Nd isotopes represent one of the best tools to investigate the processes involved in the evolution of the continental crust and mantle. This is due mainly to the similar geochemical behaviour of Sm and Nd, both light rare earth elements, which inhibits their fractionation during most varied geological processes. In order to carry out crustal evolution studies in central Brazil, the Sm-Nd isotopic method was implanted at the Geochronology Laboratory of the University of Brasília. The Sm-Nd separation methodology is basically that described in Richard et al. (1976), with the addition of some improvements. In this study we describe in detail the methodology used in Brasília. A prototoxina epsilon foi obtida a partir de cultivo líquido, rico em carboidratos, a partir de amostra de C. perfringens tipo D (INTA - Argentina), incubado em condições de anarobiose à 37ºC por oito horas. O sobrenadante da cultura foi concentrado utilizando-se o sistema Amicon com membrana de "cut off" de 10 kDa. O concentrado foi precipitado com sulfato de amônio (350 g/ml) e dissolvido em tampão fosfato 0,01 M (pH 7,2). A prototoxina purificada por cromatografia de troca iônica (DEAE-Sepharose CL 6B) e isolada no primeiro pico, mostrou uma banda única de aproximadamente 34kDa de peso molecular em SDS-PAGE, além de ser tóxica em camundongos. |

Highly purified intravenous immunoglobulin G concentrate (IV IgG) was produced with the use of polyethylene glycol associated to a single-stage precipitation by ethanol, instead of the classic Cohn-Oncley process, which employs cold alcohol as the precipitating agent, in a three-stage process. Precipitation of crude fraction containing more than 95% of immunoglobulin G was performed by liquid chromatography with a cation exchanger, CM-Sepharose, as a stationary phase. During the process, the product was subjected to two-stage viral inactivation. The first stage was performed by the action of sodium caprylate, 30 mM at pH 5.1+/- 0.1, and the second stage was performed by the action of a solvent-detergent mixture.
