Constitutional and somatic methylation status of DMRH19 and KvDMR in Wilms tumor patients Genet. Mol. Biol.
Cardoso,Leila C.A.; Castaño,Jair A. Tenorio; Pereira,Hanna S.; Lima,Maria Angélica de F.D.; Santos,Anna Cláudia E. dos; Faria,Paulo S. de; Ferman,Sima; Seuánez,Héctor N.; Nevado,Julián B.; Almeida,José Carlos Cabral de; Lapunzina,Pablo; Vargas,Fernando R..
The most frequent epigenetic alterations in Wilms tumor (WT) occur at WT2, assigned to 11p15. WT2 consists of two domains: telomeric domain 1 (DMRH19) that contains the IGF2 gene and an imprinted maternally expressed transcript (H19) and centromeric domain 2 (KvDMR) that contains the genes KCNQ1, KCNQ1OT1 and CDKN1C. In this work, we used pyrosequencing and MS-MLPA to compare the methylation patterns of DMRH19/KvDMR in blood and tumor samples from 40 WT patients. Normal constitutional KvDMR methylation indicated that most of the epigenetic alterations in WT occur at DMRH19. Constitutional DMRH19 hypermethylation (HM DMRH19) was observed in two patients with Beckwith-Wiedemann syndrome. Pyrosequencing and MS-MLPA showed HM DMRH19 in 28/34 tumor samples:...

Histopatological effects of bleaching and disease on the coral Siderastrea stellata from coastal reefs of Brazil Iheringia, Sér. Zool.
Sassi,Cristiane F. Costa; Farias,Gabriel M. de; Vasconcelos,Amanda de S.; Macedo,Rebeca S. de; França,Jéssica P. de S.; Sassi,Roberto.
ABSTRACT Histological analysis of bleached samples of Siderastrea stellata Verrill, 1868, with white plague disease and with alteration in the color pattern, revealed drastic tissue and cellular disturbances, such as lysis of the external epithelium, hyperplasia of gastrodermis, apoptosis of epithelial cells and zooxanthellae, and degradation of mesenteric filaments and reproductive cells. Fungal hyphae, diatoms, and cyanobacteria were present in diseased samples and colonies with alteration in the color pattern. Furthermore, bleached and diseased samples showed significant reductions in the number of zooxanthellae per 100 µm2 of gastrodermis and significant reductions in the diameters and volumes of stage V oocytes. We found that bleaching events,...

An infection of Ruditapes decussatus (Bivalvia) by Rickettsia ArchiMer
Mialhe, Eric; Chagot, Dominique; Boulo, Viviane; Comps, Michel; Ruano, Francisco; Grizel, Henri.
The authors report the presence of a Rickettsia in Ruditapes decussatus from natural populations of the Algarve (Portugal). Light microscopic shows infections of varying importance in gill tissues. Some enclose small procaryotic colonies (cp) scattered in all lamellae; in others, the colonies are scarce and hypertrophic (100 mu m). These different pictures suggest an amplification process. Digestive diverticulae may also be infected, but to a lesser extent. The ultrastructural features of this intravacuolar (mvd: densified vacuolar membrane) procaryote (P) and its multiplication by scission without a complex developmental cycle allows its classification in the order Rickettsiales.

Two Vibrio splendidus related strains collaborate to kill Crassostrea gigas: taxonomy and host alterations ArchiMer
Gay, Melanie; Renault, Tristan; Pons, Anne-marie; Le Roux, Frédérique.
For several years, strains phenotypically related to Vibrio splendidus have been associated with mortality outbreaks of molluscs. A former study on Crassostrea gigas demonstrated the genetic diversity of V splendidus strains associated with diseased animals. Another study suggested that different strains may act in an additive/synergistic way leading to higher C. gigas mortality rates. Here, a strain pair (31+32) was characterised at taxonomic and virulence levels. Using a polyphasic approach, these strains were confirmed to be V splendidus-related, without a clear discrimination between V. kanaloae and V. pomeroyi since hybridisation rates with both these strains were above 70%. Following experimental infection of C. gigas by injection in the adductor...

Histopathological assessment of liver and gonad pathology in continental slope fish from the northeast Atlantic Ocean ArchiMer
Feist, S. W.; Stentiford, G. D.; Kent, Maxxie; Ribeiro Santos, Ana; Lorance, Pascal.
The deep-sea environment is a sink for a wide variety of contaminants including heavy metals and organic compounds of anthropogenic origin. Life history traits of many deep-water fish species including longevity and high trophic position may predispose them to contaminant exposure and subsequent induction of pathological changes, including tumour formation. The lack of evidence for this hypothesis prompted this investigation in order to provide data on the presence of pathological changes in the liver and gonads of several deep-water fish species. Fish were obtained from the north east region of the Bay of Biscay (north east Atlantic Ocean) by trawling at depths between 700 to 1400m. Liver and gonad samples were collected on board ship and fixed for...

