Diet high in α-linolenic acid up-regulate PPAR-α gene expression in the liver of goats Electron. J. Biotechnol.
Ebrahimi,Mahdi; Rajion,Mohamed Ali; Meng,Goh Yong; Farjam,Abdoreza Soleimani; Oskoueian,Ehsan; Jafari,Saeid.
Background There is little information on the effects of diets containing high α-linolenic acid (C18:3n-3) on liver lipid composition and lipogenic gene expressions. In this study fourteen goats (Capra aegagrus hircus) were fed either a flaxseed oil (FSO) supplemented diet containing high α-linolenic acid or a control diet without added flaxseed oil (CON) for 100-d to evaluate the effects on liver lipid composition and the gene expression of peroxisome proliferator-activated receptors (PPAR-α) and stearoyl-CoA-desaturase (SCD) in the liver. Results An increase in the levels of C18:3n-3 and C20:5n-3, C22:5n-3, C22:6n-3 was observed in the liver of FSO-treated goats. There was a significant (P < 0.05) up-regulation of...

Differential gene expression between wild-type and Gulo-deficient mice supplied with vitamin C Genet. Mol. Biol.
Jiao,Yan; Zhang,Jifei; Yan,Jian; Stuart,John; Gibson,Griffin; Lu,Lu; Willaims,Robert; Wang,Yong Jun; Gu,Weikuan.
The aim of this study was to test the hypothesis that hepatic vitamin C (VC) levels in VC deficient mice rescued with high doses of VC supplements still do not reach the optimal levels present in wild-type mice. For this, we used a mouse scurvy model (sfx) in which the L-gulonolactone oxidase gene (Gulo) is deleted. Six age- (6 weeks old) and gender- (female) matched wild-type (WT) and sfx mice (rescued by administering 500 mg of VC/L) were used as the control (WT) and treatment (MT) groups (n = 3 for each group), respectively. Total hepatic RNA was used in triplicate microarray assays for each group. EDGE software was used to identify differentially expressed genes and transcriptomic analysis was used to assess the potential genetic regulation of Gulo...

Histology of the liver of Oligosarcus jenynsii (Ostariophysi, Characidae) from Los Padres Lake, Argentina Iheringia, Sér. Zool.
Petcoff,Gladys M.; Díaz,Alcira O.; Escalante,Alicia H.; Goldemberg,Adriana L..
The liver is an organ notable for its sensitivity to a great variety of environmental factors. It is composed of a parenchyma divided into irregular lobules by the exocrine pancreas or hepatopancreas; the pancreatic cells are arranged around a branch of the portal vein. The hepatocytes are radially arranged in cords around a central sinusoid. The liver histomorphology and the organization of exocrine pancreatic tissue of O. jenynsii (Günther, 1864) is similar to the acinar morphology of many teleosts, including freshwater and marine species. The aim of the present work was to carry out the histological analysis of the liver of Oligosarcus jenynsii, one of the most common species inhabiting Los Padres Lake (Buenos Aires Province, Argentina).

Identification of Hypoxia-Regulated Genes in the Liver of Common Sole (Solea solea ) Fed Different Dietary Lipid Contents ArchiMer
Mazurais, David; Ferraresso, Serena; Gatta, Pier Paolo; Desbruyeres, Elisabeth; Severe, Armelle; Corporeau, Charlotte; Claireaux, Guy; Bargelloni, Luca; Zambonino, Jose-luis.
Coastal systems could be affected by hypoxic events brought about by global change. These areas are essential nursery habitats for several fish species including the common sole (Solea solea L.). Tolerance of fish to hypoxia depends on species and also on their physiological condition and nutritional status. Indeed, high dietary lipid content has been recently shown to negatively impact the resistance of sole to a severe hypoxic challenge. In order to study the molecular mechanisms involved in the early response to hypoxic stress, the present work examined the hepatic transcriptome in common sole fed diets with low and high lipid content, exposed to severe hypoxia. The activity of AMP-activated protein kinase (AMPK) was also investigated through the...

Jaffal, Ali; Betoulle, Stephane; Biagianti-risbourg, Sylvie; Terreau, Alexandre; Sanchez, Wilfried; Paris-palacios, Severine.
The Kerguelen Islands include various species of freshwater fish such as brown trout (Salmo trutta). These trout are among the most isolated from direct anthropogenic impact worldwide. This study was designed to analyse cadmium (Cd) and copper (Cu) concentrations in the liver of Kerguelen brown trout, and to assess the possible impacts of these metals on hepatic histopathology and oxidative stress parameters (superoxide dismutase and catalase activity and glutathione levels). Trout were caught in the Chateau River, the Studer Lakes and the Ferme Pond, close to the scientific station of the Kerguelen Islands, corresponding to three morphotypes (river, lake and station). Kerguelen trouts' hepatic concentrations of Cd and Cur were similar to those reported in...

Liaison de l'oestradiol 17 beta dans les hepatocytes de truite Arc-en-ciel et induction de la vitellogenine et de son ARNm specifique ArchiMer
Maitre, J; Mercier, L; Dolo, L; Valotaire, Y.
In the liver of rainbow trout (Salmo gairdnerii ), vitellogenin (Vg) synthesis is influenced by oestradiol (E sub(2)) which is believed to act through the classical mechanism of steroid hormone action. After binding of the hormone to a soluble specific receptor protein, the oestradiol-receptor complex is translocated to the nucleus, where it interacts with DNA and modulates expression of Vg genes, leading to increased synthesis of specific mRNA and Vg. The authors show here: (i) the presence of specific oestrogen receptors in the cytosol of the male trout liver. (ii) The male liver, offering, an ideal experimental control of "zero" background, the authors followed - in the liver of male trouts - the kinetics of induction of Vg mRNA by hybridization with Vg...

