A QUESTÃO AMBIENTAL: FLORESTAS E BIODIVERSIDADE Ciências Agrárias
Bertolino, Maria Lúcia.
This work has as objective to argue the environmental question concerning the conservation of the forests and biodiversity. One major issue in this question is that the Brazilian forest sector contributes with an important parcel for the Brazilian economy, generates products for direct consumption or for exportation, generates taxes and jobs for the population and, still, bring positive and negative consequences in the conservation and preservation of both renewable and not renewable natural resources. The forest economy has been, historically, responsible for one of the five largest positive commercial balances of the country and still it supplies 85% of all the products of forest origin found in the national market, therefore, alleviating the pressure on...

Genetic diversity in natural populations of Jacaranda decurrens Cham. determined using RAPD and AFLP markers Genet. Mol. Biol.
Bertoni,Bianca W.; Telles,Mariana P. de C.; Malosso,Milena G.; Torres,Simone C.Z.; Pereira,José O.; Lourenço,Mirian V.; França,Suzelei de C.; Pereira,Ana M.S..
Jacaranda decurrens (Bignoniaceae) is an endemic species of the Cerrado with validated antitumoral activity. The genetic diversity of six populations of J. decurrens located in the State of São Paulo was determined in this study by using molecular markers for randomly amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP). Following optimization of the amplification reaction, 10 selected primers generated 78 reproducible RAPD fragments that were mostly (69.2%) polymorphic. Two hundred and five reproducible AFLP fragments were generated by using four selected primer combinations; 46.3% of these fragments were polymorphic, indicating a considerable level of genetic diversity. Analysis of molecular variance (AMOVA) using these two...

A benthic Si mass balance on the Congo margin: Origin of the 4000 m DSi anomaly and implications for the transfer of Si from land to ocean ArchiMer
Ragueneau, O.; Regaudie-de-gioux, A.; Moriceau, B.; Gallinari, M.; Vangriesheim, Annick; Baurand, F.; Khripounoff, Alexis.
To elucidate the origin of the silicic acid (DSi) anomaly observed along the 4000 isobath on the Congo margin, we have established a benthic Si mass balance and performed direct measurements of biogenic silica (bSiO(2)) dissolution in the deep waters and in the sediments. Results strongly suggest that the anomaly originates from the sediments; the intensity of DSi recycling is consistent with the degradation of organic matter, as observed from Si:O-2 ratios in the benthic fluxes compared to that ratio observed in the anomalies. Strong imbalances, observed in both the Si and C mass balances, suggest that the biogenic matter that degrades and dissolves in these sediments near 4000 m does not come from pelagic sedimentation. It is probably not coming also...

Effects of high pressure on texture and microstructure of sea bass (Dicentrarchus labrax L.) fillets ArchiMer
Cheret, Romuald; Chapleau, Nicolas; Delbarre Ladrat, Christine; Verrez-bagnis, Veronique; De Lamballerie, Marie.
High pressure is an innovative non-thermal food-preservation technology. We studied the effect of high-pressure treatment up-to 500 MPa for 5 min on physical characteristics of sea bass fillets after 0, 7, and 14 d of refrigerated storage.. Color results exhibited an increase of lightness and a slight change of hue, which might be imperceptible in cooked fish. High-pressure treatment induced a decrease of exudation and water-holding capacity Pressure treatment above 300 MPa provoked higher fish hardness after storage than in untreated sample, proving the ability of high pressure to improve textural, quality of chilled, stored fish fillet. These assessments were corroborated with microstructure observations. We showed that high-pressure treatment at 500 MPa...

