Isolation and molecular characterization of Rhizoctonia-like fungi associated with orchid roots in the Quadrilátero Ferrífero and Zona da Mata regions of the state of Minas Gerais, Brazil Acta Botanica

Mycorrhizal associations can be considered required for orchids, which depend on the fungi for germination and establishment in natural conditions. Knowledge of the mycorrhizal fungi is important for programs aimed at the reintroduction, conservation and management of orchid species. The objective of this study was the molecular characterization of Rhizoctonia-like fungi from orchids in the Quadrilátero Ferrífero ("Iron Quadrangle") and Zona da Mata ("Forest Zone") regions of the state of Minas Gerais, Brazil. The affinities of these fungi were studied by comparing the rRNA internal transcribed spacer region with that of other isolates and sequences in GenBank. Three isolates had an affinity for Epulorhiza repens, and one was the holotypeof E. epiphytica.

In the present study, nine terverticillate Penicillium isolates (P. griseofulfum, P. puberulum, P. crustosum, P. aurantiogriseum, P. chrysogenum, P. primulinum, P. expansum, P. viridicatum, Eupenicillium egyptiacum) from 56 soil samples were characterized genetically by a PCR method. Stone pine (Pinus pinea L.), like other conifers, forms ectomycorrhizas (ECM), which have beneficial impact on plant growth in natural environments and forest ecosystems. An in vitro co-culture of stone pine microshoots with pure mycelia of isolated ECM sporocarps was used to overcome the root growth cessation not only in vitro but also to improve root development during acclimation phase. ABSTRACT: Atta sexdens rubropilosa (leaf-cutter ants) has a symbiotic association with a fungus and has a negative interaction with other fungi due to parasitism of the fungus cultivated by ants; also, there are several other fungi with no exact known role occurring in their cultivated fungus garden. In the present study, we use the ITS region (internal transcribed spacer) to identify fungi in colonies treated with toxic baits. Se estudió la variabilidad de 2 genes nucleares, el intrón 4/5 del gen de Calmodulina (CaM) por amplificación PCR y la región ITS (Internal Transcribed Spacers) del ADNr, por PCR y digestión en 139 ejemplares de E. ringens provenientes de las costas de Iquique (20°13'S; 70°09'W) y Talcahuano (36°41'S; 73°06'W). Phylogenetic relationships among Chilean species of Drimys as well asD. granadensis from Central America andD. angustifolia from Brazil were reconstructed using internal transcribed spacers (ITS) of nuclear ribosomal DNA and a parsimony analysis. Our results at the family level were congruent with previous phylogenetic hypotheses. Evolutionary history of the Tip100 transposon in the genus Ipomoea Genet. Mol. Biol.

Tip100 is an Ac-like transposable element that belongs to the hAT superfamily. First discovered in Ipomoea purpurea (common morning glory), it was classified as an autonomous element capable of movement within the genome. As Tip100 data were already available in databases, the sequences of related elements in ten additional species of Ipomoea and five commercial varieties were isolated and analyzed. Molecular evidence of polyphyletism in the plant genus Carum L. (Apiaceae) Genet. Mol. Biol.

An analysis of internal transcribed spacer (ITS) DNA sequences of the four species of Carum L. (Apiaceae) known in Italy revealed that this genus is polyphyletic. Maximum parsimony with bootstrap resampling, maximum likelihood and Bayesian inference analyses resulted in three distinct clades: Carum carvi L. clustered within tribe Careae Baill. (former Aegopodium clade); Hellenocarum multiflorum (= Carum multiflorum), Carum heldreichii and Carum appuanum clustered within the tribe Pyramidoptereae Boiss.; and H. multiflorum and C. heldreichii formed a well supported clade. The discovery and characterization of informative intraspecific genetic markers is fundamental for evolutionary and conservation genetics studies. Here, we used nuclear ribosomal ITS sequences to access intraspecific genetic diversity in 23 species of the genus Passiflora L. Some degree of variation was detected in 21 of these. The Passiflora and Decaloba (DC.) Rchb. subgenera showed significant differences in the sizes of the two ITS regions and in GC content, which can be related to reproductive characteristics of species in these subgenera. Characterization of the protozoan parasite Marteilia refringens infecting the dwarf oyster Ostrea stentina in Tunisia ArchiMer

Marteilia refringens is a protozoan parasite recognized as a significant pathogen of the European flat oyster Ostrea edulis. The life cycle of this species is still poorly known, although there is evidence of the need for intermediate host(s). In the present study, we have used molecular approaches to identify this parasite in samples of the dwarf oyster Ostrea stentina after reports of massive mortality along the Tunisian coasts. In 2009 we evaluated the status of O. stentina from Monastir and checked if there was an infection with M. refringens, using polymerase chain reaction assays. Morphological and genetic diversity of Beaufort Sea diatoms with high contributions from the Chaetoceros neogracilis species complex ArchiMer

Seventy-five diatoms strains isolated from the Beaufort Sea (Canadian Arctic) in the summer of 2009 were characterized by light and electron microscopy (SEM and TEM) as well as 18S and 28S rRNA gene sequencing. These strains group into 20 genotypes and 17 morphotypes and are affiliated with the genera Arcocellulus, Attheya, Chaetoceros, Cylindrotheca, Eucampia, Nitzschia, Porosira, Pseudo-nitzschia, Shionodiscus, Thalassiosira, Synedropsis. Most of the species have a distribution confined to the northern/polar area. Chaetoceros neogracilis and Chaetoceros gelidus were the most represented taxa. Strains of C. neogracilis were morphologically similar and shared identical 18S rRNA gene sequences, but belonged to four distinct genetic clades based on 28S rRNA,...
