The enzyme glucanase from Moniliophthora perniciosa was produced in liquid medium and purified from the culture supernatant. A multivariate statistical approach (Response Surface Methodology - RSM) was employed to evaluate the effect of variables, including inducer (yeast extract) and fermentation time, on secreted glucanase activities M. perniciosa detected in the culture medium. The crude enzyme present in the supernatant was purified in two steps: precipitation with ammonium sulfate (70%) and gel filtration chromatography on Sephacryl S-200. The best inducer and fermentation time for glucanase activities were 5.9 g L-1 and 13 days, respectively. The enzyme chitinase from Moniliophthora perniciosa the causative agent of the witches' broom disease in Theobroma cacao, was partially purified with ammonium sulfate and filtration by Sephacryl S-200 using sodium phosphate as an extraction buffer. Response surface methodology (RSM) was used to determine the optimum pH and temperature conditions. Four different isoenzymes were obtained: ChitMp I, ChitMp II, ChitMp III and ChitMp IV. This report describes the cloning, sequence and expression analysis of the glyceraldehyde-3-phosphate dehydrogenase (GAPDH) gene of Moniliophthora perniciosa, the most important pathogen of cocoa in Brazil. Southern blot analysis revealed the presence of a single copy of the GAPDH gene in the M. perniciosa genome (MpGAPDH). The complete MpGAPDH coding sequence contained 1,461 bp with eight introns that were conserved in the GAPDH genes of other basidiomycete species. The cis-elements in the promoter region of the MpGAPDH gene were similar to those of other basidiomycetes. A vassoura de bruxa, doença endêmica da região amazônica, é considerada a mais prejudicial à cultura do cupuaçuzeiro. Em mudas atacadas, o sintoma mais frequente é a vassoura terminal (VT), pois o fungo infecta principalmente a gema apical. A medida mais segura e de baixo custo para o manejo da doença é o uso de cultivares resistentes a ela. Porém o ciclo no melhoramento genético da cultura é longo. Assim, o uso de metodologia que permita fazer comparações na fase de mudas é de grande valia para acelerar o processo de seleção e desenvolvimento de cultivares de cupuaçuzeiro resistentes à vassoura de bruxa, pois genótipos suscetíveis podem ser descartados precocemente. The objective of this work was to identify genotypes with high general combining ability for resistance to witches'-broom (Moniliophthora perniciosa) in populations formed from a first cycle of recurrent selection. Highly productive and resistant clones from different origins were interbred using the North Carolina II design. The clones SCA 6, CSUL 7, RB 39, CEPEC 89, OC 67, BE 4, EEG 29 and ICS 98 were used as paternal parents, while the maternal ones were NA 33, CCN 10, IMC 67, P 4B, CCN 51, CEPEC 86, SGU 54 and ICS 9. Os objetivos deste trabalho foram caracterizar a resistência à vassoura-de-bruxa de plantas de cacau originadas do cruzamento entre TSH 1188 e CCN 51 (população segregante), por meio de dois métodos de inoculação em condições de campo, e identificar marcadores microssatélites específicos para grupos de plantas resistentes e suscetíveis. As plantas-controle avaliadas pelos métodos de inoculação natural e inoculação artificial em campo produziram os mesmos padrões de sintomas. The efficacy of Ocimum selloi essential oil was evaluated for controlling the growth of mycelia and spores germination Moniliophthora perniciosa. Six compounds (99.89%) of the total oil were identified by GC-MS, of which methyl chavicol, methyl eugenol, β-caryophyllene, germacrene-D, bicyclogermacrene and spathulenol. Essential oil was tested for antifungal activity, which was determined by disc diffusion and minimum inhibitory concentration (MIC) determination methods. Application of the oil reduced mycelial growth in a dose dependent manner, with maximum inhibition being observed at concentration of 1,000 ppm.
