The lift pool method for isolation of cDNA clones from lambda phage libraries Electron. J. Biotechnol.
LeBlanc-Straceski,Janine; Sobrado,Pablo; Betz,Sharon; Zerfas,Julie; Morgan,Karen.
A PCR based strategy was developed to screen a Xenopus oocyte λgt10 cDNA library. The PCR-based lift pool (LP) method follows the same two tiered strategy as conventional screening of phage libraries by filter hybridization. Two rounds of plating, one at high density to detect the clone, and one at low density to purify the clone to homogeneity, are performed. In the first round, lysates from high density plates, termed plate pools (PP), serve as template for PCR. In the second round, phage particles adhering to plaque lifts of low density plates are washed off nitrocellulose filters to create LPs, which are used as template for PCR. The integrity of the plaques on the low-density plates is preserved. Once a positive LP has been identified,...

Un modèle asymptotique de la plaque stratifiée ArchiMer
Kail, R; Lene, F; De Roeck, Y.
In order to calculate the behaviour of thin structures of laminated composite materials, a bidimensional plate model is generated by an asymptotic development technique. From a structural analysis by plane finite elements at the middle surface, this model yields the equations leading to the computation, at each point of the laminated material, of the displacement and the stress tensor. After describing the theoretical model, numerical results are compared with the analytical solution of a test problem.

The in vitro comparative cytopathology of a porcine rotavirus and the simian prototype (SA-11) Arq. Bras. Med. Vet. Zootec.
Lonien,S.C.H.; Bolognini,A.M.; Linhares,R.E.C.; Nozawa,C.M..

Carotid intima-media thickness and plaques in internal carotid artery as surrogate markers of lower limb arterial lesions in Chinese patients with diabetic foot BJMBR
Zhang,Mei; Wen,Xiaorong; Zhou,Chenyun; Huang,Jing; He,Ying.
Peripheral arterial disease (PAD) can impair healing of diabetic foot (DF) in patients with diabetes mellitus. To determine whether carotid intima-media thickness (CIMT) can predict lower limb arterial lesions in patients with DF, this cross-sectional study enrolled patients with DF at West China Hospital (China) between January 2012 and December 2015. Ultrasonography was used to measure CIMT, assess the internal carotid arteries (ICA) for plaques, and evaluate lower limb segmental arteries for stenosis. The optimal CIMT cutoff for detecting lower limb PAD was determined by receiver operating characteristic (ROC) curve analysis. Factors associated with PAD were identified by logistic regression analyses. A total of 167 patients (mean age: 69.7±10.3 years;...

Reproducibility of carotid ultrasound measurements in the Brazilian Longitudinal Study of Adult Health (ELSA-Brasil) at baseline BJMBR
Santos-Neto,P.J.; Sena-Santos,E.H.; Meireles,D.P.; Santos,I.S.; Bensenor,I.M.; Lotufo,P.A..
Carotid artery assessment by ultrasound is a non-invasive evaluation of subclinical atherosclerosis and a predictor of cardiovascular events. However, ultrasound examinations are operator-dependent. In the Brazilian Longitudinal Study of Adult Health (ELSA-Brasil), ultrasound images have been acquired from more than 10,000 participants. In this article, we describe the reproducibility of carotid intima-media thickness (CIMT), carotid plaque detection, and carotid plaque score (defined as the number of arterial sites with plaques) using ELSA-Brasil protocol, in a subset of 118 participants. Two board-certified radiologists and a trained technician read carotid images. We calculated intra- and inter-observer intraclass correlation (ICC) for CIMT values. We...

Role of chemokines in promoting instability of coronary atherosclerotic plaques and the underlying molecular mechanism BJMBR
Zhong,Z.X.; Li,B.; Li,C.R.; Zhang,Q.F.; Liu,Z.D.; Zhang,P.F.; Gu,X.F.; Luo,H.; Li,M.J.; Luo,H.S.; Ye,G.H.; Wen,F.L..
Our aim was to investigate the role of chemokines in promoting instability of coronary atherosclerotic plaques and the underlying molecular mechanism. Coronary angiography and intravascular ultrasound (IVUS) were performed in 60 stable angina pectoris (SAP) patients and 60 unstable angina pectoris (UAP) patients. The chemotactic activity of monocytes in the 2 groups of patients was examined in Transwell chambers. High-sensitivity C-reactive protein (hs-CRP), monocyte chemoattractant protein-1 (MCP-1), regulated on activation in normal T-cell expressed and secreted (RANTES), and fractalkine in serum were examined with ELISA kits, and expression of MCP-1, RANTES, and fractalkine mRNA was examined with real-time PCR. In the SAP group, 92 plaques were detected...

Red wine consumption, coronary calcification, and long-term clinical evolution BJMBR
da Luz,P.L.; Favarato,D.; Moriguchi,E.H.; de Carli,W.; Bruscato,N.; Mochiduky,R.I.; Schwartzman,P.; Rochitte,C.E.; Laurindo,F.R..
Coronary artery calcification (CAC) is associated with atherosclerotic complications. However, elevated CAC may not always imply a worse prognosis. Herein, we report the clinical evolution of long-term red wine (RW) drinkers in relation to CAC. We followed 200 healthy male habitual RW drinkers and compared them to 154 abstainers for a period of 5.5 years. The initial evaluation included coronary computed tomography angiography (CTA), clinical, demographics, and laboratory data. CAC was quantified by the Agatston score. The follow-up process was conducted by telephone calls and/or hospital record review. The composite end-point of total death, acute myocardial infarction (AMI), or coronary revascularization (or major adverse cardiac event - MACE) was...
