Growth and extracellular laccase production in liquid cultures of Minimidochium parvum LPSC # 548 Strain Boletín de la Sociedad
Saparrat,Mario C. N.; Arambarri,Angélica M.; Balatti,Pedro A..
Minimidochium parvum LPSC # 548, a fungus isolated from litter floating on waters of Río Santiago (Provincia de Buenos Aires, Argentina) polluted with industrial effluents and crude-oil, was grown as a shaking culture on a C-limited medium to evaluate its ability to produce extracellular laccase. The effect of anthracene, CuSO4 · 5H2O, ethanol, guaiacol, humic acids, Kraft lignin, MnSO4· H2O, Tween 20 and veratryl alcohol on its growth and extracellular laccase activity levels was also analyzed. The cultures grown on basal medium produced maximum biomass (over 420 mg/100 ml) and maximum extracellular laccase activity (351.7 ±53.3 pkat/ml) after 5 days of incubation. Among the different factors tested, only the humic acids at 0.1 % (w/v) were found to...

Screening of Colletotrichum (Ascomycota) isolates, causal agents of Soybean Anthracnose, for Laccase production Boletín de la Sociedad
Levin,L.; Ramos,A .M.; Parisi,M .; Gally,M..
Colletotrichum truncatum is the most common pathogen fungus associated with soybean anthracnose. Although the lignin-degrading enzyme laccase has been implicated in pathogenicity of a wide range of plant pathogenic fungi, its biological role in the Colletotrichum -soybean disease system is unknown. The extent of the infection in our country led us to examine laccase production in Argentinean Colletotrichum strains isolated from diseased soybean plants from different geographic locations. Ten strains (eight of them identified as C. truncatum) , were screened for in vitro laccase production. Only six of the isolates, all of them C. truncatum , produced laccase activity when cultured on a defined medium based on pectin and asparagine as carbon and nitrogen...

Validation of computationally predicted substrates for laccase BABT
Reena,; Dhall,Purnima; Kumar,Rita; Kumar,Anil.
Present study reports the validation (oxidation) of computationally predicted oxidation of xenobiotic contaminants by commercially available pure laccase from Trametes versicolor. Selected contaminants were predicted as potential targets for laccase oxidation by using in-silico docking tool. The oxidation by laccase was measured by change in absorbance at specific λ max of each compound. Sinapic acid and tyrosine were taken as positive and negative controls, respectively. Oxidation was observed in m-chlorophenol, 2,4 di-chlorophenol, 2,4,6 tri-chlorophenol, captan, atrazine and thiodicarb, except malathion, which showed no activity. It could be speculated that the predicted substrates showing oxidation shared homology at structural and chemical level with...

Pentachlorophenol removal by Pleurotus pulmonarius in submerged cultures BABT
Souza,Daniela Farani de; Costa,Silvio Cláudio da; Dacome,Antonio Sérgio; Souza,Cristina Giatti Marques de; Bracht,Adelar; Peralta,Rosane Marina.
Pentachlorophenol (PCP) removal by Pleurotus pulmonarius grown in submerged cultures in the presence and absence of laccase inducers was studied in this work. When PCP was added to a final concentration of 25 mg·L-1 in submerged cultures actively producing laccase, 70% of the PCP was removed after 96 h. The removal of PCP was less than 20% in the cultures with low laccase activity. The results suggested that laccase played an important role in the biodegradation of PCP by P. pulmonarius and that for bioremediation purposes the fungus must be cultured under the conditions of active laccase production.

Copper induction and differential expression of laccase in Aspergillus flavus BJM
Gomaa,Ola M.; Momtaz,Osama A..
Aspergillus flavus was isolated from soil and exhibited laccase activity under both constitutive and copper induced conditions. Spiking the medium with 1 mM copper sulfate resulted in an increase in the activity which reached 51.84 U/mL, a distinctive protein band was detected at 60 kDa. The extracellular enzyme was purified 81 fold using gel filtration chromatography and resulted in two different laccase fractions L1 and L2, the latter had a higher enzymatic activity which reached 79.57 U/mL and specific activity of 64.17 U/μg protein. The analysis of the spectrum of the L2 fraction showed a shoulder at 330 nm which is characteristic for T2/T3 copper centers; both copper and zinc were detected suggesting that this is an unconventional white laccase....

Laccase produced by a thermotolerant strain of Trametes trogii LK13 BJM
Yan,Jinping; Chen,Yuhui; Niu,Jiezhen; Chen,Daidi; Chagan,Irbis.
Thermophilic and thermotolerant micro-organisms strains have served as the natural source of industrially relevant and thermostable enzymes. Although some strains of the Trametes genus are thermotolerant, few Trametes strains were studied at the temperature above 30 °C until now. In this paper, the laccase activity and the mycelial growth rate for Trametes trogii LK13 are superior at 37 °C. Thermostability and organic cosolvent tolerance assays of the laccase produced at 37 °C indicated that the enzyme possessed fair thermostability with 50% of its initial activity at 80 °C for 5 min, and could remain 50% enzyme activity treated with organic cosolvent at the concentration range of 25%–50% (v/v). Furthermore, the test on production of laccase and...

