Accumulation and depuration of hydrocarbons in the mangrove oyster Crassostrea rhizophorae ArchiMer

Adult specimens of Crassostrea rhizophorae were exposed to diesel oil dispersed in seawater. The experiment consisted of a 10 days contamination period and 15 days depuration period. Oyster samples were extracted, and analysed by gas chromatography and UV fluorescence spectroscopy. The concentrations of n-alkanes, branched alkanes, unresolved complex mixture (UCM) and aromatic hydrocarbons were determined. The oysters accumulated the hydrocarbons rapidly, reaching a maximum concentration of 387.5 ug/g (wet weight). After exposure to uncontaminated seawater, the oysters were able to reduce their hydrocarbon load to 5.6 ug/g. Genetic basis of summer mortality in juvenile cupped oysters ArchiMer

The French project "Morest", coordinated by IFREMER, aims to understand the causes of the summer mortalities in Crassostrea gigas. In 2001, three sets of families were bred following a nested half-sib mating design. 17 half-sib families (HSF) were obtained in this first generation (G1) and reared in 3 sites. Significant differences in survival were observed among HSF, and some HSF showed high levels of mortality in all sites, clearly indicating a genetic basis for survival. In 2002, a second generation (G2), including divergent selection and inbred lines, was constituted. Monitoring of survival and growth of G2s were the same as in 2001. An experimental hatchery for bivalves and the different techniques used in Arcachon are described: sea water treatment, phytoplankton culture, conditionning of oysters for spawning and larval culture. This experimental unit has been organised to study the reasons why natural reproduction is disturbed in the bay. The quality of the oysters and the sea water was analysed to ascertain their capacity to acquire normal oyster development. Characterization of Vibrio isolated from Crassostrea gigas spat suffering summer mortality outbreaks (slides) ArchiMer

The pathogens related to summer mortality outbreaks are a herpes virus and two bacterial strains one belonging to Vibrio splendidus biovar II and the other to Vibrio splendidus spp. However, the feature pathogen/opportunist of these strains is still unknown. Several strains belonging to the genus Vibrio have been identified as pathogen for different mollusk species. In the context of the French program Morest, experiments of cohabitation have been used to demonstrate the potential presence of a transmissible infectious agent in batches of oysters suffering from summer mortality outbreaks. Of the several bacterial diseases which may result from consumption of shellfish, those caused by marine bacteria of the genus Vibrio are the most abundant. In the United States, up to 10,000 non-fatal cases per year are estimated to be caused by the various members of this genus. More than 95% of all deaths in the United States which result from seafood consumption are caused by a single bacterium, Vibrio vulnificus. The bacterium is a normal inhabitant of estuarine waters, and occurs naturally in especially high numbers in molluscan shellfish. Infections following consuption of raw or undercooked shellfish, especially oysters, result in fatality rates of over 60%. Vibrio gigantis sp nov., isolated from the haemolymph of cultured oysters (Crassostrea gigas) ArchiMer

Polyphasic analysis of four new Vibrio isolates originating from the haemolymph of diseased cultured oysters is described. The new isolates were closely related to Vibrio splendidus, having 98 % 16S rRNA gene sequence similarity. Phylogenetic analysis based on DNA gyrase subunit B (gyrB), RNA polymerase Quo factor (rpoD), replication origin-binding protein (rctB) and transmembrane regulatory protein (toxR) genes, fluorescent amplified fragment length polymorphism and DNA-DNA hybridization experiments clearly showed that the new isolates form a tight genomic group that is different from the currently known Vibrio species. Microalgal production in oyster ponds: Nutrition and dissolved organic nitrogen ArchiMer

Microalgal population of oyster ponds is dominated by several tychobenthic diatoms which provide a very good food to grow oysters. The mechanisms which allow them to dominate have been put under research. H.P.L.C. analysis demonstrated 17 amino acids amounted 10-17% of total DON, i.e. 0.6-11.5 mu g-at/l nitrogen. Serin and glycin were the most abundant. None of the substances was related to the presence of oyster. Glutamic acid, alanin and ornithin concentrations showed the greatest variations; they are suspected to play the major role as nutrient source. The boats cruising in the oyster growing areas are protected against biofouling by paints which release toxic compounds such as copper oxide and organotins into the water. After an estimation of the inputs of copper and organotins in the "Marennes-Oleron", Arcachon (Atlantic) and Thau (Mediterranean Sea) areas, their influence on larval fixation is discussed, taking into account the tributyltin toxicity against D larvae of Crassostrea gigas. Experiments in the hatchery rearing of C. gigas larvae showed that: the antifouling paints represent a real danger for spatting areas; the larvae of Arcachon breeding stocks grown in the seawater taken from the Bay develop normally in the laboratory; and, the growth of the larvae, collected from the Bay of Arcachon and reared in hatchery at 18 degree C, confirms that the last summer temperature regimes could not explain the abnormalities. It also confirms the good quality of the seawater of the bay for C. gigas reproduction.

Persistence of atrazine impact on aneuploidy in the Pacific oyster, Crassostrea gigas ArchiMer

Aneuploidy is the alteration of the normal diploid chromosome number. In the Pacific oyster, Crassostrea gigas, hypodiploid aneuploid cells have regularly been reported as has a negative correlation between this phenomenon and growth and evidence for a genetic basis. We previously demonstrated a positive relationship between a pollutant, atrazine, and aneuploidy in Crassostrea gigas adults and juveniles. To evaluate the persistence of this impact, the present study focused on a sample of the same juveniles exposed to different atrazine treatments (0.01 mg/l which represents a peak value found in a polluted environment and 0.1 mg/l) for three and a half months and evaluated them for aneuploidy after another two and a half months in non polluted conditions....

Observations complementaires sur les causes possibles des anomalies de la reproduction de Crassostrea gigas (Thunberg) dans le bassin d'Arcachon. ArchiMer

The spat production in the Bay of Arcachon has been recently disturbed. Bioassays using algae used as food for oyster larvae have shown that the organotin paints affect or inhibit growth of Isochrysis galbana and Chaetoceros calcitrans . Fresh waters coming into the bay and shore sediment did not affect the growth of these algae. Bioassays proposed by Woelke (1967) using embryos and larvae of Crassostrea gigas have shown that the quality of the sea water from Arcachon harbour has been improved a few months after the use of organotin paints has been prohibited. Likewise fresh waters are free from any micropollutants that could drastically disturb the reproduction of C. gigas in the bay.

Composition of biofouling communities on suspended oyster cultures: an in situ study of their interactions with the water column ArchiMer

This study was based on in situ experiments conducted in a shellfish lagoon (Thau, France) to determine the interactions between suspended oyster Crassostrea gigas Thunberg cultures and their environment at a seasonal scale. Three groups of descriptors were considered simultaneously during the course of the study: the taxonomic composition of an oyster culture unit (OCU), the nutrient and oxygen exchanges at the OCU-water interface (fluxes) and the water-column characteristics (water). The impact of suspended oyster culture (Crassostrea gigas, Thunberg) on oxygen and nutrient fluxes has been studied in situ, in a coastal lagoon (Thau, France), during a seasonal cycle. On the first plan of the multiple factorial correspondences analysis (MCA), seasons were well discriminated. The fluxes were maximum in summer and minimum in winter. However, this seasonal pattern was not only linked to the water temperature, as autumn and spring (similar temperatures of about 12 degrees C) were distinct in the second factorial plan (2.3).
