Inhibition mechanism of Listeria monocytogenes by a bioprotective bacteria Lactococcus piscium CNCM I-4031 ArchiMer

Listeria monocytogenes is a pathogenic Gram positive bacterium and the etiologic agent of listeriosis, a severe food-borne disease. Lactococcus piscium CNCM I-4031 has the capacity to prevent the growth of L. monocytogenes in contaminated peeled and cooked shrimp. To investigate the inhibititory mechanism, a chemically defined medium (MSMA) based on shrimp composition and reproducing the inhibition observed in shrimp was developed. In co-culture at 26°C, L. monocytogenes was reduced by 3-4 log CFU g-1 after 24 h. This article describes a method for high-throughput competition assays using a bioluminescent strain of L. monocytogenes. This method is based on the use of the luminescent indicator strain L. monocytogenes EGDelux. The luminescence of this strain is correlated to growth, which make it suitable to monitor the growth of L. monocytogenes in mixed cultures. Microbial seafood-borne disease represents 10 to 20% of the total food-borne outbreaks. Most of them are from bacterial origin and involve seafood products that have been contaminated by pathogenic Vibrio, Listeria monocytogenes and histaminogen bacteria. The aim of this study was to demonstrate the inhibitory capacity of Carnobacterium strains against a collection of Listeria monocytogenes strains in cold-smoked salmon (CSS). Methods and Results: Three bacteriocin-producing strains, Carnobacterium divergens V41, C. piscicola V1 and C. piscicola SF668, were screened for their antilisterial activity against a collection of 57 L. monocytogenes strains selected from the French smoked salmon industry, using an agar spot test. In order to develop a biopreservation strategy in cold-smoked salmon, the inhibitory capacity of 3 bacteriocin-producing Carnobacterium strains isolated from fish have been tested towards Listeria monocytogenes strains. C. divergens V41 maintains the level of Listeria below 50 CFU/g in sterile cold-smoked salmon during the 28 days of vacuum storage at 4 and 8°C, thanks to the divercin V41 action. For its use in the product, the culture of the strain was optimized in a growth medium without animal protein. On commercial cold-smoked salmon slices, the strain inhibits slightly the endogenous microflora and doesn't modify the sensorial quality, but it produces tyramine, a biogenic amine. A total of 30 strains of Listeria monocytogenes isolated from different foods (16 of differents kinds of sausage, 14 cheese,) purchased at groceries of São Paulo City were ribotyped and analysed for the presence and expression of hemolysin gene and production of phosphatidylinositol-specific phosphalipase C - PI-PLC enzyme. The L. monocytogenes strains were differentiated into six ribotype classes. Se caracterizaron in vitro 17 cepas de Lactobacillus spp. aisladas del tracto intestinal de terneros neonatos, para seleccionar las mejores como candidatos probióticos y realizarles posteriormente los estudios in vivo. Ocorrência de Listeria monocytogenes em queijo artesanal tipo coalho comercializado na cidade de Manaus-AM, Brasil Acta Amazonica

A ocorrência de Listeria sp. em queijo artesanal tipo coalho comercializado na cidade de Manaus foi estudada. No período de março a maio de 1998 foram coletadas 58 amostras do produto em feiras e mercados localizados em seis zonas da cidade de Manaus. Em duas amostras de queijo foram identificadas duas espécies de Listeria: L. monocytogenes e L. innocua. A primeira espécie é reconhecidamente patogênica para o ser humano. Os resultados apresentados demonstraram haver risco de transmissão de enfermidades pelo consumo de queijos processados sem os devidos cuidados de higiene.

Actividad antagonista de bacterias lácticas aisladas del medio marino contra cepas de Listeria ABCL

En el presente trabajo se estudiaron 74 bacterias lácticas para determinar su capacidad de inhibir el desarrollo de Listeria monocytogenes ATCC 7644 o L. innocua ATCC 33090. Dentro de las 7 que exhibieron actividad, 2 se seleccionaron para estudios complementarios y se identificaron por métodos fenotípicos y genotípicos como Enterococcus mundtii Tw 56 y Lactococcus lactis subsp. lactis Tw 34. Relatam-se dois casos de listeriose ocorridos em ovinos leiteiros da raça Lacaune, mantidos em confinamento e alimentados com silagem de milho. O exame histológico do tecido nervoso mostrou meningoencefalite supurada, e a imunoistoquímica foi positiva para Listeria monocytogenes com isolamento da bactéria do tecido nervoso. Enfatiza-se a ocorrência dessa enfermidade em ovinos leiteiros que apresentavam lesões na cavidade oral e eram alimentados com silagem de baixa qualidade.

The presented study aimed to verify the effect of different pH values, enzyme solutions and heat treatments on the antimicrobial activity of the bacteriocinogenic strain Lactococcus lactis subsp. lactis Lc08 and to test their antimicrobial activity against Listeria monocytogenes in reconstituted skim milk at refrigeration temperatures. Avaliou-se a qualidade microbiológica da silagem produzida em Portugal e otimizaram-se alguns aspectos relacionados com a eficácia de detecção de Listeria monocytogenes mediante protocolo baseado na técnica de hibridação in situ fluorescente (FISH) para detecção directa desse microrganismo em amostras de silagens. O protocolo foi aplicado a 74 amostras, em simultâneo com o método bacteriológico convencional. Este último permitiu a detecção de L. monocytogenes em 11 silos (15%). ABSTRACT This study focused on assessing the microbiology of conveyor belts surfaces, cleaned or not with pressurized water, and verification of the influence of this process on the microbiological quality of poultry meat. A reduction on mesophilic, psychrotrophic and Enterobacteriaceae counts (P<0.05) on dry conveyor belts compared to wet ones was observed. For the chicken leg (consisting of drumstick and thigh) samples, no statistically significant differences were detected on counts of indicators of microorganisms on poultry legs (composed by drumstick and thigh). Effect of eugenol on growth and listeriolysin o production by Listeria monocytogenes BABT

The inhibitory effect of eugenol, a naturally occurring compound mainly present in the essential oil fraction of cloves, was studied on the growth and listeriolysin O (LLO) production by Listeria monocytogenes. Potassium efflux from cells promoted by eugenol was also determined after 24 h incubation in phosphate buffered saline. Eugenol promoted a delay on the growth of L. monocytogenes at concentrations of 100, 300 and 500 µg mL-1and above 800 µg mL-1 the effect was bactericidal. Production of LLO by L. monocytogenes in the presence of eugenol was reduced 80-100%. Antigenic characterization defined 49% of serotype 4b samples, followed by 28% of serotype 1/2b, 14% of serotype 1/2c, 8% of serotype 1/2a, and 1% of serotype 3b. Both type of samples from human and food origin express the same serotype distribution. Multiplex PCR analysis showed 13 strains of type 4b with the amplification profile 4b-VI (Variant I). Virulence genes hly, inlA, inlB, inlC, inlJ, actA, plcA, and prfA were detected in all samples, highlighting a deletion of 105pb on the actA gene in 23% of serotype 4b...
