Oligomerization of Cry9Aa in solution without receptor binding, is not related with insecticidal activity Electron. J. Biotechnol.
Fang,Longfa; Wang,Bo; Zhou,Zishan; Yang,Sujuan; Shu,Changlong; Song,Fuping; Bravo,Alejandra; Soberón,Mario; Zhang,Jie.
Background: Bacillus thuringiensis Cry toxins bind with different insect midgut proteins leading to toxin oligomerization, membrane insertion and pore formation. However, different Cry toxins had been shown to readily form high molecular weight oligomers or aggregates in solution in the absence of receptor interaction. The role of Cry oligomers formed in solution remains uncertain. The Cry9A proteins show high toxicity against different Lepidoptera, and no-cross resistance with Cry1A. Results: Cry9Aa655 protein formed oligomers easily in solution mediated by disulfide bonds, according to SDS-PAGE analysis under non-reducing and reducing conditions. However, oligomerization is not observed if Cry9Aa655 is activated with trypsin, suggesting that cysteine...

The chitinase C gene PsChiC from Pseudomonas sp. and its synergistic effects on larvicidal activity Genet. Mol. Biol.
Zhong,Wanfang; Ding,Shaojun; Guo,Huifang.
Pseudomonas sp. strain TXG6-1, a chitinolytic gram-negative bacterium, was isolated from a vegetable field in Taixing city, Jiangsu Province, China. In this study, a Pseudomonas chitinase C gene (PsChiC) was isolated from the chromosomal DNA of this bacterium using a pair of specific primers. The PsChiC gene consisted of an open reading frame of 1443 nucleotides and encoded 480 amino acid residues with a calculated molecular mass of 51.66 kDa. The deduced PsChiC amino acid sequence lacked a signal sequence and consisted of a glycoside hydrolase family 18 catalytic domain responsible for chitinase activity, a fibronectin type III-like domain (FLD) and a C-terminal chitin-binding domain (ChBD). The amino acid sequence of PsChiCshowed high sequence homology...

Mortality of Plutella xylostella larvae treated with Aspidosperma pyrifolium ethanol extracts PAB
Trindade,Roseane Cristina Prédes; Silva,Paulo Pedro da; Araújo-Júnior,João Xavier de; Lima,Ivanildo Soares de; Paula,José Elias de; Sant'Ana,Antônio Euzébio Goulart.
The objective of this work was to assess the effects of Aspidosperma pyrifolium ethanol extracts on cabbage moth (Plutella xylostella) larvae. The ethanol extracts of the stem bark, fruits and roots of A. pyrifolium were obtained by classical phytochemical methods, and the resulting subfractions were tested on P. xylostella, using 4 and 5 mg L-1. The crude ethanol extract of the stem bark was more lethal. The alkaloid-rich aqueous subfraction derived from the stem bark extract caused 100% larval mortality at 4 mg L-1. Insecticidal activity was associated with the presence of the monoterpenoid indole alkaloids aspidofractinine, 15-demethoxypyrifoline, and N-formylaspidofractinine. These alkaloids presented excellent insecticidal properties against P....

Synthesis of new α-amino nitriles with insecticidal action on Aedes aegypti (Diptera: Culicidae) Rev. Bras. entomol.
Rueda,Andrés G.; Carreño Otero,Aurora L.; Duque,Jonny E.; Kouznetsov,Vladimir V..
Abstract Aedes aegypti is the principal vector of arboviral pathogens that may cause diseases as dengue fever, chikungunya and zika. The harmful environmental effects of commercial pesticides coalesced with the development of insecticide-resistant populations encourage the discovery and generation of new alternative products as a tool to reduce the incidence of vector-borne diseases. In this work, through the classic three component Strecker reaction of commercial benzaldehydes, cyclic secondary amines and KCN, a new series of nine α-amino nitriles, girgensohnine analogs, has been synthetized and screened for larvicide and adulticide properties against A. aegypti, one of the dominant vectors of dengue, chikungunya and zika in tropical and subtropical areas...

Oryzophagus oryzae (Costa Lima 1936) (Coleoptera: Curculionidae) and Spodoptera frugiperda (J E Smith, 1797) (Lepidoptera: Noctuidae) cause important crop losses in southern Brazil. Control is possible by the use of the bacteria Bacillus thuringiensis and extracts of Melia azedarach. This study aimed to evaluate the mortality, in vivo, of O. oryzae and S. frugiperda submitted to two isolates of B. thuringiensis and the aqueous extract of M. azedarach. The LC50 for O. oryzae due to bacteria was 5.40μg/mL (Bt 2014-2) and due to plant extract 0.90μg/mL. For S. frugiperda, the Bt 1958-2 bacterial suspension (1.10(10)UFC/mL) caused a 100% of corrected mortality, showing that the purified Cry proteins caused a CL10 of 268μg/mL five days after the treatments, and...

Expression of cry1Ab gene from a novel Bacillus thuringiensis strain SY49-1 active on pest insects BJM
Azizoglu,Ugur; Ayvaz,Abdurrahman; Yılmaz,Semih; Karabörklü,Salih; Temizgul,Rıdvan.
ABSTRACT In this study, the cry1Ab gene of previously characterized and Lepidoptera-, Diptera-, and Coleoptera-active Bacillus thuringiensis SY49-1 strain was cloned, expressed and individually tested on Ephestia kuehniella (Lepidoptera: Pyralidae) and Plodia interpunctella (Lepidoptera: Pyralidae) larvae. pET-cry1Ab plasmids were constructed by ligating the cry1Ab into pET28a (+) expression vector. Constructed plasmids were transferred to an Escherichia coli BL21 (DE3) strain rendered competent with CaCl2. Isopropyl β-D-1-thiogalactopyranoside was used to induce the expression of cry1Ab in E. coli BL21(DE3), and consequently, ∼130 kDa of Cry1Ab was obtained. Bioassay results indicated that recombinant Cry1Ab at a dose of 1000 µg g-1 caused 40% and 64%...
