Expression of RNA polymerase IV and V in Oryza sativa Electron. J. Biotechnol.
Moo,Leydi de Rocio Canche; González,Angela Kú; Rodríguez-Zapata,Luis Carlos; Suarez,Victor; Castaño,Enrique.
RNA polymerase IV and V are principal players in the RdDM pathway, where their current study has shown interaction of several factors that control DNA silencing of intergenic regions and siRNA production. DNA silencing is an important process during cell differentiation, nuclear structure and viral control. However, RNA pol IV and V are yet to be study in model monocot systems like Oryza sativa that can provide further information on genetic silence mechanism in plats. We show the expression pattern of these polymerases in tissues extracts of Oryza sativa. Detectable amounts of these polymerases are found in specific adult plant tissues and particularly expressed during somatic embryogenesis but not during early stages of normal embryo development. The use...

Screening for propagation suitability in vitro of different Cyclamen species Electron. J. Biotechnol.
Seyring,Martina; Ewald,Aloma; Mueller,Anke; Haensch,Klaus-Thomas.
In the present study we examined the possibility of propagating different Cyclamen species (C. africanum Boiss. and Reut., C. cilicium Boiss. and Heldr., C. coum Mill., C. hederifolium Ait., C. persicum Mill., C. purpurascens Mill.) including some of their subspecies and cultivars in vitro using explants of adult plants. For this purpose two protocols have been applied to eleven genotypes combined with mostly four explant types (placentas with ovules, leaves, petioles and peduncles). The use of these protocols has given rise to either somatic embryo-like structures and/or adventitious shoots in all genotypes. This way it was possible to propagate each of the examined genotypes in vitro using explants of adult plants in a time less than one year. These...

Influence of 2,4-D and BAP on callus growth and the subsequent regeneration of somatic embryos in long-term cultures of Pelargonium x domesticum cv. Madame Layal Electron. J. Biotechnol.
Haensch,Klaus-Thomas.
This long-term study demonstrates for the first time that it is possible to propagate embryogenic cultures in pelargoniums and to subsequently initiate the differentiation of embryos using the cultivar Madame Layal (Pelargonium x domesticum). Propagation of callus was only possible with combinations of 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP), which gave rise to embryos from the primary culture stage on. However, the propagation of cells, as well as the differentiation of embryos, was inhibited by a continuous application of these growth regulators. For this reason, a long period on medium lacking growth regulators was necessary before the differentiation of embryos occurred again. The consequences for improving the propagation...

Induction of somatic embryogenesis as an example of stress-related plant reactions Electron. J. Biotechnol.
Zavattieri,Maria Amely; Frederico,António Miguel; Lima,Mónica; Sabino,Rui; Arnholdt-Schmitt,Birgit.
In this review, we address the role of stress as one of the principal causes for a cell or tissue to change its pre-existing somatic program, reprogramming itself to express the embryogenic pathway. The focus of this paper is the effect of different stress conditions on the induction phase of plant somatic embryogenesis, as well as the development of embryogenic competence as a result of the applied stresses. We also present a variety of data that link plant somatic embryogenesis, DNA methylation and oxidative stress response.

Somatic embryogenesis and plant regeneration capacity in Argentinean maize (Zea mays L.) inbred lines Electron. J. Biotechnol.
González,Germán Andrés; Pacheco,María Gabriela; Oneto,Cecilia Décima; Etchart,Valeria J; Kandus,Mariana V; Salerno,Juan Carlos; Eyherabide,Guillermo; Presello,Daniel; Lewi,Dalia Marcela.
Somatic embryogenesis, which is still the method of choice for tissue culture, regeneration and transformation of maize, is largely considered highly genotype-dependent. The Hi II, a highly embryogenic genotype, has been extensively used in transformation protocols. However, this is not an inbred line; instead, it has a proportion of the undesirable A-188 background, and the progeny segregates for phenotypic characteristics and shows poor agronomic performance. In an effort to identify genotypes that combine a high somatic embryogenic response and good agronomic performance, we evaluated 48 advanced inbred lines developed at INTA. Callus development and somatic embryogenesis capacity were measured in 200 immature embryos per line. Embryogenic capacity [EC...

Selection of reference genes for normalization of quantitative real-time PCR in cell cultures of Cyclamen persicum Electron. J. Biotechnol.
Hoenemann,Claudia; Hohe,Annette.
As a prerequisite for gene expression analyses in cell cultures of the ornamental crop Cyclamen persicum basic parameters for quantitative real-time polymerase chain reaction (qRT-PCR) have been established including the selection of reference genes using the software tools ‘geNorm’ and ‘NormFinder’. Five potential reference genes have been tested (elongation factor tu (Ef-Tu), putative ABC transporter ATPase, putative conserved oligomeric Golgi (COG) complex component, V-ATPase G subunit 1 and Histone H3-K9 methyltransferase 4 (H3-K9-HMTase 4)). ‘NormFinder’ as well as ‘geNorm’ identified Ef-Tu to be the least stable reference gene while the ranking of the most stable genes...

