Genome analysis of thirteen Colombian clostridial strains by pulsed field gel electrophoresis Electron. J. Biotechnol.
Quilaguy Ayure,Diana Milena; Suárez Moreno,Zulma Rocío; Aristizábal Gutierrez,Fabio Ancizar; Bernal Morales,Jose Mauricio; Montoya Castaño,Dolly.
Pulsed field gel electrophoresis was used for estimating the size of the genome and evaluating the presence of megaplasmids in 13 native Colombian solventogenic Clostridium strains. DNA preparation and purification were optimised for obtaining differentiated restriction fragments in electrophoresis. Genomic DNA was digested with ApaI, Eco52I, SmaI and XhoI enzymes. Estimated genome size for native strains ranged from 4.0 to 4.2 mega base pairs. Larger sized plasmids were detected and the presence of genes related to megaplasmid pSOL1 was determined by polymerase chain reaction. adc gene region amplification suggested that genes related to solventogenesis in native strains may be located in an extra-chromosomal element. Determining genome size provides...

Staphylococcus aureus infections are growing problems worldwide with important implications in hospitals. The organism is normally present in the nasal vestibule of about 35% of apparently healthy individuals and its carriage varies between different ethnic and age groups. Staphylococcal nasal carriage among health workers is particularly important to establish new clones and track origin of infections during outbreak situations. To determine the carriage rate and compare the pulsed field gel patterns of the strains, nasal swabs were collected from 185 medical students in a teaching hospital in Lagos, Nigeria. Isolates of S. aureus were tested for heamolysin production, methicillin sensitivity and Pulsed Field Gel Electrophoresis (PFGE) was performed. The...

Carbapenem-resistant Pseudomonas aeruginosa outbreak in an intensive care unit of a teaching hospital BJID
Gales,Ana C.; Torres,Patrícia L.; Vilarinho,Dominique S. O.; Melo,Renata S.; Silva,Claudinéia F. L.; Cereda,Rosângela F..
The genetic similarity of carbapenem-resistant Pseudomonas aeruginosa strains isolated in the Hospital Universitário São Francisco, Bragança Paulista, São Paulo, Brasil, was evaluated by pulsed field gel electrophoresis (PFGE). A unique clone was detected among 5 of 7 isolates, suggesting that cross-contamination might have played a role in the spread of carbapenem-resistant P. aeruginosa strains. Interestingly, a similar PFGE pattern was encountered in a P. aeruginosa strain isolated from Hospital São Paulo that was used as a PFGE control.

Mucoid Burkholderia cepacia morphotype emerged within a nine year follow-up of a cystic fibrosis patient. Clinical data suggested a linkage between the mucoid phenotype isolation and the deterioration of the patient's condition. Despite of the phenotypic variation, molecular typing showed that the patient was chronically infected with B. cepacia complex isolates belonging to a same genetic clone.

Epidemiological Survey of Listeria monocytogenes in a gravlax salmon processing line BJM
Cruz,C.D.; Silvestre,F.A.; Kinoshita,E.M.; Landgraf,M.; Franco,B.D.G.M.; Destro,M.T..
Listeria monocytogenes is a cause of concern to food industries, mainly for those producing ready-to-eat (RTE) products. This microorganism can survive processing steps such as curing and cold smoking and is capable of growing under refrigeration temperatures. Its presence in RTE fish products with extended shelf life may be a risk to the susceptible population. One example of such a product is gravlax salmon; a refrigerated fish product not exposed to listericidal processes and was the subject of this study. In order to evaluate the incidence and dissemination of L. monocytogenes 415 samples were collected at different steps of a gravlax salmon processing line in São Paulo state, Brazil. L. monocytogenes was confirmed in salmon samples (41%), food contact...
