Characterization and expression analysis of chymotrypsin after bacterial challenge in the mud crab, Scylla paramamosain Genet. Mol. Biol.

Chymotrypsin is one of the serine proteases families that have various biological functions. A chymotrypsin gene was isolated from hepatopancreas of the mud crab, Scylla paramamosain (designated SpCHY) in this study. The full-length cDNA of SpCHY contained 942 nucleotides with a polyadenylation sequence and encoded a peptide of 270 amino acids with a signal peptide of 17 amino acids. The SpCHY gene contains seven exons, six introns, a TATA box and several transcription factor binding sites that were found in 5'-promoter region which is 1221 bp in length-time quantitative PCR analysis indicated that the expression level of SpCHY mRNA in hepatopancreas was significantly higher than that in other tissues. HER2/neu Status in Breast Cancer Specimens: Comparison of Immunohistochemistry (IHC) and Fluorescence in situ Hybridization (FISH) Methods International Journal of Morphology

HER2 amplification or overexpression is considered as disease outcome and a predictive marker of response to treatment in breast cancer. The present study aimed to compare the results of IHC and FISH for determining HER2 and to search the interpretational differences. Samples (n= 169), of which 31 were the paraffin blocks sent from outer centers, that underwent FISH analysis for HER-2 were included. Samples were re-reviewed by IHC in our laboratory. FISH test was negative in 131 (77.5%) and positive in 38 (22.5%). When those with previous IHC 0-1+ were re-reviewed, the results were found again 0-1+ and none of them was FISH positive. Inconsistency between re-reviewed IHC and previous IHC results was 25% for those with 2+ score and 11% for those with 3+...

In this study thirty shrimp samples from commercial marine shrimp (L. vannamei) farms of southern region of Brazil were obtained. Hepatopancreas and shell scrapings fragments collected in these animals were processed by transmission electron microscopy using negative staining (rapid preparation), immunoelectron microscopy and immunocytochemistry (immunolabelling with colloidal gold particles) techniques. Cytomorphology of Circulating Tumor Cells and Cell Clusters in Chilean Patients with Colo-Rectal Cancer International Journal of Morphology

The aim of this study was to determine the cytomorphological characteristics of circulating tumor cells (CTCS) in patients with colo-rectal cancer and compare them with the primary tumor and metastasis. CTCS were obtained from blood using differential gel centrifugation and detected using standard immunocytochemistry using anti-CEA. Primary CTCs were defined as those detected before surgery and secondary CTCs those detected after. Surgical specimens of the primary tumor and metastasis were evaluated using standard histological methods with hematoxillin and eosin. ABSTRACT Heparan sulfate proteoglycans (HSPGs) are present on the cell surface and in the extracellular matrix in all metazoans. HSPGs interact with growth factors and receptors through heparan sulfate (HS) chains. The sulfation pattern of heparan sulfate chains influences signaling events mediated by heparan sulfate proteoglycans located on the cell surface. SULF1 and SULF2 are two endo-sulfatases that can cleave specific 6-O-sulfate groups within the heparan chains. Mapping of serotonin-immunoreactive neurons of Anastrepha obliqua Macquart larvae Zoologia

Serotonin-immunoreactive neurons were identified in the central nervous system (CNS) of Anastrepha obliqua Macquart, 1835 wandering stage larvae. The PAP immunocytochemical method was applied to the entire CNS (whole mounts). About 90 neurons were visualized in the CNS (20 in the brain and 70 in the ventral ganglion). Both somata and axons were strongly stained. These neurons showed a segmental arrangement and bilateral symmetry. All processes presented a basic projection pattern, in which the major fibres travel contralaterally. Comparison of these neurons with serotonergic neurons described in other insects suggests order-specific traits such as cerebral clusters and presence of only one 5-HT immunoreactive neuron in the 8th abdominal neuromere as well.

Binding patterns of seminal plasma plasma proteins on bovine epididymal and ejaculated sperm membrane Arq. Bras. Med. Vet. Zootec.

The present study was designed to investigate the topographical distribution of seminal plasma (SP) proteins on epididymal and ejaculated bovine sperm. Using immunocytochemistry and confocal microscopy the binding patterns of bovine SP proteins BSP-A3, albumin, transferrin, prostaglandin D-synthase (PGDS) and nucleobindin in ejaculated and cauda epididymal sperm from adult bulls were evaluated. Experiments were performed using sperm from 5 males. Data showed a positive signal, only detected for anti-PGDS, in the acrosomal cap of epididymal and ejaculated sperm. The trabecular meshwork (TM) is the main outflow pathway in the mammalian eye. Oxidative damage to TM cells has been suggested to be an important cause of impairment of TM functions, leading to deficient drainage of aqueous humor, with deleterious consequences to the eye. Transferrin, a metalloprotein involved in iron transport, has been characterized as an intrinsic eye protein.
