Avaliou-se a qualidade microbiológica da silagem produzida em Portugal e otimizaram-se alguns aspectos relacionados com a eficácia de detecção de Listeria monocytogenes mediante protocolo baseado na técnica de hibridação in situ fluorescente (FISH) para detecção directa desse microrganismo em amostras de silagens. O protocolo foi aplicado a 74 amostras, em simultâneo com o método bacteriológico convencional. Este último permitiu a detecção de L. monocytogenes em 11 silos (15%). Por meio do protocolo FISH, observou-se que 22 silos (29,7%) se encontravam contaminados. ABSTRACT This study focused on assessing the microbiology of conveyor belts surfaces, cleaned or not with pressurized water, and verification of the influence of this process on the microbiological quality of poultry meat. A reduction on mesophilic, psychrotrophic and Enterobacteriaceae counts (P<0.05) on dry conveyor belts compared to wet ones was observed. For the chicken leg (consisting of drumstick and thigh) samples, no statistically significant differences were detected on counts of indicators of microorganisms on poultry legs (composed by drumstick and thigh). ABSTRACT The herein presented assay provided a bacteriological and molecular characterization of 100 samples of L. monocytogenes isolated from human (43) and food (57) sources, from several regions of Brazil, and collected between 1975 and 2013. Antigenic characterization defined 49% of serotype 4b samples, followed by 28% of serotype 1/2b, 14% of serotype 1/2c, 8% of serotype 1/2a, and 1% of serotype 3b. Both type of samples from human and food origin express the same serotype distribution. Multiplex PCR analysis showed 13 strains of type 4b with the amplification profile 4b-VI (Variant I). Fluctuations in the Brazilian milk market force small milk producers to find temporary trade alternatives, which include selling raw milk to people who prefer this type of milk rather than heat-processed milk. Considering the lack of information available in Brazil concerning listeriosis, the present study aimed to analyze phenotypic and genotypically Listeria monocytogenes clinical isolates from the Southwestern region of the State of São Paulo, Brazil. From January 1995 to May 2005, thirteen isolates of L. monocytogenes from twelve patients with listeriosis were sent to the Adolfo Lutz Institute (Campinas Regional Laboratory, São Paulo, Brazil). ABSTRACT The inhibition of Listeria monocytogenes ATCC 7644 on fresh-cut tomato was investigated using nisin alone, and in combinations with organic salts. Nisin at a concentration of 5000 UI/mL was introduced alone or in combination with an organic salt (sodium citrate or sodium acetate each at 3 and 5 g/100 mL each) on fresh-cut tomato previously inoculated with 108 CFU/mL of L. monocytogenes ATCC 7644. This study evaluated the growth of naturally occurring L. monocytogenes in sliced, vacuum-packed mortadella samples during storage at 5ºC until the expiration date. Tukey's test indicated that counts of L. monocytogenes on 0, 10, 20, 30 and 40 days of storage were significantly different (p<0.05), indicating growth during shelf life. In three trials, the mean increase was 1.72 log cycles. Vacuum packing and storage under refrigeration were not effective in controlling the growth of L. monocytogenes in sliced mortadella, indicating that good manufacturing practices and implemented HACCP programs are essential to assure safety of this product. |
