Elevated atmospheric CO2 increases Eucalyptus urophylla S. T. Blake stem diameter by stimulating cell proliferation and reducing lignin deposition Acta Botanica
Innocente,Patricia Sitta; Teixeira,Simone Pádua; Balbuena,Tiago Santana.
ABSTRACT In 2019, the atmospheric CO2 concentration exceeded the 415 ppm milestone for the first time in the human history. According to projections of the Intergovernmental Panel on Climate Change (IPCC), CO2 levels will continue to rise in the future, potentially affecting all living organisms. Plants with C3 metabolism may benefit from rising CO2 levels because the significant losses of photosynthesis, driven by photorespiration could be diminished under this scenario. This study addressed the anatomical changes in the stems of young Eucalyptus urophylla plants induced through cultivation in elevated CO2 (eCO2). Plants cultivated under eCO2 showed increased stem diameter (i.e., radial width of the secondary xylem, secondary phloem and cortex tissues)....

Primary leaf growth and coleoptile length in maize seedlings Agronomy
Lopes, Camila Aparecida; Carvalho, Maria Laene Moreira de; Vasconcelos, Michelle Conceição; Ribeiro, Ana Maria Pereira; Santos, Heloísa Oliveira dos.
Evaluation of the physiological potential of maize seeds during the certification process is performed by a germination test. However, some tests show a delay in the growth of aerial seedling organs that does not reflect seed lot behavior in the field. This study aims to investigate the effects of genotype, water, light and temperature on differential primary leaf growth compared to coleoptile length in maize seedlings. Seeds from two simple hybrids and their reciprocal crosses that showed slow primary leaf growth were evaluated according to physiological quality, morphology, histochemistry, and enzymatic expression. There were differences among maize genotypes regarding initial primary leaf growth when compared to the coleoptile, especially when...

Zonetti, Patricia da Costa; Cesumar; Suzuki, Letícia Sayuri; UEM; Bonini, Edicléia Aparecida; UEM; Ferrarese, Maria de Lourdes Lucio; UEM; Ferrarese Filho, Osvaldo; UEM.
Glyphosate resistance is conferred to soybean (Glycine max L. Merril) by incorporating a gene encoding a glyphosate-insensitive enzyme (CP4-EPSP synthase) that acts in the shikimate/chorismate pathway, an important metabolic route in the lignification process. The aim of this work was to investigate the root growth and lignin contents of susceptible (CD 201 and OC 14) and glyphosate-resistant (CD 214RR and CD 213RR) soybean cultivars. To this end, three-day-old seedlings were cultivated in half-strength Hoagland nutrient solution (pH 6.0) in a growth chamber (25°C, 12-h photoperiod, irradiance of 280 µmol m-2 s-1) for 24 to 96 hours. The results revealed that glyphosate-resistant (CD 213RR and CD 214RR) cultivars showed high root growth when...

Optimization of RT-PCR reactions in studies with genes of lignin biosynthetic route in Saccharum spontaneum Anais da ABC (AABC)
LLERENA,JUAN P.P.; ARAÚJO,PEDRO; MAZZAFERA,PAULO.
ABSTRACT Saccharum spontaneum has been used for the development of energy cane a crop aimed to be used for the production of second-generation ethanol, or lignocellulosic ethanol. Lignin is a main challenge in the conversion of cell wall sugars into ethanol. In our studies to isolate the genes the lignin biosynthesis in S. spontaneum we have had great difficulty in RT-PCR reactions. Thus, we evaluated the effectiveness of different additives in the amplification of these genes. While COMT and CCoAOMT genes did not need any additives for other genes there was no amplification (HCT, F5H, 4CL and CCR) or the yield was very low (CAD and C4H). The application of supplementary cDNA was enough to overcome the non-specificity and low yield for C4H and C3H, while...

A supramolecular proposal of lignin structure and its relation with the wood properties Anais da ABC (AABC)
Abreu,Heber S.; Latorraca,João V.F.; Pereira,Regina P.W.; Monteiro,Maria Beatriz O.; Abreu,Fábio A.; Amparado,Kelysson F..
In spite of the great importance of cellulose the lignin is considered the second most abundant substance of the wood. However, little attention has been given it, mainly to wood properties. The lignin as well as other structural compounds (cellulose and hemicelluloses), has obviously an important role on the wood properties, probably due its composition and existent bonds. In general lignins have β-O-4 (Alkyl Aril Ether) as majoritary bond. This bond in a continued structure form big molecules with spiral conformation as virtual model. Based on this idea, lignins that have high/low β-O-4 content may have differentiated spiraled structures, suggesting different behaviors on the wood properties, which shows that the lignins (Guaicyl:Syringyl (GS)) of...