Camassola,Marli; Rosa,Letícia O. da; Calloni,Raquel; Gaio,Tamara A.; Dillon,Aldo J.P..
Pleurotus species secrete phenol oxidase enzymes: laccase (Lcc) and manganese peroxidase (MnP). New genotypes of these species show potential to be used in processes aiming at the degradation of phenolic compounds, polycyclic aromatic hydrocarbons and dyes. Hence, a screening of some strains of Pleurotus towards Lcc and MnP production was performed in this work. Ten strains were grown through solid-state fermentation on a medium based on Pinus spp. sawdust, wheat bran and calcium carbonate. High Lcc and MnP activities were found with these strains. Highest Lcc activity, 741 ± 245 U gdm-1 of solid state-cultivation medium, was detected on strain IB11 after 32 days, while the highest MnP activity occurred with strains IB05, IB09, and IB11 (5,333 ± 357; 4,701...

Lignocellulolytic enzyme production of Pleurotus ostreatus growth in agroindustrial wastes BJM
Luz,José Maria Rodrigues da; Nunes,Mateus Dias; Paes,Sirlaine Albino; Torres,Denise Pereira; Silva,Marliane de Cássia Soares da; Kasuya,Maria Catarina Megumi.
The mushroom Pleurotus ostreatus has nutritional and medicinal characteristics that depend on the growth substrate. In nature, this fungus grows on dead wood, but it can be artificially cultivated on agricultural wastes (coffee husks, eucalyptus sawdust, corncobs and sugar cane bagasse). The degradation of agricultural wastes involves some enzyme complexes made up of oxidative (laccase, manganese peroxidase and lignin peroxidase) and hydrolytic enzymes (cellulases, xylanases and tanases). Understanding how these enzymes work will help to improve the productivity of mushroom cultures and decrease the potential pollution that can be caused by inadequate discharge of the agroindustrial residues. The objective of this work was to assess the activity of the...

Screening of inducers for laccase production by Lentinula edodes in liquid medium BJM
Cavallazzi,José Renato P.; Kasuya,Catarina M.; Soares,Marcos A..
Laccases are enzymes involved in lignin degradation and are produced by various organisms. Due to their low substrate specificity their potential to be used in biotechnological applications has received attention. The addition of laccase inducers to the culture medium of microorganisms can enhance laccase production and facilitate its purification and utilization. The aim of this study was to investigate the effect of some compounds as laccase inducers in cultures of Lentinula edodes (shiitake). First, it was selected a culture medium suitable for laccase production by shiitake using two levels of N (2.6 and 26 mM) and seven levels of Cu (0, 50, 100, 150, 200, 250 and 300 µM). The medium with 2.6 mM N and 250 µM Cu was found to provide the highest laccase...

Ligninolytic enzymes production and Remazol brilliant blue R decolorization by tropical brazilian basidiomycetes fungi BJM
Machado,Kátia M. G.; Matheus,Dácio R.; Bononi,Vera L. R..
Remazol Brilliant Blue R (RBBR) dye was used as substrate to evaluate ligninolytic activity in 125 basidiomycetous fungi isolated from tropical ecosystems. The extracellular RBBR decolorizing activity produced when selected fungi were grown in solid media and in soil contaminated with organochlorines was also evaluated. A total of 106 fungi decolorized the RBBR during the growth in malt extract agar (MEA, 2%); 96 fungi showed a mycelia growth and decolorization activity stronger than the P. chrysosporium used as reference. Extracellular extracts of 35 selected fungi grown on solid medium with sugar cane bagasse (BGS) were evaluated for RBBR decolorization and peroxidase activity. All fungi showed peroxidase activities, but 5 of those were unable to...

Cloning, characterization and expression of a novel laccase gene Pclac2 from Phytophthora capsici BJM
Feng,Bao Zhen; Li,Peiqian.
Laccases are blue copper oxidases (E.C. 1.10.3.2) that catalyze the one-electron oxidation of phenolics, aromatic amines, and other electron-rich substrates with the concomitant reduction of O2 to H2O. A novel laccase gene pclac2 and its corresponding full-length cDNA were cloned and characterized from Phytophthora capsici for the first time. The 1683 bp full-length cDNA of pclac2 encoded a mature laccase protein containing 560 amino acids preceded by a signal peptide of 23 amino acids. The deduced protein sequence of PCLAC2 showed high similarity with other known fungal laccases and contained four copper-binding conserved domains of typical laccase protein. In order to achieve a high level secretion and full activity expression of PCLAC2, expression...
