Comparação entre métodos de tuberculinização no diagnóstico da infecção por agentes do complexo Mycobacterium avium ou M. bovis em suínos Arq. Bras. Med. Vet. Zootec.
Morés,N.; Amaral,A.L.; Ventura,L.; Silva,R.A.M.; Silva,V.S.; Barioni Junior,W..

BCGiosis as a presenting feature of a child with chronic granulomatous disease BJID
Movahedi,Zahra; Norouzi,Sayna; Mamishi,Setareh; Rezaei,Nima.
Bacillus Calmette Guerin (BCG) vaccine, which is administered to all newborns in some regions, could lead to serious complication ranging from local disease (known as BCGitis) to disseminated disease (BCGosis) in a group of patients with primary immunodeficiency diseases. We are reporting here a 3.5 year-old girl with a history of prolonged BCGitis, which developed to disseminated disease without any other special features. Immunological studies with nitro-blue tetrazolium test confirmed the diagnosis of chronic granulomatous disease in this patient. Chronic granulomatous disease should be considered in the list of differential diagnosis in all children with BCGosis, even in the absence of any other manifestations related to immunodeficiency.

Identification of Mycobacterium bovis antigens by analysis of bovine T-cell responses after infection with a virulent strain BJMBR
Alito,A.; McNair,J.; Girvin,R.M.; Zumarraga,M.; Bigi,F.; Pollock,J.M.; Cataldi,A..
Purification and characterization of individual antigenic proteins are essential for the understanding of the pathogenic mechanisms of mycobacteria and the immune response against them. In the present study, we used anion-exchange chromatography to fractionate cell extracts and culture supernatant proteins from Mycobacterium bovis to identify T-cell-stimulating antigens. These fractions were incubated with peripheral blood mononuclear cells (PBMC) from M. bovis-infected cattle in lymphoproliferation assays. This procedure does not denature proteins and permits the testing of mixtures of potential antigens that could be later identified. We characterized protein fractions with high stimulation indices from both culture supernatants and cell extracts....

A single strain of Mycobacterium bovis bacillus Calmette-Guérin (BCG) grown in two different media evokes distinct humoral immune responses in mice BJMBR
Petricevich,V.L.; Ueda,C.; Alves,R.C.B.; da Silva,M.A.; Moreno,C.; Melo,A.R.; Dias da Silva,W..
Two attenuated bacillus Calmette-Guérin (BCG) preparations derived from the same Moreau strain, Copenhagen but grown in Sauton medium containing starch and bacto-peptone (onco BCG, O-BCG), or asparagine (intradermal BCG, ID-BCG), exhibited indistinguishable DNA sequences and bacterial morphology. The number of viable bacilli recovered from spleen, liver and lungs was approximately the same in mice inoculated with the vaccines and was similarly reduced (over 90%) in mice previously immunized with either BCG vaccine. The humoral immune response evoked by the vaccines was, however, distinct. Spleen cell proliferation accompanying the growth of bacilli in tissue was significantly higher in mice inoculated with O-BCG. These cells proliferated in vitro upon...

Effects of diazepam on Mycobacterium bovis-induced infection in hamsters BJMBR
Righi,D.A.; Pinheiro,S.R.; Guerra,J.L.; Palermo-Neto,J..
The in utero exposure of hamsters to low doses of diazepam results in impaired host defense against Mycobacterium bovis during adulthood. Delayed developmental immunotoxicity, however, represents a specific situation that might not be general. The present experiment was undertaken to investigate the effects of diazepam on hamster resistance to M. bovis using adult animals. The effects of diazepam treatment on serum cortisol levels were also studied. Adult hamsters (N = 10 for each group) were treated with diazepam (E1 = 1.0, E2 = 2.0 or E3 = 3.0 mg kg-1 day-1 subcutaneously) or with control solution (C) for 30 days. Seven days after the beginning of the treatment, the animals received identical inoculum concentrations of M. bovis. Hamsters treated with the...

Comparison of three decontamination methods for Mycobacterium bovis isolation BJM
Ambrosio,Simone Rodrigues; Oliveira,Eugenia Márcia de Deus; Rodriguez,Cesar Alejandro Rosales; Ferreira Neto,José Soares; Amaku,Marcos.
Sixty samples of tissue fragments with lesions suggestive of tuberculosis from bovine abattoirs, kept in saturated solution of sodium borate, were subjected to four treatments: 4% NaOH (Petroff Method), 12 % H2SO4 and 1.5% HPC (1-Hexadecylpyridinium Chloride) decontamination, and physiological saline solution (control). The HPC method showed the lowest contamination rate (3%) when compared to control (88%, p<0.001), NaOH (33%, p<0.001) and H2SO4 (21.7%, p<0.002). Regarding the isolation success, the HPC method was better (40%) than the control (3%, p<0.001), NaOH (13%, p=0.001) and H2SO4 (1.7%, p<0.001) methods. These results indicate that HPC is an alternative to the Petroff method.

