Statistical tools application on dextranase production from Pochonia chlamydosporia (VC4) and its application on dextran removal from sugarcane juice Anais da ABC (AABC)
SUFIATE,BRUNA L.; SOARES,FILIPPE E.F.; GOUVEIA,ANGÉLICA S.; MOREIRA,SAMARA S.; CARDOSO,EVANDRO F.; TAVARES,GABRIELLA P.; BRAGA,FABIO R.; ARAÚJO,JACKSON V. DE; QUEIROZ,JOSÉ H. DE.
ABSTRACT The aim of this study was to optimize the dextranase production by fungus Pochonia chlamydosporia (VC4) and evaluate its activity in dextran reduction in sugarcane juice. The effects, over the P. chlamydosporia dextranase production, of different components from the culture medium were analyzed by Plackett-Burman design and central composite design. The response surface was utilized to determine the levels that, among the variables that influence dextranase production, provide higher production of these enzymes. The enzymatic effect on the removal of dextran present in sugarcane juice was also evaluated. It was observed that only NaNO3 and pH showed significant effect (p<0.05) over dextranase production and was determined that the levels which...

Biodegradation of crude petroleum oil and environmental pollutants by Candida tropicalis strain BABT
Farag,Soha; Soliman,Nadia A..
A local yeast isolate named A was isolated from polluted area of Abou-Qir gulf (Alexandria, Egypt), identified according to a partial sequence of 18sRNA as Candida tropicalis. The isolate showed a high potency in petroleum oil biodegradation as well some hydrocarbons. Morphological changes in cell diameter of this yeast were recognized upon growing the target cell in sea water medium supplemented with petroleum oil as sole carbon source in comparison to the growth in enriched medium. Statistically-based experimental design was applied to evaluate the significance of factors on petroleum oil biodegradation by this yeast isolate. Eleven culture conditions were examined by implementing Plackett-Burman factorial design where aeration, NH4Cl and K2HPO4 had the...

Exopolysaccharide production from Bacillus velezensis KY471306 using statistical experimental design BJM
Moghannem,Saad A.M.; Farag,Mohamed M.S.; Shehab,Amr M.; Azab,Mohamed S..
Abstract Exopolysaccharide (EPS) biopolymers produced by microorganisms play a crucial role in the environment such as health and bio-nanotechnology sectors, gelling agents in food and cosmetic industries in addition to bio-flocculants in the environmental sector as they are degradable, nontoxic. This study focuses on the improvement of EPS production through manipulation of different culture and environmental conditions using response surface methodology (RSM). Plackett-Burman design indicated that; molasses, yeast extract and incubation temperature are the most effective parameters. Box-Behnken RSM indicated that; the optimum concentration for each parameter was 12% (w/v) for molasses, 6 g/L yeast extract and 30 °C for incubation temperature. The most...

Optimizing a culture medium for biomass and phenolic compounds production using Ganoderma lucidum BJM
Zárate-Chaves,Carlos Andrés; Romero-Rodríguez,María Camila; Niño-Arias,Fabián Camilo; Robles-Camargo,Jorge; Linares-Linares,Melva; Rodríguez-Bocanegra,María Ximena; Gutiérrez-Rojas,Ivonne.
The present work was aimed at optimizing a culture medium for biomass production and phenolic compounds by using Ganoderma lucidum. The culture was optimized in two stages; a Plackett-Burman design was used in the first one for identifying key components in the medium and a central composite design was used in the second one for optimizing their concentration. Both responses (biomass and phenolic compounds) were simultaneously optimized by the latter methodology regarding desirability, and the optimal concentrations obtained were 50.00 g/L sucrose, 13.29 g/L yeast extract and 2.99 g/L olive oil. Maximum biomass production identified in these optimal conditions was 9.5 g/L and that for phenolic compounds was 0.0452 g/L, this being 100% better than that...

Statistical evaluation of nutritional components impacting phycocyanin production in Synechocystis SP BJM
Deshmukh,Devendra V.; Puranik,Pravin R..
Alkaliphilic cyanobacterial cultures were isolated from Lonar lake (MS, India). Among the set of cultures, Synechocystis sp, was studied for phycocyanin production. A maximum yield was obtained in BG-11 medium at optimized conditions (pH 10 and 16 h light). In order to increase the phycocyanin yield media optimization based on the eight media components a Plackett-Burman design of the 12 experimental trials was used. As per the analysis CaCl2.2H2O and Na2CO3 have been found to be the most influencing media components at 95% significance. Further the optimum concentrations of these components were estimated following a Box Wilson Central Composite Design (CCD) with four star points and five replicates at the center points for each of two factors was adopted...