Sensitivity of clumped isotope temperatures in fossil benthic and planktic foraminifera to diagenetic alteration ArchiMer
Leutert, Thomas J.; Sexton, Philip F.; Tripati, Aradhna; Piasecki, Alison; Ling Ho, Sze; Nele Meckler, A..
Applying the clumped isotope (Δ47) thermometer to foraminifer microfossils offers the potential to significantly improve paleoclimate reconstructions, owing to its insensitivity to the isotopic composition of seawater (unlike traditional oxygen isotope (δ18O) analyses). However, the extent to which primary Δ47 signatures of foraminiferal calcites can be overprinted during diagenesis is not well known. Here, we present Δ47 data as well as high-resolution (∼10 kyr) δ18O and δ13C middle Eocene time series, measured on benthic and planktic foraminifera from ODP/IODP Sites 1408, 1409, 1410, 1050, 1260 and 1263 in the Atlantic Ocean. The sites examined span various oceanographic regimes, including the western tropical to mid-latitude North Atlantic, and the...

Pavlova lutheri: Production, preservation and use as food for Crassostrea gigas larvae ArchiMer
Ponis, Emanuele; Parisi, G; Chini Zittelli, G; Lavista, F; Robert, Rene; Tredici, M. R..
The haptophyte Pavlova lutheri was cultivated in a 4-l flat alveolar photobioreactor (FAP) in semi-continuous mode during 35 days, achieving a mean volumetric productivity of 0.4 g l-1 day-1. The microalgal culture was harvested from the photobioreactor at an average concentration of 3.5 g l-1 and subjected to different storage conditions in which the effects of temperature (1 °C, 4 °C and 25 °C), air supply and preservative (ascorbic or propionic acid) addition on the viability of the stored suspensions were investigated. Low temperatures (1 and 4 °C) and air-bubbling extended the shelf-life of the stored suspension, while no beneficial effects of ascorbic or propionic acid were found. The influence of temperature and air-bubbling on fatty acid content...

Conservation des laminaires apres sechage a tres hautes temperatures. ArchiMer
Perez, René.
Laminaria digitata is the only brown alga used in the French alginate industry. Air-dried and flash-dried (in a current of air at 200 to 850 degree C) algae were compared. Quality was assessed viscometrically and the results expressed graphically. Quality after drying and keeping properties were found better in flash-dried algae and the method was shown to be the cheapest of methods currently available.

Freezing and chemical preservatives alter the stable isotope values of carbon and nitrogen of the Asiatic clam (Corbicula fluminea) ArchiMer
Syvaeranta, Jari; Martino, Aurelia; Kopp, Dorothee; Cereghino, Regis; Santoul, Frederic.
We tested the impacts of most common sample preservation methods used for aquatic sample materials on the stable isotope ratios of carbon and nitrogen in clams, a typical baseline indicator organism for many aquatic food web studies utilising stable isotope analysis (SIA). In addition to common chemical preservatives ethanol and formalin, we also assessed the potential impacts of freezing on delta C-13 and delta N-15 values and compared the preserved samples against freshly dried and analysed samples. All preservation methods, including freezing, had significant impacts on delta C-13 and delta N-15 values and the effects in general were greater on the carbon isotope values (1.3-2.2aEuro degrees difference) than on the nitrogen isotope values (0.9-1.0aEuro...

An Improved Protocol for Flow Cytometry Analysis of Phytoplankton Cultures and Natural Samples ArchiMer
Marie, Dominique; Rigaut-jalabert, Fabienne; Vaulot, Daniel.
Preservation of cells, choice of fixative, storage, and thawing conditions are recurrent issues for the analysis of phytoplankton by flow cytometry. We examined the effects of addition of the surfactant Pluronic F68 to glutaraldehyde-fixed photosynthetic organisms in cultures and natural samples. In particular, we examined cell losses and modifications of side scatter (a proxy of cell size) and fluorescence of natural pigments. We found that different marine phytoplankton species react differently to the action of Pluronic F68. In particular, photosynthetic prokaryotes are less sensitive than eukaryotes. Observed cell losses may result from cell lysis or from cell adhesion to the walls of plastic tubes that are commonly used for flow cytometry analysis....