Interference of tuberculosis on the performance of ELISAs used in the diagnosis of paratuberculosis in cattle BJM
Lilenbaum,Walter; Ferreira,Rachel; Marassi,Carla Dray; Ristow,Paula; Oelemann,Walter Martins Roland; Fonseca,Leila de Souza.
Forty-four cows from five herds infected with tuberculosis (TB) and without paratuberculosis (PTB), and 21 cows from a herd without either infection were studied. The cattle presented concordant results in both the skin test and gamma-interferon assay for TB and two commercial ELISAs for PTB. Animals were divided according to TB test results into Group A with 28 TB-infected animals, Group B with 16 TB-negative animals from infected herds, and Group C with 21 TB-negative cows from a tuberculosis-free herd (which were used as controls). Twenty of 28 animals from Group A (71.4%), 6/16 from Group B (37.5%) and none from Group C were reactive to PTB ELISAs, suggesting that these commercial kits were unable to distinguish between PTB and TB. It is proposed that...

Potential application of new diagnostic methods for controlling bovine Tuberculosis in Brazil BJM
Medeiros,Luciana dos Santos; Marassi,Carla Dray; Figueiredo,Eduardo Eustáquio S.; Lilenbaum,Walter.
Bovine tuberculosis, a chronic infection in cattle caused by Mycobacterium bovis, remains an economic and public health problem for several countries. Due to its economic impact on international trade, contagious nature, and implications for human health, global programs to eradicate the disease were implemented worldwide. Those programs are based on slaughtering PPD-reactive animals. Despite the National Programs in Brazil, complete eradication has not been achieved, and the disease remains, albeit at a lower prevalence. The central purpose of this review is to address diagnostic tests for tuberculosis. Considering the course of the infection in cattle, at least two tests, ideally complementary to one another, may be necessary for an adequate diagnosis:...

Thin layer microcolony culture associated with PCR for early identification of Mycobacterium bovis BJM
Rosário,Tatiana Reis do; Dib,Cristina Corsi; Roxo,Eliana; Pinheiro,Sônia Regina; Vasconcellos,Silvio Arruda; Benites,Nilson Roberti.
The initial growth of mycobacteria from 49 samples of cattle and buffalo organs collected in commercial slaughterhouses was compared between modified Middlebrook 7H11 thin layer microcolony culture and Stonebrink medium used in the isolation of Mycobacterium bovis. Aliquots were decontaminated by Petroff's method, processed and cultured in both media. The identity of the acid-fast bacilli stained by Ziehl-Neelsen was confirmed by PCR. Optical microscopy showed that results of the early observation of Mycobacterium bovis colonies in thin layer culture were similar to those obtained in macroscopic observation of the colonies in Stonebrink medium. However, early observation of the colonies enabled early confirmation by PCR, given the shorter time to the...

Infection by Mycobacterium bovis in a dog from Brazil BJM
Rocha,Vivianne Cambuí Figueiredo; Figueiredo,Salomão Cambuí de; Rosales,Cesar Alejandro Rodriguez; Porto,Camila Dias; Sequeira,Julio Lopes; Ferreira Neto,José Soares; Paes,Antônio Carlos; Salgado,Vanessa Riesz.
Abstract Tuberculosis (TB) is a chronic disease caused by bacteria belonging to the Mycobacterium tuberculosis complex (MtbC). This disease rarely affects dogs. Canine infections are usually caused by M. tuberculosis. Mycobacterium bovis infections are rare in dogs and associated with consumption of raw milk or contaminated products. Here, we report a Boxer dog who had a M. bovis infection and was admitted to a Brazilian veterinary hospital with a presumptive diagnosis of chronic ehrlichiosis. Despite receiving treatment for chronic ehrlichiosis, it progressed to death. TB was diagnosed during post-mortem examinations using histopathological analysis. Ziehl-Neelsen staining revealed acid-fast bacilli in the kidneys, liver, mesentery, and a mass adhered to...

Tuberculosis determined by Mycobacterium bovis in captive waterbucks (Kobus ellipsiprymnus) in São Paulo, Brazil BJM
Rocha,V.C.M.; Corrêa,S.H.R.; Oliveira,E.M.D.; Rodriguez,C.A.R.; Fedullo,J.D.; Matrone,M.; Setzer,A.; Ikuta,C.Y.; Vejarano,M.P.; Figueiredo,S.M.; Ferreira Neto,J.S..
Two waterbucks from São Paulo Zoo Foundation exhibited respiratory symptoms in July 2004. After euthanasia, granulommas in lungs and mediastinic lymph nodes were observed. Acid-fast bacilli isolated were identified as Mycobacterium bovis spoligotype SB0121 by PRA and spoligotyping. They were born and kept in the same enclosure with the same group, without any contact to other species housed in the zoo. This is the first detailed description of M. bovis infection in Kobus ellipsiprymnus.

