Eddy-Current Modeling of a Continous Conductivity Profile Resulting From a Diffusion Process ArchiMer
Lambert, Marc; Nouguier, Frederic; Zorgati, Riadh.
This paper deals with the modelization of eddy-current measurements over combustion turbine blade coatings affected by depletion of aluminum. First, we model the response of an eddy-current coil over a layered metallic structure with a top over-aluminized coating by extending the analytical Uzal-Rose's model for one hyperbolic tangent conductivity profile to a conductivity profile using two hyperbolic tangents for taking inward and outward depletion of aluminum inside the coating into account. Results obtained with this model are similar to those obtained with a numerical multilayer model but with a reduced computing time.

Specificity Re-evaluation of Oligonucleotide Probes for the Detection of Marine Picoplankton by Tyramide Signal Amplification-Fluorescent In Situ Hybridization ArchiMer
Riou, Virginie; Périot, Marine; Biegala, Isabelle.
Oligonucleotide probes are increasingly being used to characterize natural microbial assemblages by Tyramide Signal Amplification-Fluorescent in situ Hybridization (TSA-FISH, or CAtalysed Reporter Deposition CARD-FISH). In view of the fast-growing rRNA databases, we re-evaluated the in silico specificity of eleven bacterial and eukaryotic probes and competitor frequently used for the quantification of marine picoplankton. We performed tests on cell cultures to decrease the risk for non-specific hybridization, before they are used on environmental samples. The probes were confronted to recent databases and hybridization conditions were tested against target strains matching perfectly with the probes, and against the closest non-target strains presenting one...

Garlic viral complex: identification of Potyviruses and Carlavirus in Central Brazil Trop. Plant Pathol.
FAJARDO,THOR V. M.; NISHIJIMA,MARTA; BUSO,JOSÉ A.; TORRES,ANTÔNIO C.; ÁVILA,ANTÔNIO C.; RESENDE,RENATO O..
Garlic viruses often occur in complex infections in nature. In this study, a garlic virus complex, collected in fields in Brazil, was purified. RT-PCR was performed using specific primers designed from the consensus regions of the coat protein genes of Onion yellow dwarf virus, a garlic strain (OYDV-G) and Leek yellow stripe virus (LYSV). cDNA of Garlic common latent virus (GCLV) was synthesized using oligo-dT and random primers. By these procedures individual garlic virus genomes were isolated and sequenced. The nucleotide sequence analysis associated with serological data reveals the presence of two Potyvirus OYDV-G and LYSV, and GCLV, a Carlavirus, simultaneously infecting garlic plants. Deduced amino acid sequences of the Brazilian isolates were...

Homology among extra-cryptic DNA bands and the typical plasmids in Brazilian Yersinia pestis strains BJM
Leal,Nilma Cintra; Sobreira,Marise; Leal,Tereza Cristina Arcanjo; Almeida,Alzira Maria Paiva de.
Yersinia pestis, the etiologic agent of plague, harbors three well-characterized plasmids: pFra (90-110kb), pYV (70 kb) and pPst (9.5 kb). Furthermore, some extra-cryptic DNA bands have been observed in a number of wild strains from several foci of the world. Additional bands have also been reported in Brazilian strains. Looking for any relationship among these cryptic DNA bands and the three-prototypical plasmids, we analyzed twelve strains displaying different plasmid content. The DNA bands were hybridized by southern blot with probes directed at the genes caf1, lcrV and pla located respectively on the plasmids pFra, pYV and pPst. The probes were constructed by PCR amplification and labeled with digoxigenin. The Pla probe hybridized with its target...
