In vitro morphogenesis and cell suspension culture establishment in Piper solmsianum C. DC. (Piperaceae) Acta Botanica

Piper solmsianum is a shrub from Southeast Brazil in which many biologically active compounds were identified. The aim of this work was to establish a cell suspension culture system for this species. With this in mind, petiole and leaf explants obtained from in vitro plantlets were cultured in the presence of different plant growth regulator combinations (IAA, NAA, 2,4-D and BA). Root and indirect shoot adventitious formation, detected by histological analysis, was observed. Morphogenetic, structural variables and leaf biomass flows of Italian ryegrass (Lolium multiflorum Lam.) were evaluated under two stocking rates: ‘Low’ and ‘High’. These rates were determined by heifers exclusively on pasture or on pasture and supplemented with corn grain. The experimental design was completely randomized following a repeated measure arrangement, two stocking rates, two and four replications of area for the stocking rates ‘low’ and ‘high’, respectively. Biomass flows and defoliation patterns of alexandergrass pasture grazed by beef heifers, receiving or not protein salt Animal Sciences

The experiment aimed at evaluating leaf biomass flow, intensity and frequency of defoliation of Alexandergrass (Urochloa plantaginea (Link) Hitch) by beef heifers exclusively on pasture or that grazed and received protein salt. A completely randomized experimental design was used with repeated measures over time, two treatments and two area replications, with three Angus test heifers by paddock. Leaf blade intake flow is 18% superior for heifers exclusively on pasture. Grazing intensity (59.8%) is similar when heifers receive protein salt or not. Heifers receiving protein salt reduce by one day the defoliation frequency of tillers. This study aimed to evaluate the effects of temperature variation during incubation on testicular morphology of post-hatch chicks. We utilized 60 eggs incubated under different temperatures: Group 1 – 37.5°C; Group 2 – 39.5°C; and Group 3 – 34°C. Chicks were weighted and the testes were histologically analyzed. All eggs from Group 3 showed embryonic death. There were no significant differences in body weight and testicular morphology between Groups 1 and 2; however, there was a difference in the diameter of the seminiferous tubules (p < 0.001). The increase in temperature to 39.5°C during incubation causes a decrease in diameter of the seminiferous tubules.

This study aims to evaluate the morphogenetic characteristics of three cultivars of Brachiaria brizantha subjected to nitrogen fertilization. The design was a randomized block in factorial arrangement 4x3; three cultivars of B. brizantha - Marandu, Piatã, Xaraés and four nitrogen levels - 0, 80, 160 and 240 kg/ha, with three replications. The experimental units consisted of plastic pots filled with 5 dm3 of soil. Histological analysis of the callogenesis and organogenesis from root segments of Curcuma zedoaria Roscoe BABT

Callus was induced from root segments taken from in vitro grown plants of Curcuma zedoaria Roscoe. The explants were cultured on agar-solidified Murashige and Skoog medium supplemented with 13.4muM of alpha-naphthaleneacetic acid and 2.2muM of 6-benzylaminopurine at 25ºC in the dark. Histological analysis revealed that callus was formed from the hypertrophied cortical parenchyma cells of the explant. Some of these cells underwent division while the surrounding cells accumulated starch. Callus was capable of shoot bud regeneration after 70 days when it was transfered to liquid medium of the same composition. After 30 days in liquid medium, buds developed from nodular structures. Selective agent and A. tumefaciens overgrowth-control antibiotics in Eucalyptus camaldulensis Cotiledonary culture BABT

The objectives of the present work were to establish the minimal lethal dose of the selective agent to determine the type and concentration of appropriate antibiotics for the elimination of Agrobacterium tumefaciens inoculated explants, without interfering with the regenerative potential of the E. camaldulensis cotyledonary explants. Non-transformed explants were cultivated in medium supplemented with kanamycin. The results showed that the antibiotic was suitable for the selection of transformed cells in the concentration of 9 mg L-1 as it inhibited the growth of non-transformed cells. ABSTRACT The present study reports a shoot organogenesis-based system for in vitro regeneration of Passiflora miniata, an Amazonia passion fruit species. Root segments were cultured in Murashige and Skoog (MS) medium supplemented with different concentrations (range 2-9 µM) of 6-benzyladenine (BA); thidiazuron (TDZ) or kinetin (KIN). Plant growth regulators were not added to the control treatment. Root explants have showed a high regenerative potential. After 30 days of in vitro culture, the root explants showed several shoots formed direct and indirectly. Tissue cultures of Alternanthera brasiliana (L.) Kuntze were treated with different growth regulators (Kinetin and 2,4-D), tyrosine and Ultraviolet A radiation (UV-A; 320 - 400 nm) to investigate their effects on development and pigment production. Nodal segments of plantlets grown from seeds were inoculated in all tested media and different light conditions. After eight weeks, this material was used to evaluate biomass, chlorophyll and betacyanin production. The Murashige and Skoog (MS) + kinetin medium resulted in development of approximately four shoots/explant. Effect of alcohols on filamentation, growth, viability and biofilm development in Candida albicans BJM

In this study we report the potential of alcohols as morphogenetic regulators in Candida albicans. All the alcohols tested influenced various modes of growth like planktonic as well as biofilm forms. Viability was affected at high concentrations. Among the alcohols, the response of C. albicans to amyl alcohol (pentanol) was noteworthy. Amyl alcohol at a concentration 0.5% which was not inhibitory to growth and viability specifically inhibited morphogenetic switching from yeast to hyphal forms. It also inhibited normal biofilm development favoring yeast dominated biofilms. A method for regeneration of the commercially important common bean (Phaseolus vulgaris ) using N6-benzylaminopurine(BAP) and adenine sulphate (AS) was established. Embryogenic axes of the Costa Rican common bean cultivars Bribrí, Brunca, Guaymí, Huetar and Telire were cultured on Murashige and Skoog medium supplemented with 100 mgl-1 myo-inositol, 1 mgl-1 thiamine, 30 gl-1 sucrose, BAP (0, 5 and 10 mgl-1), AS (0, 20 and 40 mgl-1) and 8 gl-1 agar. Their assembling gives rise to a five-layered continuous structure of about 37 nm thick. Afterwards, the inner epicuticle is added inside close to the cuticulin layer but dense plaques are not concerned with the phenomenon. The deposition of the fibrous surface coat outside to the cuticulin layer takes place only during late postmolt stages (D2-D3). In mineralized cuticles, this typical epicuticle structure persists during all intermolt long.