Detection of Mycobacterium bovis DNA in nasal swabs from tuberculous cattle by a multiplex PCR BJM
Figueiredo,Eduardo Eustáquio de Souza; Carvalho,Ricardo Cezar Tavares; Silvestre,Flávia Galindo; Lilenbaum,Walter; Fonseca,Leila Sousa; Silva,Joab Trajano; Paschoalin,Vânia Margaret Flosi.
Detection of tuberculosis in cattle relies on the intradermal tuberculin test (ITT), but a definitive diagnosis requires identification of the pathogen after the animal is slaughtered. DNA in nasal swabs from 50 cows was analyzed by m-PCR, targeting for the RvD1-Rv2031c and IS6110 sequences. M. bovis was identified in two of 34 tuberculous cows (5.9%). The use of mPCR of nasal swabs as an in vivo diagnostic tool for bovine tuberculosis is suggested.

Molecular typing of Mycobacterium bovis isolated in the south of Brazil BJM
Ramos,Daniela Fernandes; Silva,Ana Bárbara Scholante; Fagundes,Michel Quevedo; von Groll,Andrea; Silva,Pedro Eduardo Almeida da; Dellagostin,Odir Antônio.
Bovine tuberculosis is a major infectious disease of the cattle. In this study, 85 M. bovis isolates from 162 lymph nodes, obtained from a herd of cattle on a farm in southern Brazil, were evaluated using spoligotyping and VNTR. The strains were grouped into five clusters and five orphans, showing a heterogenic genetic profile, what could represent diverse geographic origins of the introduced cows and/or the frequent movement of cattle between different properties.

Influence of initial L-asparagine and glycerol concentrations on the batch growth kinetics of Mycobacterium bovis BCG BJM
Leal,Maria Betania Batista; Baruque-Ramos,Júlia; Hiss,Haroldo; Paz,Marcelo Fossa da; Sakai,Maria Cristina; Vassoler,Umbelina Macedo; Arauz,Luciana Juncioni de; Raw,Isaías.
The influences of the L-asparagine and glycerol initial concentrations in Sauton medium on the productivities of biomass and colony forming units were studied. The submerged batch cultivations of Mycobacterium bovis were carried out in a 20 L bioreactor. The L-asparagine and glycerol initial concentrations of 4.54 g/L and 25 mL/L, respectively, corresponded to the best biomass productivity, namely 2.5 g/L.day. On the other hand, the concentrations of 2.27 g/L and 25 mL/L, respectively, led to the highest productivity in terms of colony forming units, namely 2.7·10(6) colonies/mg.day. In addition, by means of the relative consumption analysis of L-asparagine and glycerol (50 and 26% respectively), it was concluded that the concentrations of such components...

Abstract For the definitive diagnosis of bovine tuberculosis, isolation of the etiologic agent is required. However, there is no consensus on the best methodology for isolation of Mycobacterium bovis in Brazil. This study evaluated the most used decontaminants and culture media in the country, in order to identify the best combination for the Brazilian samples. Three decontaminants - 2% sodium hydroxide (w/v), 0.75% hexadecylpiridinium chloride (w/v) and 5% sulphuric acid (v/v) and four culture media - 7H11 Middlebrook with additives and OADC supplement “A” (7H11 A), the same media with another supplement trademark (7H11 B), tuberculosis blood agar (B83) and Stonebrink's medium were compared. Regarding the isolation, there were no significant differences...

Molecular typing of Mycobacterium bovis isolates: a review BJM
Ramos,Daniela Fernandes; Tavares,Lucas; Silva,Pedro Eduardo Almeida da; Dellagostin,Odir Antônio.
Mycobacterium bovis is the main causative agent of animal tuberculosis (TB) and it may cause TB in humans. Molecular typing of M. bovis isolates provides precise epidemiological data on issues of inter- or intra-herd transmission and wildlife reservoirs. Techniques used for typing M. bovis have evolved over the last 2 decades, and PCR-based methods such as spoligotyping and mycobacterial interspersed repetitive unit-variable number tandem repeat (MIRU-VNTR) have been extensively used. These techniques can provide epidemiological information about isolates of M. Bovis that may help control bovine TB by indicating possible links between diseased animals, detecting and sampling outbreaks, and even demonstrating cases of laboratory cross-contamination between...

Isolation and identification of Mycobacterium bovis in milk from cows in northeastern Brazil Ciência Rural
Ramos,Joelson Marcolino; Heinemann,Marcos Bryan; Ferreira Neto,José Soares; Filho,Antônio Francisco de Souza; Cárdenas,Nicolás Céspedes; Alves,Clebert José; Azevedo,Sérgio Santos de.
ABSTRACT: Milk samples from 16 cows that tested positive on the tuberculin test in the state of Paraíba, northeastern Brazil, were used for mycobacteria isolation and identification. Mycobacteria were isolated from five (31.25%) of the 16 milk samples; three samples were classified as M. bovis, and two as belonging to the Mycobacterium genus. This is probably the first study of isolation and identification of M. bovis in milk from cows in Northeastern Brazil, which suggests that humans are at risk of contamination by ingestion.
