In the state of Rio Grande do Sul, Brazil, yerba mate (Ilex paraguariensis A. St.-Hil.) is an economically and culturally important species. Because of the difficulty in obtaining plantlets from seeds, micropropagation has emerged as an alternative that could increase production. This work aimed to evaluate the effect that treatment with 6-benzyladenine (BA), at concentrations of 2.2, 8.9, and 20 µM and associated with naphthaleneacetic acid (NAA, 0.5 µM), has on the in vitro growth and leaf emission of I. paraguariensis nodal segments cultivated in 0.5 X Murashige and Skoog (MS) medium or 0.5 X Woody Plant Medium (WPM). The most efficient treatment was MS medium with 8.9 µM BA+NAA, in which I. paraguariensis presented greater shoot elongation and higher... |

ABSTRACTOil palm is one of the most economically valuable oil seed plants, but the expansion of plantations has been limited by availability of seedlings, as the conventional propagation is through seeds, which have low germination rates. One possible solution for the large-scale production is the use of somatic embryogenesis. The aim of this study was evaluate the effects auxins 2,4-D and picloram on the induction of pro-embryogenic masses inE.guineenesis hybrid leaf explants and characterize, regarding embryogenic characteristics, with cytochemical and ultrastructural analisys. Specifically, in vitro plantlets leaves fragments were inoculated in Y3 culture medium supplemented by 2.4-D or picloram at different concentrations (0.0, 1.0, 3.0, 6.0 and 9.0 mg... |

Nanism is one of the most frequent type of mutant in micropropagated banana plants from the Cavendish subgroup. The present study aimed at studying some of the hormone factors involved in this type of mutation. Rhizomes from normal and dwarf plants from the cultivar Grand Naine were incubated for 5 d in the presence of [³H]-L-tryptophan, [³H]-indole-3-acetic acid and gibberellin, to quantify the endogenous levels of indole-3-acetic acid-ester, indole-3-acetic acid-amide, free indole-3-acetic acid, and cytokinins. The endogenous levels of indole-3-acetic acid and its ester- and amide-conjugated forms were measured in normal and dwarf plants incubated for 30 d with gibberellin, indole-3-acetic acid, and L- and D-tryptophan. In normal plants, the use of... |

This work focuses on the evaluation of flowering and vegetative growth of olive tree submitted to pruning and paclobutrazol application under field conditions with low temperatures during the winter. Two-years-old olive plant variety Grappolo 575, were submitted to light pruning, removing apical dominance, before treatments application. The treatments were organized in a 4×2 factorial scheme, respective to four PBZ concentrations tested: 0, 200, 400 and 800 mg L-1 of PBZ and two plants groups, with and without pruning, in randomized block with four replications. Paclobutrazol concentrations tested did not affect the olive tree flowering. The plant vegetative growth was reduced until 60 days after paclobutrazol application. Pruning resulted in stimulation... |

Annona glabra is a tropical species that has significant agronomic potential in terms of furnishing fruits for in natura consumption and for the production of phyto-pharmaceuticals. In vitro cultivation has been considered the most promising form of propagation for this species, although large scale utilization of this technique is currently limited by high rates of leaf abscission, reduced rates of explant multiplication and slow bud growth. The present work evaluated the effects of different cytokinins on mineral accumulation in shoots of A. glabra cultivated in vitro, and their effects on growth and survival of these plants. Buds of A. glabra were cultivated in Wood Plant Medium (WPM) in the presence of 6-benzilaminopurine (BAP), thidiazuron (TDZ),... |

ABSTRACT: The availability of an efficient protocol for in vitro regeneration is imperative for genetic transformation. Using genotypes adapted to the target region as a transgenic platform accelerates the development of cultivars. Therefore, this study aimed to adapt an in vitro regeneration protocol for Brazilian wheat genotypes. For this purpose, the in vitro regeneration capacity of immature embryos from six Brazilian wheat genotypes using two protocols of regeneration of somatic embryos was analysed. Furthermore, combinations of 2,4-D and picloram in the callus induction medium were tested in order to improve regeneration efficiency. Genotypes with higher regeneration efficiency were BR18-Terena and PF020037, yielding 0.42 and 1.13 shoots per explant... |

A large number of micropropagated plantlets of banana, Musa acuminata var. Nanjanagudu Rasabale (NR), that were developed from axillary shoot bud explants over 10 years ago were screened for genetic variation, if any, using RAPD (Random Amplified Polymorphic DNA) and ISSR (Inter-Simple Sequence Repeats) markers. Of the 4000 in vitro plantlets, 11 were used for screening that involved shoot cultures with distinct variation in morphological characteristics (morphotypes). Similarly, the mother maintained in the field was also subjected for genetic analysis. Out of the 50 RAPD and 25 ISSR primers screened, 30 RAPD and 5 ISSR primers produced totally 424 clear, distinct and reproducible band classes resulting in a total of 5088 bands where the banding patterns... |

ABSTRACT The aim of this study was to evaluate the effect of salicylic acid (SA) application on pre-transplant acclimatization of native populations of tomato. SA is a growth regulator that modifies plant growth and development by inducing changes in cell processes, physiology and morphology. Five populations of native tomato were sown in polystyrene trays. Peat moss was used as substrate and plants were maintained at field capacity continuously. After emergence, seedlings were applied during three weeks with different concentration of SA (0.0, 0.01, 0.1, 0.5 and 1.0 μM). A completely random experimental design was used with five replications per treatment. The growth parameters evaluated were height, stem diameter, number of leaves, hypocotyl length,... |

Aechmea blanchetiana and Aechmea distichantha are Brazilian native bromeliads with great ornamental value, although none of the species are produced commercially. Therefore, market demands are met through nature predatory exploitation. This study intended to (1) evaluate the in vitro seed germination of A. blanchetiana and A. distichantha and (2) establish micropropagation protocols for both species, aiming at producing plantlets for the market, as well as to germplasm in vitro conservation. Germination experiments were carried out in completely randomized designs with two treatments (presence and absence of light) and seven and eight replications respectively for A. blanchetiana and A. distichantha. In vitro seedlings, produced under light conditions,... |