Differential gene expression after total replacement of dietary fish meal and fish oil by plant products in rainbow trout (Oncorhynchus mykiss) liver ArchiMer
Panserat, S.; Hortopan, G. A.; Plagnes-juan, E.; Kolditz, C.; Lansard, M.; Skiba-cassy, S.; Esquerre, D.; Geurden, I.; Medale, Francoise; Kaushik, Sadasivam; Corraze, Genevieve.
Our objective was to analyse the hepatic transcriptomes of juvenile rainbow trout fed with a plant-based diet. We focused our analysis on the total replacement of fish meal (FM) and fish oil (FO) by a 100% plant-based diet (0% FM, 0% FO). We analysed the postprandial hepatic transcriptomes of rainbow trout fed the two diets 8 h after feeding. Six total hepatic RNAs from each dietary group were hybridised against a trout cDNA microarray (9K). After treatment of the data respecting the standard MIAME (Minimum Information About a Microarray Experiment) protocol, we found that 176 hepatic genes were differentially expressed between fish fed the two diets: 96 and 80 were over-expressed and under-expressed, respectively, in trout fed the plant-based diet. Etude cytologique ultrastructurale des anomalies hepatiques du loup, de la daurade, et de l'anguille, induites par une alimentation artificielle ArchiMer
Bac, N; Biagianti, S; Brusle, J.
In the three species Dicentrarchus labrax, Sparus auratus , and Anguilla anguilla an artificial feeding (dry pellets) disturbs liver cells as revealed at ultrastructural level. In hepatocytes, anomalies consist of lipids storage (liposomes) considered as deviation in fatty acids metabolism. Moreover are observed cellular alterations (mitochondrial vacuolization, nuclear pycnosis) related to degenerative process. It is of great interest to understand wether liver modifications are just a reaction to an unbalanced feeding or a true trophic pathology.

Variation in Metabolic Rate among Individuals Is Related to Tissue-Specific Differences in Mitochondrial Leak Respiration ArchiMer
Salin, Karine; Auer, Sonya K.; Rudolf, Agata M.; Anderson, Graeme J.; Selman, Colin; Metcalfe, Neil B..
Standard metabolic rate (SMR) and maximum metabolic rate (MMR) typically vary two-or threefold among conspecifics, with both traits assumed to significantly impact fitness. However, the underlying mechanisms that determine such intraspecific variation are not well understood. We examined the influence of mitochondrial properties on intraspecific variation in SMR and MMR and hypothesized that if SMR supports the cost of maintaining the metabolic machinery required for MMR, then the mitochondrial properties underlying these traits should be shared. Mitochondrial respiratory capacity (leak and phosphorylating respiration) and mitochondrial content (cytochrome c oxidase activity) were determined in the liver and white muscle of brown trout Salmo trutta of...

Anatomical Description of the Liver, Hepatic Ligaments and Omenta in the Coypu (Myocastor coypus) International Journal of Morphology
Pérez,W; Lima,M.
The objective of this work is to give a complementary description of the hepatic lobulation, the hepatic ligaments and the omenta of the nutria. Thirty nutrias were studied by gross dissection. The liver of the nutria was divided into six lobules as follows: left lateral, left medial, quadrate, right medial, right lateral, and caudate lobes. The caudate lobe was divided into a papillary and a caudate process. A whole falciform ligament, extending as far as the navel, was found in all animals. This one was the only ligament that contained fat in between its sheets, and it was abundant in the umbilical part. The left triangular ligament had two parts. One of them was attached to the left lateral lobe of the liver and the other one to the left medial lobe....

Effects of Biodegradable Detergents in the Accumulation of Lipofuscin (Age Pigment) in Gill and Liver of Two Neotropical Fish Species International Journal of Morphology
Pereira,Bruno Fiorelini; Alves,Anderson Luis; Senhorini,José Algusto; Rocha,Rita de Cássia Gimenes de Alcântara; Pitol,Dimítrius Leonardo; Caetano,Flávio Henrique.
Currently there is a growing concern, in both population and governments, to identify the effects of substances commonly disposed of into rivers and lakes, on aquatic fauna and flora. Thus the objective of the present study was to verify effects of biodegradable detergents and water from an urban lake on gills and liver of two neotropical fish species of great economic importance, Astyanax altiparanae and Prochilodus lineatus. In order to do so, lipofuscin, also called the ageing pigment, was used as bioindicator. After one and five months of experiment both tissues accumulated this pigment. These data are discussed from physiological points of view, related with lipid peroxidation and mitochondrial damage.

Ethanol Intake and Toxicity: In Search of New Treatments International Journal of Morphology
Sandoval,Cristian; Vásquez,Bélgica; Mandarim-de-Lacerda,Carlos; del Sol,Mariano.
Prolonged alcohol consumption has consequences on the liver, producing necrotic precipitates and fibrosis, on the pancreas, causing the pancreatic acini to atrophy and destroying insulin-producing cells, and on the central nervous system (CNS), causing the gray and white matter in the frontal lobes of the brain and cerebellum to atrophy. Generally, alcohol is metabolized via oxidative pathways, where the enzymes alcohol dehydrogenase and aldehyde dehydrogenase participate during its metabolization in the liver and CNS, or via non-oxidative pathways during its metabolization in the pancreas. Ethanol metabolism can produce oxidative stress and tissue damage mediated by free radicals, causing morphological and functional alterations in the liver. In the...
