Effect of the chronic administration of caffeine on adipose mass and lipid profile of Wistar rats - doi: 10.4025/actascibiolsci.v35i2.11085 Biological Sciences
Nakagawa, Priscila; Universidade Estadual de Maringá; Pedrosa, Maria Montserrat Diaz; Universidade Estadual de Maringá.
This work aimed at verifying the effect of the chronic ingestion of caffeine on body weight and adiposity of Wistar rats. Sixteen male Wistar rats weighting on average 240 g were divided into two groups, one control and the other caffeine-treated (5 mg kg-1, orally) for five weeks. At the end, the groups were evaluated for their differences in body weight; weight of the periepididymal, retroperitoneal, subcutaneous and mesenteric fat pads; size of the retroperitoneal adipocytes; liver and heart weight; glycemia and plasma lipids. Statistically significant differences were observed in adipocyte size and total serum cholesterol, while the results for the other parameters were not statistically different. Therefore, this study showed that, using an oral dose...

Lipolytic response of adipose tissue and metabolic adaptations to long periods of fasting in red tilapia (Oreochromis sp., Teleostei: Cichlidae) Anais da ABC (AABC)
DIAS JUNIOR,WALTER; BAVIERA,AMANDA M.; ZANON,NEUSA M.; GALBAN,VICTOR D.; GARÓFALO,MARIA ANTONIETA R.; MACHADO,CELIO R.; BAILÃO,ELISA F.L.C.; KETTELHUT,ISIS C..
ABSTRACT Adaptive changes of carbohydrate and lipid metabolism induced by 7, 15, 30, 60, 90, 150 and 200 days of fasting were investigated in red tilapia (Oreochromis sp.). Plasma glucose, lactate and free fatty acids (FFA) levels, liver and muscle glycogen and total lipid contents and rates of FFA release from mesenteric adipose tissue (MAT) were measured. Plasma glucose levels showed significant differences only after 90 days of fasting, when glycemia was 34% lower (50±5mg.dL-1) than fed fish values (74±1mg.dL-1), remaining relatively constant until 200 days of fasting. The content of liver glycogen ("15%) in fed tilapia fell 40% in 7 days of food deprivation. In 60, 90 and 150 days of fasting, plasma FFA levels increased 49%, 64% and 90%, respectively,...

Differences in lipogenesis and lipolysis in obese and non-obese adult human adipocytes Biol. Res.
CIFUENTES,MARIANA; ALBALA,CECILIA; ROJAS,CECILIA V.
It has been proposed that differences in adipocyte function and/or metabolism between obese and lean individuáis may manifest themselves in functional adipose tissue abnormalities that lead to metabolic disorders in obesity. We studied lipogenesis and lipolysis of omental adipocytes from obese (OB) and non-obese (NOB) humans. The specific activity of the lipogenic marker enzyme G3PDH was 50% lower in total adipocytes of OB compared to that of NOB subjects. Omental adipocytes from OB subjects also had lower basal lipolytic levéis, and a lower lipolytic response to p-adrenergic stimulus. Cholesterol depletion of adipocyte plasma membrane using methyl β-cyclodextrin caused a lipolytic effect on adipocytes of both groups together, but when obese and...

Effect of ractopamine on lipid metabolism in vivo - a systematic review BABT
Ferreira,Matheus Soares da Silva; Garbossa,Cesar Augusto Pospissil; Oberlender,Guilherme; Pereira,Luciano José; Zangeronimo,Márcio Gilberto; Sousa,Raimundo Vicente de; Cantarelli,Vinícius de Souza.
The aim of this article was to evaluate the in vivo mechanism of action through which ractopamine reduces the amount of adipose tissue in carcass of animals fed diets supplemented with this drug. Literature search was carried out, using the keywords Ractopamine, lipogenesis, lipolysis, fatty acid and adipose tissue. The scoring system was designed for the adequacy of the methodology. A total of eight papers were found - all published in the English language. The determination of lipolysis and lipogenesis was carried out using different methods - from the quantification of non-esterified fatty acids in the blood to molecular analyses of adipose tissue. The review analysis suggested that, at least in the pigs, the use of this drug reduced lipid deposition in...