Evaluation of toxicity of Deepwater Horizon slick oil on spat of the oyster Crassostrea virginica ArchiMer
Vignier, Julien; Rolton, Anne; Soudant, Philippe; Chu, Fu-lin E.; Robert, Rene; Volety, Aswani K..
The 2010 explosion of the Deepwater Horizon (DWH) oil rig generated the largest marine oil spill in US history with millions of barrels of crude oil released in the Gulf of Mexico (GoM). The eastern oyster, Crassostrea virginica, is an ecologically and economically important species in the northern GoM. Due to its biological characteristics (sessile, filter feeding), juvenile oysters may have been affected. This study investigated the effects of surface-collected DWH oil prepared as high-energy water-accommodated fraction (HEWAF) on the survival of 2-month-old oyster spat, and evaluated the potential impacts of HEWAF on particle clearance rate and spat tissue. Exposure of oysters to a range of oil/HEWAF (0-7-66-147-908-3450 mu g tPAH50 (sum of 50...

Comparison of the effects of the dietary addition of two lactic acid bacteria on the development and conformation of sea bass larvae, Dicentrarchus labrax, and the influence on associated microbiota ArchiMer
Lamari, Faouzi; Castex, Mathieu; Larcher, Thibaut; Ledevin, Mireille; Mazurais, David; Bakhrouf, Amina; Gatesoupe, Joel.
Probiotics may have many effects on health and development of fish larvae. One of the most promising is related to spinal conformation, though the mode of action is not clearly understood. The present study attempted to investigate the effects of two strains of lactic acid bacteria on associated microbiota, histological development and gene expression. Sea bass larvae were fed since 5 dph (day post hatch) with either a standard control diet (Diet C), or the same diet supplemented with Pediococcus acidilactici MA18/5M (Diet P), or with an autochthonous strain of Lactobacillus casei (X2; Diet L). The two lactic acid bacteria were incorporated in the diets at the levels of 106 and 107 CFU (colony forming units) g-1 in two consecutive experiments,...

Effect of fish meal replacement by plant protein sources on non-specific defence mechanisms and oxidative stress in gilthead sea bream (Sparus aurata) ArchiMer
Sitja Bobadilla, A; Pena Llopis, S; Gomez Requeni, P; Medale, Francoise; Kaushik, Sadasivam; Perez Sanchez, J.
Partial or total replacement of fish meal by a mixture of plant protein (PP) sources (corn gluten, wheat gluten, extruded peas, rapeseed meal and sweet white lupin) balanced with indispensable amino acids was examined in juvenile gilthead sea bream over the course of a 6-month growth trial. A diet with fish meal (FM) as the sole protein source was compared to diets with 50%, 75% and 100% of replacement (PP50, PP75, and PP100). The possible influence of diets on growth performance, plasma metabolites, gut integrity, liver structure, anti-oxidant and immune status was evaluated. Final body weight was progressively decreased with PP inclusion, but in PP50 and PP75-fed fish, feed efficiency (FE) was significantly improved and specific growth rates remained...

Gill and Liver Histopathology in Goodea atripinnis Jordan, Related to Oxidative Stress in Yuriria Lake, Mexico International Journal of Morphology
Ruiz-Picos,R; López-López,E.
In aquatic ecosystems, the complex mixture of pollutants may mediate the formation of free radicals and cause oxidative damage to the biota. Yuriria Lake (a Ramsar site in Central Mexico) receives input of wastewater from its tributaries, agricultural runoff, and municipal discharge. We studied the lipid peroxidation (LPO), antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx), and histopathology of gill and liver of the native fish Goodea atripinnis in Yuriria Lake. Results were compared to a control group of fish cultivated in the laboratory. LPO, SOD, and CAT showed no significant differences compared to controls, but GPx showed greater and significant differences in both tissues. Three class sizes were...

Martins,A. M. C. R. P. F; Bersano,J. G; Ogata,R; Amante,G; Nastari,B. D. B; Catroxo,M. H. B.
The porcine transmissible gastroenteritis is a highly severe contagious disease, caused by virus of the Coronaviridae family, genus Coronavirus. Its epizootic shape can reach a rate of up to 100% mortality in piglets under two weeks of age as a result of severe dehydration. In this study fragments of small intestine and stool samples were collected from 75 autopsied pigs from properties. The samples of the fragments were frozen and sent to the Laboratory of Electron Microscopy, Instituto Biológico, SP, Brazil, for histological and transmission electron microscopic analyses. According to histological H&E technique, atrophy, villous necrosis and destruction of the enterocytes were observed in 35 (46.6%) out of the 75 fragments of the small intestine...
