Crustacean hyperglycemic hormone (CHH) is the most abundant and best studied member of the CHH/MIH/GIH neuropeptide hormone family. CHH plays a major role in controlling glucose levels in the hemolymph, and it also has significance in regulating molting, reproduction, and osmoregulation. In contrast, molt-inhibiting hormone (MIH) is responsible for maintaining animals in an intermolt stage. The Pacific white shrimp, Litopenaeus vannamei (Penaeidae), represents about 95% of all Brazilian shrimp production. The Brazilian L. vannamei foundation broodstock was made up of specimens collected from different American Pacific sites, but little information was collected on the genetic structure of the broodstock. We used the fluorescence amplified fragment length polymorphism (fAFLP) method to study the genetic diversity of L. vannamei broodstock lines 03CMF1 and 03CBF1 originally produced by breeder-shrimps imported mainly from Panama and Ecuador, although wild individuals from other localities may also have been used in producing these two lines. We investigated the effects of family selection on the growth rates of four genetic lines of Litopenaeus vannamei, the Pacific white shrimp, from Venezuela. The mean family weight at selection was 16.79 g and the mean growth rate was 0.169 g d-1 at a mean age of 138 days. The mean growth rates per generation were 0.141 g d-1 for the parental generation (P0), 0.173 g d-1 for the F1 and 0.191 g d-1 for the F2. Survival varied from 83.3% to 94.4%. There were statistically significant differences between lines, with lines B and C, growing at higher temperatures and lower salinities, showing the fastest growth. A study was conducted to evaluate variations of digestive enzyme activities in Litopenaeus vannamei (Boone) reared in commercial ponds under semi-intensive conditions. Shrimp were collected at each body weight increase of 2 g. As the shrimp grew (2-12 g), significant increases in the activities of lipase and chymotrypsin were observed. The total protease activity decreased from 6 g onwards. Trypsin activity showed a peak at 6 g and amylase activity increased two-fold after 2 g. Improvement in the osmoregulation capacity via nutritional supplies is vitally important in shrimp aquaculture. The effects of dietary protein levels on the osmoregulation capacity of the Pacific white shrimp (L. vannamei) were investigated. This involved an examination of growth performance, glutamate dehydrogenase (GDH) and Na+-K+ ATPase mRNA expression,, and GDH activity in muscles and gills. The pathogenicity of Vibrio penaeicida Strains KH-1 and AM101, their culture-free supernatant (CFS), and their protein fraction obtained by 40% of ammonium sulfate precipitation (PFs40) were assessed in experimental challenges against juvenile Litopenaeus vannamei. Live Vibrio cells, CFS, and PFs40 from the AM101 strain produced a significantly higher mortality (p < 0.05) compared to the KH-1 strain. We evaluated changes in gene expression of Litopenaeus vannamei shrimp in response to a change in diet by comparing specimens which were fed with an animal protein diet against those which underwent a change from an animal protein diet to a plant protein diet. Biochemical changes, specifically in hemocyanin and glucose concentrations in hemolymph, glycogen in the digestive gland. were studied throughout the molt cycle in juveniles of Litopenaeus vannamei in a high salinity condition (36), and related to variations in the osmotic capacity of organisms. O objetivo deste trabalho foi avaliar a proteção antiviral específica via RNA de interferência (RNAi) contra o vírus da síndrome da mancha-branca (WSSV), em camarões marinhos (Litopenaeus vannamei). Os camarões foram injetados com uma sequência dsRNA específica (vp28 do envelope viral), seguida por desafio com WSSV após 48 horas. Avaliaram-se o hemograma às 0, 3, 6, 24, 48 e 72 horas após o desafio, e a taxa de mortalidade durante 30 dias. Nos animais tratados com dsRNA vp28, a infecção viral foi limitada, e a sobrevivência (73%) e a "clearance" viral (80%) foram maiores do que nos camarões infectados, não tratados, que apresentaram 100% de mortalidade em cinco dias. The objective of this work was to select surviving breeders of Litopenaeus vannamei from white spot syndrome virus (WSSV) outbreak, adapted to local climatic conditions and negatively diagnosed for WSSV and infectious hypodermal and hematopoietic necrosis virus (IHHNV), and to evaluate if this strategy is a viable alternative for production in Santa Catarina, Brazil.
