O objetivo do trabalho foi padronizar a metodologia de pré-condicionamento das sementes de mamoneira (Ricinus communis L.) para a avaliação do potencial fisiológico pelo teste de tetrazólio. Testaram-se os seguintes métodos de pré-condicionamento: sementes com tegumento entre papel umedecido a 30, 35 e 40°C por 6, 8, 10, 12, 14, 16 e 18h; sementes sem tegumento entre papel umedecido e sementes com tegumento imersas em água a 25, 30, 35 e 40° C por 1, 2, 3, 4, 5 e 6h. Após o pré-condicionamento, removeu-se o tegumento das sementes, que foram cortadas no sentido do comprimento e imersas na solução de tetrazólio a 0,2%, por 120 min., a 35°C. Heart rate variability (HRV) analysis consists in a well-established tool for the assessment of cardiac autonomic control, both in humans and in animal models. Conventional methods for HRV analysis in rats rely on conscious state electrocardiogram (ECG) recording based on prior invasive surgical procedures for electrodes/transmitters implants. The aim of the present study was to test a noninvasive and inexpensive method for ECG recording in conscious rats, assessing its feasibility for HRV analysis. This study aimed to evaluate the effect of penetration rate and the size of the cone base on the resistance to penetration under different soil moistures and soil bulk density. The experimental design was completely randomized in a 4x2x2x2 factorial arrangement, with the factors, soil bulk density of 1.0; 1.2; 1.4 and 1.6 Mg m-3, soil moisture at the evaluation of 0.16 and 0.22 kg kg-1, penetration rates of 0.166 and 30 mm s-1 and areas of the cone base of 10.98 and 129.28 mm² resulting in 32 treatments with 8 replicates. In this work, several major procedures of the electrophoretic mobility shift assay (EMSA) were modified including swift extraction of the nucleic protein, labeling of the probe and radioautography. The modified assay required shorter time, simplified the nucleic protein extraction, increased the radioactivity of the labeling probe, skipped the tedious process of gel drying, and produced clear images. Its results were comparable, reproducible and stable. It thus has merited for wide application. |

The present article is the adapted version of an electronic symposium organized by the Brazilian Society of Neuroscience and Behavior (SBNeC) which took place on June 14, 2000. The text is divided into three sections: I. The main issues, II. Chronodrugs, and III. Methods. The first section is dedicated to the perspectives of chronobiology for the next decade, with opinions about the trends of future research being emitted and discussed. The second section deals mostly with drugs acting or potentially acting on the organism's timing systems. Competing claims on natural resources become increasingly acute, with the poor being most vulnerable to adverse outcomes of such competition. A major challenge for science and policy is to progress from facilitating univocal use to guiding stakeholders in dealing with potentially conflicting uses of natural resources. The development of novel, more equitable, management options that reduce rural poverty is key to achieving sustainable use of natural resources and the resolution of conflicts over them. Complex systems are shaped by cross-scale interactions, nonlinear feedbacks, and uncertainty, among other factors. Transdisciplinary approaches that combine participatory and conventional methods and democratize knowledge to enable diverse inputs, including those from local, informal experts, are essential tools in understanding such systems. The challenge confronting those seeking to understand the institutional dimensions of global environmental change and patterns of land-use and land-cover change is to find effective methods for analyzing the dynamics of socio-ecological systems. Such systems exhibit a number of characteristics that pose problems for the most commonly used statistical techniques and may require additional and innovative analytic tools. Scholars of comanagement are faced with a difficult methodological challenge. As comanagement has evolved and diversified it has increasingly merged with the field of adaptive management and related concepts that derive from resilience thinking and complex adaptive systems theory.
