Structural organization of polygalacturonase-encoding genes from Penicillium griseoroseum Genet. Mol. Biol.

The pectinolytic system of Penicillium griseoroseum has been studied as a model to investigate aspects of gene organization in filamentous fungi. Here we show that the endopolygalacturonase-coding genes previously isolated exist as single copies in the fungus genome. DNA blot analysis revealed the presence of corresponding genes in other Penicillium species, although only one or two genes were found in opposition to the endoPG gene family reported for other filamentous fungi. Isotermas de adsorção do pedúnculo seco do caju AGRIAMBI

A atividade de água constitui um fator importante no processo de fermentação semi-sólida, haja vista sua relação com a quantidade de água disponível ao microrganismo responsável pelo metabolismo do produto, sendo necessário à obtenção de isotermas de sorção para caracterização do substrato. Ante o exposto, objetivou-se a construção das isotermas de adsorção do pedúnculo seco do caju (Anacardium occidentale L.) nas temperaturas usuais de fermentação (25, 30, 35 e 40 ºC). Ajustaram-se as isotermas com os modelos de BET, GAB, Oswin, Henderson e Smith. ABSTRACT: Biological degumming is an eco-friendly, efficient, high-quality and low-cost method that has become the leading bast fiber degumming technology. However, bacterial strains with short degumming cycles, high gum removal rates and small fiber damage are few. To screen high quality microbial resources with bast-fiber biological degumming function, soil samples were collected from a continuously cultivated banana plantation and then used to be enriched by ramie and kenaf materials in turn. Screening and Molecular Identification of New Microbial Strains for Production of Enzymes of Biotechnological Interest BABT

ABSTRACT: This research focused on isolation, identification and characterization of new strains of fungi and bacteria, which were able to produce extracellular xylanase, mannanase, pectinase and α-amylase. Fungi isolates were identified on the basis of analyses of 18S gene sequencing and internal transcribed spacer region. The closest phylogenetic neighbors according to 18S gene sequence and ITS region data for the two isolates M1 and SE were Aspergillus fumigatus and Aspergillus sydowii, respectively. I4 was identified as Bacillus mojavensis on the basis of the 16S rRNA gene sequencing and biochemical properties. Influence of medium composition and pH on the production of polygalacturonases by Aspergillus oryzae BABT

A liquid medium containing wheat bran, salts and a source of inducer (pectin) was found to be suitable for the production of exo- and endo-polygalacturonases by Aspergillus oryzae CCT3940. Induction of polygalacturonases by purified pectin was significantly higher than when rinds of citrus fruits were used as inducer. A. oryzae growth was favoured by pH close to 4, although a drop of pH to around 3 was needed for enzymes production. Afterwards, decreasing activities were observed with the normal increase in pH to near neutrality. The highest activities were achieved with an initial pH of 4 and controlled when it decreased to a value slightly below 3 (159 units endo-PG.mL-1 at 83 h and 45 units exo-PG.mL-1 at 64 h), being the loss in polygalacturonases...

This study aimed to determine the influence of process variables and production, of polygalacturonase (PG) and polymetylgalacturonase (PMG) by solid substrate cultivation using a fixed bed column bioreactor. A fractional factorial design (FFD) was used to study the effect of the following variables: microorganism (Aspergillus oryzae and Aspergillus niger), substratum (wheat bran and defatted rice bran), aeration (40 and 60 ml h-1g-1), pectin (5 and 10 g g-1) and nitrogen (urea and ammonium sulfate). Partial purification and characterization of exopolygalacturonase II and III of Penicillium frequentans BJM

Previous studies from our laboratory have demonstrated that the fungus Penicillium frequentans produces high levels of polygalacturonase and pectinesterase. Endopolygalacturonase I (Endo-PG I) and Exopolygalacturonase I (Exo-PG I) were previously purified and characterized. In the present study two extracellular polygalacturonases were separated, partially purified and biochemically characterized. Both were characterized as exopolygalacturonases so they were named exopolygalacturonase II (Exo-PG II) and exopolygalacturonase III (Exo-PG III) which had a molecular mass of 63 kDa (Exo-PG II) and 79 kDa (Exo-PG III). Pectinase and polygalacturonase production by a thermophilic Aspergillus fumigatus isolated from decomposting orange peels BJM

A thermophilic fungal strain producing both pectinase and polygalacturonase was isolated after primary screening of 120 different isolates. The fungus was identified as Aspergillus fumigatus Fres. MTCC 4163. Using solid-state cultivation, the optimum levels of variables for pectinase and polygalacturonase (PG) production were determined. Maximal levels of enzyme activities were achieved upon growing the culture in a medium containing wheat bran, sucrose, yeast extract and (NH4)2SO4 after 2-3 days of incubation at a temperature of 50ºC. Highest enzyme activities of 1116 Ug-1 for pectinase and 1270 Ug-1 for polygalacturonase were obtained at pH 4.0 and 5.0, respectively.

Many enzymes produced by fungi have relevant biotechnological applications in several industrial areas. The purpose of this study was to collect and isolate filamentous fungi from soil and humus, plants and sugar cane bagasse of different regions of the São Paulo state. Forty isolates were examined for their ability to produce xylanase, glucose-oxidase, alkaline phosphatase, acid phosphatase, phytase, pectinase and amylase. Among these, twenty three isolates exhibited enzymatic potential. The xylanases produced by two of these isolates (Aspergillus caespitosus and A. phoenicis) showed good potential for pulp bleaching.
