The use of raw rice husk as substrate allows the use of crop systems that promote the recirculation of leachate in long crop cycles. Mini tomatoes present relatively low demand for photoassimilates. Thus, partial defoliation of the sympodium could benefit the crop without damage to the production or quality of the fruits. The objective of this work was to evaluate the plant growth, fruit yield and fruit quality of Cherry Hybrid Wanda and Grape Hybrid Dolcetto mini tomatoes cultivated in two recirculation crop systems (pots and troughs), using raw rice husk as substrate, under three defoliation conditions (without defoliation, removal of one and two leaves of the sympodium). Grape quality mapping for vineyard differential harvesting BABT

An experiment was carried out from 2005 to 2008, to calibrated NIR-based instrumentation and explore within field grape quality variability and map potential grape quality descriptors along vineyards, as a subsidy for differential harvesting,. The quality indicators (anthocyanin content, pH, titratable acidity and soluble solids) were subject to geo-spatial modeling. Subsequently, the data set was utilized to delineate "within-field" grape quality zone and to determine the timing of the harvest. Differential machine harvesting was implemented and segregation of wine grapes was done "on-the-go". The approach for field prediction of grape quality parameters and zone delineation allowed for separated fermentation for at least two wine styles.

Intralaboratory optimization and validation of a method for patulin determination in grapes by Thin-Layer Chromatography BJM

The aim of this work was to optimize and validate, by intralaboratorial procedures, a method for the determination of patulin in grapes by thin-layer chromatography. The steps of extraction, cleanup, detection and quantification were optimized. For the validation of the method, recovery assays with standard solutions and artificially contaminated samples were carried out. The mean recovery and the variation coefficient were 65.4% and 7.58%, respectively. The optimized conditions were: 50 mL of grape juice, three extraction stages (with 100 mL of ethyl acetate in the first stage and 50 mL in second and third stages), and 100 µL of ethanol to solubilize the extract. The enzimatic activity of peroxidase (POD) and polyphenoloxidase (PPO) extracted from three grape cultivars (Vitis vinifera L.), cultivated in Marialva city, state of Paraná, was evaluated in this study. The enzymatic extracts were prepared starting from the Rubi, Borbon and Benitaka grape cultivars pulp and peel. The activity of the peroxidase was 53.00 units/100 g in the extract from the Rubi cultivar peel, and 327.00 units/100 g from the Benitaka cultivar, these values being superior to those observed in the same cultivars pulp extracts, which were 7.67 units/100 g and 44.00 units/100 g respectively. Abstract Brazilian wine production is characterized by Vitis labrusca grape varieties, especially the economically important Isabel cultivar, with over 80% of its production destined for table wine production. The objective of this study was to optimize and validate the conditions for extracting volatile compounds from wine with the solid-phase microextraction technique, using the response surface method. Based on the response surface analysis, it can be concluded that the central point values maximize the process of extracting volatile compounds from wine, i.e., an equilibrium time of 15 minutes, an extraction time of 35 minutes, and an extraction temperature of 30 °C. Abstract A comprehensive investigation was conducted to determine the effects of exogenous melatonin treatment of pre-veraison grapes on phenolic components and antioxidants of grape berries and wine. The results showed an increase in the concentration of cyanin-3-O-glucoside, peonidin derivatives, and two flavanols [namely (+)-catechins and (−)-epicatechins] of grapes, as well as flavonols in both grapes and wine due to the application of melatonin. These compounds are derivatives of the catalysate of flavonoid 3’-hydroxylase (F3’H) in the flavonoid synthesis pathway of plants. ABSTRACT: The objective of this study is to research the genetic diversity of the ‘ Zuijinxiang ’ grape and its mutant breeding F1 plants, we screened the excellent mutant plants with potential breeding value. 50 mutated single plants obtained from 137Cs-γ irradiated ‘Zuijinxiang’ grape seeds were used as research objects, and SCoT molecular marker technology was used for genetic diversity and variation analysis, and clustering research was carried out. The results showed that: (1) 36 SCoT primers produced abundant polymorphisms, and the amplification results showed obvious bright bands, and the amplification efficiency and polymorphism rate were 100%. Extraction of RNA from plant tissue containing high levels of polyphenols and polysaccharides is tedious and difficult in grapes. Although several protocols have been published for plant RNA isolation, most have failed to yield high quality RNA in sufficient quantity from mature and diseased grape tissue. We describe a protocol for isolating intact and high quality RNA from various grape tissues as evident by high A260/A280 absorbance ratio (1.8 to 1.9) and electrophoretic profile on denaturing formaldehyde agarose gel. Constatou-se excelentes resultados com a enxertia de mesa, realizada no Campo Experimental de Mandacaru. Este método de enxertia apresenta uma série de vantagens. São apresentados os seguintes ítens, para a realização deste método: preparo dos enxertos; preparo dos porta-enxertos; enxertia; plantio da estaca enxertada e transplantio da muda para o campo.
