian rhythmicity which is expressed by a nocturnal glucose rise. This study shows that injections of serotonin and dopamine result in hyperglycemia and that injections of met-enkephalin decrease hemolymph glucose level as well as the nocturnal circadian glucose peak. Using immunocytochemistry, these neurotransmitters have been located in the eyestalk and their relationship with the CHH cell system is described.

A study was conducted to evaluate the effect of two dietary carbohydrate sources (waxy maize starch and glucose) on the metabolic adaptation of sea bass juveniles (initial weight: 24 g) to a heat shock treatment (temperature rise from 18 degrees C to 25 degrees C within 24 h). Two isonitrogenous and isolipidic diets were fon-nulated to contain 20% waxy maize starch (WS diet) or 20% glucose (GLU diet). Triplicate groups of fish were fed to near satiation for 4 weeks at both temperatures (18 degrees C and 25 degrees C). In the Pacific oyster, Crassostrea gigas, the energy necessary for general metabolism and gametogenesis is mainly provided by the glycogen stored in the vesicular cells. The first level of glycogen metabolism control is the entrance of glucose into cells. The monitoring of the uptake capacity of [U-14C] D-glucose revealed the existence of a saturable component with kinetic constants: Km = 0.71 mM ; Vmax = 0.22 nmol/h/106cells. Effects of environmental conditions on the sessile existence of an estuarine sediment bacterium. ArchiMer

A submerged glass coverslip technique was developed to determine dissolved organic nutrient uptake by adherent cells of a sediment isolate of Enterobacter cloacae . Cells which colonized the coverslips in swirling culture flasks during anaerobic growth remained firmly attached during manipulations employed to determine uptake of radiolabeled glucose by the adherent population. The attached cells were capable of a more rapid rate of glucose uptake than free cells. Lineweaver-Burk plots demonstrated different glucose uptake kinetics for the 2 cell populations. The data suggest that physiological changes occur in cells soon after they become attached to surfaces. These changes appear to enhance the metabolic activity of the adherent population.

Some nutrients known as non essential in vertebrates could be essential in invertebrates and could be related to some "unknown growth factors" which were evidenced or suspected, mainly in mollusc or crustacean flesh or meal, by several authors working on different species of crustaceans. A set of experiments was carried out in our laboratory in order to isolate one of these factors found in squid meal.

This study was conducted to evaluate the effect of Tribulus terrestris hydroalcoholic extract administration on the serum level of glucose and reproductive parameters in diabetic male rats. ABSTRACT The objective of this study was to evaluate stress responses in dog snapper (Lutjanus jocu) during transport by evaluating their hematological and biochemical responses. Twenty-five wild dog snapper specimens were used in the experiment (220 ± 68 g and 24.5 ± 2.5 cm total length). Blood samples were collected prior to transport (control), and fish were placed in two transport boxes, one with anesthetic and one without anesthetic. ABSTRACT The aim of this study was to evaluate if Lippia alba has different chemotypes according to the chemical composition of the essential oil (EO) considering collection site, and if the EO may have different effects on blood and plasma parameters in silver catfish, Rhamdia quelen, during and immediately after anesthesia. The citral (EO-C) and linalool (EO-L) chemotypes were identified, and both presented similar anesthetic effects for silver catfish.
