La deficiencia en la capacidad de unión de leucocitos bovinos a los antígenos, más conocida como BLAD, es una enfermedad hereditaria que resulta letal para el ganado de la raza Holstein. Su principal característica es ser una enfermedad autosómica recesiva que puede ser transmitida a la descendencia. En muchas áreas sembradas con papa en México se manifiesta el síndrome de la punta morada, que reduce el rendimiento y la calidad del tubérculo. Un factor principal de este síndrome es la presencia de fitoplasmas y el inadecuado manejo de su insecto vector. Una opción para controlar esta enfermedad es generar genotipos de papa resistentes y un diagnóstico oportuno para evitar su diseminación. Recientemente en el estado de Tlaxcala, México el "síntoma de escoba de bruja" (proliferación de brotes y reducción de entrenudos) fue observado en amaranto (Amaranthus hypochondriacus L.). Esta sintomatología en campo se distribuye uniformemente en la plantación y no en forma azarosa y de manchones; además, el mayor daño es en la panícula que aloja a las semillas lo cual genera considerables perdidas en la cosecha. Informações sobre peso à desmama de bezerros Nelore foram utilizadas após ajuste para idade padrão aos 205 dias, sexo, idade da mãe, touro e mês de desmama, para separar as reprodutrizes em dois grupos, segundo o peso de suas crias. As médias de peso dos bezerros ajustadas pelo método dos quadrados mínimos e erros-padrão (LSM± SE) foram para os grupos pesados (P) e leves (L) 163,21± 2,18kg e 134,44± 2,18kg, respectivamente, com 41 animais em cada grupo. The population structure and genotypic diversity of Septoria tritici Rob ex Desm. from two crop field populations in Buenos Aires Province were studied with DNA restriction fragment length polymorphism. Hierarchical samples were taken from different cultivars. A total of 120 single spore isolates was subject to molecular analysis to compare the genetic structure of Los Hornos and Balcarce populations separated by 500km. Genetic structure of Brazilian populations of white mouth croaker Micropogonias furnieri (Perciformes : Sciaenidae) BABT

Mitochondrial (mt) DNA population structure was examined by using restriction fragment length polymorphism analysis (RFLP) among 149 white mouth croakers (Micropogonias furnieri) sampled from six locations in the Brazilian coast. Heterogeneity tests revealed no differentiation in mtDNA haplotype frequencies within the region between 23°S and 34°S (P=0.263), but significant heterogeneity was detected between north and south of 23°S (P<0.003). High quality DNA from human papillomavirus (HPV) for PCR/RFLPs BABT

The analysis of DNA in clinical samples for a secure diagnostic has become indispensable nowadays. Techniques approaching isolation of high molecular weigth DNA of HPV could lead to efficient amplification and early clinical diagnosis of the virus DNA by PCR (polymerase chain reaction). We describe a fast, non-toxical, efficient and cheap method for DNA isolation of human papilloma virus (HPV) from cervical smears using guanidine (DNAzol solution). Mycobacterium simiae infection in a patient with Acquired Immunodeficiency Syndrome BJID

Mycobacterium simiae is usually an environmental contaminant rarely associated with human disease. We report a fatal case of M.simiae infection in a 37 year old, HIV positive, male from whom the organism was isolated from blood culture. The identification of M.simiae was performed using DNA amplification followed by analysis on 3% agarose gel of the amplicon fragments after digestion by restriction endonucleases. The precise identification of mycobacterial isolates to the species level is important, with both epidemiological and therapeutic implications.

Typing class I HLA-A gene using a nested PCR-RFLP procedure BJMBR

In order to detect several new HLA-A class I alleles that have been described since 1998, the original PCR-RFLP method developed to identify the 78 alleles recognized at that time at high resolution level was adapted by us for low and medium resolution levels using a nested PCR-RFLP approach. The results obtained from blood samples of 23 subjects using both the PCR-RFLP method and a commercial kit (MicroSSP1A®, One Lambda Inc.) showed an agreement higher than 95%. The PCR-RFLP adapted method was effective in low and medium resolution histocompatibility evaluations.

Characterization and lytic activity of Pseudomonas fluorescens phages from sewage BJM

Pseudomonas fluorescens phages from sewage were tested against P. fluorescens isolates of soil and sewage. The phages were characterized as to host range, morphology, structural proteins and genome fingerprint. Of the seven phages isolated, one was found to be abundant in sewage (5.9×10(7) pfu/mL), having broad host range, and distinct protein and DNA profile when compared to the other six phages. DNA restriction and protein profiles of the phages and their morphology indicate the diversity in the sewage environment. None of the isolates from the rhizosphere regions of various cultivated soils were susceptible to phages isolated from sewage.

Genotypic characterization of psittacid herpesvirus isolates from Brazil BJM

Abstract Thirty-six isolates of psittacid herpesvirus (PsHV), obtained from 12 different species of psittacids in Brazil, were genotypically characterized by restriction fragment length polymorphism (RFLP) analysis and PCR amplification. RFLP analysis with the PstI enzyme revealed four distinct restriction patterns (A1, X, W and Y), of which only A1 (corresponding to PsHV-1) had previously been described. To study PCR amplification patterns, six pairs of primers were used. Identification of Leptospira serovars by RFLP of the RNA polymerase beta subunit gene (rpoB) BJM

Leptospires are usually classified by methods based on DNA-DNA hybridization and the conventional cross-agglutination absorption test, which uses polyclonal antibodies against lipopolysaccharides. In this study, the amplification of the rpoB gene, which encodes the beta-subunit of RNA polymerase, was used as an alternative tool to identify Leptospira. Human cytomegalovirus (HCMV) displays genetic variability in several regions, supposed to be related with strain-specific tissue tropism and immunopathogenesis. Based on sequence variation in the UL55 gene that encodes gB glycoprotein, HCMV strains can be assigned to one of four genotypes. Previous studies have addressed gB genotyping mostly by investigating strains derived from immunosuppressed patients, sometimes without previous knowledge about genotype distribution in a geographic area.