Nuclear translocation of STAT3 by in vitro metreleptin administration causes lipolysis in human primary adipocytes BABT
Choi,Seung Kug; Park,Sunmi; Moon,Hyun-Seuk.
We utilized subcutaneous (SC)- and omental (OM)-derived human primary adipocytes (hPA) from obese male, and investigated whether synthetic analog of leptin, metreleptin, may regulate lipolysis via translocation of STAT3 to the nucleus. We observed that 50 ng/mL of metreleptin increases STAT3 phosphorylation in both SC- and OM-derived hPA. Importantly, we found for the first time that metreleptin is capable of trans-locating STAT3 to the nucleus and STAT3 blockade inhibits metreleptin-induced lipolysis. Our initial data provide novel insights into the role of STAT3 as probable mediator of the action of metreleptin in regulating metabolism.

New concepts in white adipose tissue physiology BJMBR
Proença,A.R.G.; Sertié,R.A.L.; Oliveira,A.C.; Campaãa,A.B.; Caminhotto,R.O.; Chimin,P.; Lima,F.B..
Numerous studies address the physiology of adipose tissue (AT). The interest surrounding the physiology of AT is primarily the result of the epidemic outburst of obesity in various contemporary societies. Briefly, the two primary metabolic activities of white AT include lipogenesis and lipolysis. Throughout the last two decades, a new model of AT physiology has emerged. Although AT was considered to be primarily an abundant energy source, it is currently considered to be a prolific producer of biologically active substances, and, consequently, is now recognized as an endocrine organ. In addition to leptin, other biologically active substances secreted by AT, generally classified as cytokines, include adiponectin, interleukin-6, tumor necrosis factor-alpha,...

Renin-angiotensin system blockers regulate the metabolism of isolated fat cells in vitro BJMBR
Caminhotto,R de O.; Sertié,R.A.L.; Andreotti,S.; Campaãa,A.B.; Lima,F.B..
Due to the presence of the renin-angiotensin system (RAS) in tissues and its specific influence on white adipose tissue, fat cells are possible targets of pharmacological RAS blockers commonly used as anti-hypertensive drugs. In the present study, we investigated the effects of different RAS blockers on fat cell metabolism, more specifically on lipolysis, lipogenesis and oxidation of energy substrates. Isolated primary adipocytes were incubated with different RAS blockers (aliskiren, captopril and losartan) in vitro for 24 h and lipolysis, lipogenesis and glucose oxidation capacities were determined in dose-response assays to a β-adrenergic agonist and to insulin. Although no change was found in lipolytic capacity, the RAS blockers modulated lipogenesis...

The addition of the thermophilic esterase EST2 influences the fatty acids and volatile compound profiles of semi hard cheeses Ciênc. Tecnol. Aliment.
DE LUCA,Valentina; PEROTTI,Maria Cristina; WOLF,Irma Veronica; MEINARDI,Carlos Alberto; MANDRICH,Luigi.
Abstract During cheese ripening the hydrolytic activities of proteases and lipases are essential to develop typical flavours; usually these activities come from lactic bacteria. A way to increase cheese flavour and/or to reduce the time of ripening is the addition of exogenous enzymes during cheese making. Actually some commercial lipases are used in cheese making, here we have evaluated the effect of the addition of the thermophilic esterase EST2, from Alyciclobacillus acidocaldarius, on the lipolysis and volatile compounds profiles of semi-hard cheeses. The addition of EST2 produced an increase of the degree of lipolysis up to 30% and the relative proportion of short- and medium- chain fatty acids, with respect to the control cheese. By SPME-GC-FID/MS...

Spoilage potential of Paenibacillussp. in Brazilian raw milk Ciência Rural
Ribeiro Júnior,José Carlos; Alcântara,Brígida Kussumoto de; Beloti,Vanerli.
ABSTRACT: Bacterial spores are widespread in the environment and can contaminate milk. Spores are resistant to thermal conditions and your germination reduces milk shelf-life because the aerobic bacteria that are sporulated produce proteases and lipases. The aim of this study was identify Paenibacillus sp., the spoilage microbiota, arising from the germination of spores in raw milk and your spoilage potential. Twenty different milk samples were treated at 80°C/12min and plated to isolate spore-forming bacteria. These strains were picked in milk agar and tributyrin agar for verification of their potential proteolytic and lipolytic activities, respectively. Amplification and sequencing of the 16S rRNA gene of the strains for identification by similarity to...