Lidija,Begović; Lepeduš,Hrvoje; Lalić,Alojzije; Štolfa,Ivna; Jurković,Zorica; Kovačević,Josip; Cesar,Vera.
ABSTRACT The aim of this study was to investigate anatomical characteristics of barley (Hordeum vulgare L.) internodes and to reveal the connection with activity of guaiacol-peroxidases, their isoenzyme profiles, lignin, and cellulose content. Three lower internodes were sampled at elongation stage and differed in their anatomical features by showing that lignin deposition follows developmental pattern. Epidermis and hypodermis in the first internode were characterized by continuous layer of the lignified cells while in the second and third internode thin walled parenchyma cells could be observed in the hypodermis. These observations were in accordance with increment of peroxidase activity from the first to the third internode. Two acidic and 2 basic...

Naringenin inhibits the growth and stimulates the lignification of soybean root BABT
Bido,Graciene de Souza; Ferrarese,Maria de Lourdes Lucio; Marchiosi,Rogério; Ferrarese-Filho,Osvaldo.
The flavanone naringenin, an intermediate in flavonoid biosynthesis, was tested for its effect on root growth, phenylalanine ammonia-lyase (PAL) and peroxidase (POD) activities, as well as phenolic compounds and lignin contents in soybean (Glycine max L. Merrill) seedlings. Three-day-old seedlings were cultivated in half-strength Hoagland nutrient solution (pH 6.0), with or without 0.1 to 0.4 mM naringenin in a growth chamber (25°C, 12-h photoperiod, irradiance of 280 µmol m-2 s-1) for 24 h. Inhibitory effects on root growth (length, weight, cell viability), PAL and soluble POD activities were detected after naringenin treatments. These effects were associated with stimulatory activity of the cell wall-bound POD followed by an increase in the lignin...

Gréggio,Thiago Claudino; Assis,Luiz Carlos; Nahas,Ely.
The decomposition of soil litter contributes to maintaining agricultural sustainability, since the nutrients released by microbial activity are determinants of the net productivity of the agroecosystem. The decomposition of rubber tree (Hevea brasiliensis [Willd. ex A. Juss.] Müll. Arg.) leaves located on the surface and buried in at 10 cm depth in Forest and Savannah (Cerradão) soils (Oxisols) was studied, with emphasis on the production of CO2 and the monthly variation of the remaining amounts of litter mass, soluble substances, cellulose and lignin. To evaluate CO2 production, H. brasiliensis leaves were incubated for 30 days in soils from the 0-2 cm and 10-12 cm layers. CO2 production increased in the 0-2 cm layer in comparison to the 10-12 cm layer....

Use of markers to determine the ileal digestibility of amino acids for swine Ciência e Agrotecnologia
Pozza,Paulo Cesar; Gomes,Paulo Cezar; Pozza,Magali Soares Santos; Nunes,Ricardo Vianna; Ferreira,Rony Antônio; Rodrigueiro,Ramalho José Barbosa.
Many markers can be used in digestibility assays for swine, but needs to be evaluated before its use. An assay was carried out with the objective of measuring the coefficients of ileal apparent (CIAD) and standardized digestibility (CISD) of amino acids in meat and bone meal for swine using different markers. Eight crossbred barrows were equipped with T-cannulas in the distal ileum. Four animals received a free protein diet and another four received a diet containing meat and bone meal as the only protein source in the diet. The treatments consisted in evaluating the chromic oxide, acid-insoluble ash naturally occurring in the diet, cellulose and lignin as markers to determine the apparent and standardized digestibility of the amino acids contained in the...

Generation and analysis of an Eucalyptus globulus cDNA library constructed from seedlings subjected to low temperature conditions Electron. J. Biotechnol.
Rasmussen-Poblete,Susana; Valdes,Jorge; Gamboa,Maria Cecilia; Valenzuela,Pablo D.T; Krauskopf,Erwin.
Eucalyptus globulus is the most important commercial temperate hardwood in the world because of its wood properties and due to its characteristics for biofuel production. However, only a very low number of expressed sequence tags (ESTs) are publicly available for this tree species. We constructed a cDNA from E. globulus seedlings subjected to low temperature and sequenced 9,913 randomly selected clones, generating 8,737 curated ESTs. The assembly produced 1,062 contigs and 3,879 singletons forming a Eucalyptus unigene set. Based on BLASTX analysis, 89.3% of the contigs and 88.5% of the singletons had significant similarity to known genes in the non-redundant database of GenBank. The Eucalyptus unigene set generated is a valuable public resource that...

Genes encoding enzymes of the lignin biosynthesis pathway in Eucalyptus Genet. Mol. Biol.
Harakava,Ricardo.
Eucalyptus ESTs libraries were screened for genes involved in lignin biosynthesis. This search was performed under the perspective of recent revisions on the monolignols biosynthetic pathway. Eucalyptus orthologues of all genes of the phenylpropanoid pathway leading to lignin biosynthesis reported in other plant species were identified. A library made with mRNAs extracted from wood was enriched for genes involved in lignin biosynthesis and allowed to infer the isoforms of each gene family that play a major role in wood lignin formation. Analysis of the wood library suggests that, besides the enzymes of the phenylpropanoids pathway, chitinases, laccases, and dirigent proteins are also important for lignification. Colocalization of several enzymes on the...
