Mineral supplementation and productivity of the Shiitake mushroom on eucalyptus logs Scientia Agricola
Queiroz,Elvio Cardoso; Marino,Regina Helena; Eira,Augusto Ferreira da.
Brazilian reports on Shiitake mushroom [Lentinula edodes (Berkeley) Pegler] productivity grown on Eucalyptus saligna (Sm.) logs are optimistic. The current production on this kind of wood is, however, low and variable. Aiming at increasing Shiitake mushroom productivity, the present work studied interactions among three concentrations of mineral supplement and three Shiitake strains (Le 95/01, 96/17 and 96/18) grown on seventh cut eucalyptus logs. Mineral fertilization was carried out when the logs were submerged in water to induce fruiting bodies, using the following saline concentrations: zero (no fertilizers added), 0.05% (equivalent to 0.35 g L-1 of ammonium sulfate with 18% nitrogen and 0.15 g L-1 of superphosphate with 34% phosphorus) and 0.50%....

Residual polysaccharides from fungi reduce the bacterial spot in tomato plants Bragantia
Aguiar,Tarsis; Luiz,Caroline; Rocha Neto,Argus Cezar; Di Piero,Robson Marcelo.
Abstract Polysaccharides from fungal wastes were partially characterized and evaluated for their protective effects against bacterial spot caused by Xanthomonas gardneri on four tomato cultivars: Santa Cruz Kada, Natália, BRS Sena and Forty. The polysaccharides were extracted from spent mushroom substrate of Pleurotus ostreatus, residual brewery yeast (Saccharomyces cerevisiae), and basidiocarps discarded from Lentinula edodes production. These polysaccharides were characterized for total carbohydrates, phenolics and proteins content, pH, scatter intensity, conductivity, Zeta potential, DPPH scavenging assay and infrared spectroscopy. The effects of time interval between treatment and inoculation (4 or 7 days) and polysaccharide concentrations (0.5 or 1.5...

Cellulase activity of a Lentinula edodes (Berk.) Pegl. strain grown in media containing Carboximetilcellulose or microcrystalline cellulose BABT
Pereira Júnior,José Antônio de Sousa; Correia,Marcos José; Oliveira,Neiva Tinti de.
Endoglucanase and exocellobiohydrolase produced b Lentinula edodes (Berk.) Pegl. strain thatt was cultivated in carboxymetilcellulose (CMC) or microcrystalline cellulose (Avicel) liquid media. The concentration and type of cellulose influenced the enzyme activity and production. Extra-cellular cellobiase activity was not detected in CMC or Avicel media. This enzyme was detected in mycelial extracts only. With 1.7% Avicel liquid medium, the strain did not produce exocellobiohydrolase, but 74 µmol RBBR/mg protein/min was detected with 0.5% Avicel. The substitution of Avicel by 0.5% CMC reduced this activity. Endoglucanase also had maximum activity in 0.5% Avicel medium (approximately 820 UI/mg protein) after 96 h incubation. In supernatants from 0.5% CMC,...

Antimicrobial activity and mineral composition of shiitake mushrooms cultivated on agricultural waste BABT
Casaril,Kérley Braga Pereira Bento; Kasuya,Maria Catarina Megumi; Vanetti,Maria Cristina Dantas.
The antimicrobial activity and mineral composition of shiitake mushrooms were evaluated in four isolates of Lentinula edodes. Mushrooms were cultivated on artificial logs, based on eucalyptus sawdust enriched with 20% rice, wheat, or soybean bran, or combination of 10% of two of these supplements. The substrates were humidified with a 0.1% mate tea extract or water. Logs of Eucalyptus grandis were also used to cultivate the shiitake mushrooms. The antimicrobial activity of an aqueous extract, corresponding to 40 mg of mushroom dry matter, was in some cases, depending on the isolate, able to inhibit both Bacillus subtilis and Escherichia coli K-12, independent of substrate composition or the growth stage of the mushrooms. Nitrogen, phosphorus, potassium,...

Supplementation of sugarcane bagasse with rice bran and sugarcane molasses for shiitake (Lentinula edodes) spawn production BJM
Rossi,Ivan Henrique; Monteiro,Antonio Carlos; Machado,José Octavio; Barbosa,José Carlos.
The purpose of this study was to assess the myceliation rate, mycelial vigor and "estimated biomass" of Lentinula edodes (Berk.) Pegler, grown on a sugarcane bagasse substrate enriched with rice bran and sugarcane molasses for spawn production. The proportions of rice bran used were 0, 10, 15, 20, 25, 30 and 40% (dry weight/dry weight of bagasse) and the sugarcane molasses concentrations tested were 0, 10, 20, 30, 40, 50 and 60 g/kg (dry weight/dry weight of bagasse plus rice bran). The myceliation rate was decreased by the addition of the higher quantities of rice bran. The 25 and 30% rice bran proportions induced the highest stimulation of mycelial vigor. The addition of sugarcane molasses did not change myceliation rate or mycelial vigor. The "estimated...

ANTIBACTERIAL ACTIVITY OF LENTINULA EDODES GROWN IN LIQUID MEDIUM BJM
Ishikawa,Noemia Kazue; Kasuya,Maria Catarina Megumi; Vanetti,Maria Cristina Dantas.
The antibacterial activity of 35 isolates of Lentinula edodes, a shiitake mushroom, against Bacillus subtilis was evaluated by diffusion technique in agar with a semi-solid overlay. All isolates inhibited B. subtilis and the isolate Le1 promoted the formation of the largest inhibition zone. L. edodes Le1 also presented antibacterial activity against foodborne pathogens and food contaminant bacteria, particularly Gram-positive species. The antibacterial activity of the culture filtrate after 18-25 days of cultivation of L. edodes in broth at 25ºC was high. The inhibitory activity was observed only in the organic layer when the culture filtrate was partitioned between ethyl acetate and water, suggesting that the inhibitory substances have low polarity. The...

Thermal and mechanical shocks affecting the first flush of production of Lentinula edodes on Eucalyptus saligna logs BJM
Shiomi,Humberto Franco; Minhoni,Marli Teixeira de Almeida; Machado,José Octávio; Cargnelutti Filho,Alberto.
The aim of this work was to evaluate the effect of thermal and mechanical shocks on the productivity of Lentinula edodes colonized on 140 Eucalyptus saligna logs at different immersion times in water and in the first flush of production. The logs were immersed in water at low temperature (16ºC) or environmental temperature (22ºC), for 6, 10, 14, 18, 22, 26 and 38 hours. The mechanical shock treatment was accomplished by dropping the logs onto the floor three consecutive times, in a vertical position. Water temperature and immersion time affected L. edodes yield, which increased two to four times when the logs were immersed in cool water for the shortest times (6 and 10 hours). The mechanical shock treatment did not increase the sporophore yield.