Freeze-dried Spermatozoa and Freeze-dried Sperm Injection into Oocytes OAK
SUZUKI, Hiroshi; 鈴木, 宏志.
Attempts to freeze-dry spermatozoa are not new. Successful conception and full-term development using freeze-dried spermatozoa were reported in the cow and rabbit prior to 1960. However, these preliminary results have not been confirmed. Spermatozoa become defective in motility after freeze-drying and are unable to fertilize eggs both in vivo and in vitro. However, in 1998, freeze-drying of mammalian spermatozoa was demonstrated without loss of genetic or reproductive potential. Freeze-dried spermatozoa support normal development when injected directly into oocytes by means of an intracytoplasmic sperm injection (ICSI) technique. Recent investigations of freeze-dried sperm have focused on the factors affecting freeze-drying in spermatozoa in an effort to...

Salting kinetics and salt diffusivities in farmed Pantanal caiman muscle PAB
Telis,Vânia Regina Nicoletti; Romanelli,Pedro Fernando; Gabas,Ana Lúcia; Telis-Romero,Javier.
The legal Pantanal caiman (Caiman crocodilus yacare) farming, in Brazil, has been stimulated and among meat preservation techniques the salting process is a relatively simple and low-cost method. The objective of this work was to study the sodium chloride diffusion kinetics in farmed caiman muscle during salting. Limited volumes of brine were employed, with salting essays carried at 3, 4 and 5 brine/muscle ratios, at 15%, 20% and 25% w/w brine concentrations, and brine temperatures of 10, 15 and 20ºC. The analytical solution of second Fick's law considering one-dimensional diffusion through an infinite slab in contact with a well-stirred solution of limited volume was used to calculate effective salt diffusion coefficients and to predict the sodium...

Land Preservation in British Columbia: An Empirical Analysis of the Factors Underlying Public Support and Willingness to Pay AgEcon
Androkovich, Robert A.; Desjardins, Ivan; Tarzwell, Gordon; Tsogaris, Peter.
This study extends previous empirical research on land preservation by considering an actual land preservation scheme, the agricultural land reserve in British Columbia, Canada. The reserve was established in 1973 to ensure that development did not occur on the province’s most productive agricultural land. ‘To ensure that local food production is maintained,’ ‘the economic importance of British Columbia’s agricultural sector,’ and ‘to protect the environment’ are the most important factors that underlie support for the reserve. Aggregate, provincewide willingness to pay to maintain the land reserve is substantial, with our most conservative estimate being Can$91.18 million per year.

Evaluation of glycerin as preserving agent of chicken serum for plate agglutination test Rev. Bras. Ciênc. Avic.
Freitas,ES de; Soldá,A; Calegari,DL; Munaretto,FC; Dietrich,GF; Back,A.
Serum is widely used for the purpose of monitoring and diagnosis support for most of poultry diseases. In the case of the serum plate agglutination test (SPA), commonly used to detect antibodies for Salmonella Pullorum (SP), Mycoplasma gallisepticum (MG) and Mycoplasma synoviae (MS), serum cannot be frozen because it may result in false positive. Without freezing, serum can last only for a few days. In this experiment, glycerin was evaluated as a serum preservering agent. About 50 samples for each disease and analyzed by SPA test previously were separated. Glycerin was added to serum from commercial chickens, with and without antibodies for SP, MG and MS, in the proportion of 1:1 (serum:glycerin) and kept at refrigerated conditions (2 to 8 ºC). For four...

Lactobacillus spp. Strains Isolation, Identification, Preservation and Quantitative Determinations from Gut Content of 45-Day-Old Chickens Broilers Rev. Bras. Ciênc. Avic.
Sorescu,I; Dumitru,M; Ciurescu,G.
ABSTRACT The objective of this study was to isolate, identify, preserve and determine the quantitative level of the Lactobacillus strains from the gut content of 45-day-old chickens broilers; to test the viability of these strains preserved at 4 ºC and room temperature (20 ± 2 ºC). Lactobacillus strains were isolated, phenotypically identified and preserved from the gut content of 17 chickens broilers. Identification was performed by morphological, cultural and biochemical characters examination, using apiwebTM and ABIS online software. The quantitative level of Lactobacillus strains in intestinal content (105 - 109 CFU/g) and the viability of strains preserved at 4 ºC and at room temperature (from 8 days to 9 months) was also determined. Twenty-three...