AgNO3: a potential regulator of ethylene activity and plant growth modulator Electron. J. Biotechnol.
Kumar,Vinod; Parvatam,Giridhar; Ravishankar,Gokare Aswathanarayana.
The aim of this review is to critically analyze the role of silver nitrate (AgNO3) in modulating plant growth and development. In recent years, basic studies on ethylene regulation opened new vistas for applied research in the area of micro-propagation, somatic embryogenesis, in vitro flowering, growth promotion, fruit ripening, and sex expression. Silver nitrate has proved to be a very potent inhibitor of ethylene action and is widely used in plant tissue culture. Few properties of silver nitrate such as easy availability, solubility in water, specificity and stability make it very useful for various applications in exploiting plant growth regulation and morphogenesis in vivo and in vitro. Silver ion mediated responses seem to be involved in polyamines,...

Characterization of the third SERK gene in pineapple (Ananas comosus) and analysis of its expression and autophosphorylation activity in vitro Genet. Mol. Biol.
Ma,Jun; He,Yehua; Hu,Zhongyi; Xu,Wentian; Xia,Jingxian; Guo,Cuihong; Lin,Shunquan; Chen,Chengjie; Wu,Chenghou; Zhang,Junli.
Two somatic embryogenesis receptor-like kinase genes (identified as AcSERK1 and AcSERK2) have previously been characterized from pineapple (Ananas comosus). In this work, we describe the characterization of a third gene (AcSERK3) in this family. AcSERK3 had all the characteristic domains and shared extensive sequence homology with other plant SERKs. AcSERK3 expression was studied by in situ hybridization and quantitative real-time PCR to analyze its function. Intense in situ hybridization signals were observed only in single competent cells and competent cell clusters; no hybridization signal was detected in the subsequent stages of somatic embryogenesis. AcSERK3 was highly expressed in embryogenic callus compared to other organs, e.g., 20-80 fold more...

Identification of histone methylation modifiers and their expression patterns during somatic embryogenesis in Hevea brasiliensis Genet. Mol. Biol.
Li,Hui-Liang; Guo,Dong; Zhu,Jia-Hong; Wang,Ying; Peng,Shi-Qing.
Abstract Histone methylation plays a crucial role in various biological processes, from heterochromatin formation to transcriptional regulation. Currently, no information is available regarding histone methylation modifiers in the important rubber-producing plant Hevea brasiliensis. Here, we identified 47 histone methyltransferase (HMT) genes and 25 histone demethylase (HDM) genes as possible members of the histone methylation modifiers in the rubber tree genome. According to the structural features of HMT and HDM, the HbHMTs were classified into two groups (HbPRMs and HbSDGs), the HbHDMs have two groups (HbLSDs and HbJMJs). Expression patterns were analyzed in five different tissues and at different phases of somatic embryogenesis. HbSDG10, 21, 25, 33,...

Assessment of somaclonal variation in indirect morphogenesis-derived plants of Arracacia xanthorrhiza PAB
Vitamvas,Jan; Viehmannova,Iva; Cepkova,Petra Hlasna; Mrhalova,Hana; Eliasova,Katerina.
Abstract: The objective of this work was to induce and detect somaclonal variation in arracacha (Arracacia xanthorrhiza) plants regenerated via indirect morphogenesis, in order to evaluate the potential of this technique to produce new genotypes for breeding purposes of this crop. Calli were induced from petiole segments on Murashige & Skoog (MS) medium supplied with 0.1 mg L-1 2,4-dichlorophenoxyacetic acid. The regeneration of plants via indirect morphogenesis was carried out on half-strength MS medium without plant growth regulators. Fifteen randomly chosen plants were subjected to flow cytometry and “inter-simple sequence repeat” (ISSR) analysis. Ploidy level remained stable in all tested regenerants (2n=4x=44), with no changes in the genome....

Physiological and biochemical changes during organogenesis and somatic embryogenesis of HBsAg-transgenic cherry tomato mutant Phyton
Guan,Z-J; Lu,S-B; Huo,Y-L; Guo,B; Guan,Z-P; Wei,Y-H.
Leaf explants of the HBsAg-transgenic cherry tomato (Solanum lycopersicum) mutant were cultured on Murashige and Skoog (MS) basal medium, supplemented with 1.0 mg/L 6-BA and 0.05 mg/L IAA for callus induction, to clarify the physiological and biochemical characteristics of morphogenesis development. Therefore, the physiological and biochemical changes during the development of organogenic shoots and somatic embryos in the mutant were studied. Superoxide dismutase (SOD) activities of the mutant had only one peak value on the 21st day. Peroxidase (POD) activities of the mutant declined less sharply since the explants were cultured. IAA oxidase activity of the mutant increased steadily until 42 days from culturing and then decreased sharply. Malondialdehyde...

In vitro somatic embryogenesis and adventitious root initiation have a common origin in eggplant (Solanum melongena L.) Rev. Bras. Bot.
Tarré,Erika; Magioli,Claudia; Margis-Pinheiro,Márcia; Sachetto-Martins,Gilberto; Mansur,Elisabeth; Santiago-Fernandes,Lygia D. R..
Somatic embryogenesis was induced from cotyledon explants of eggplant cultured on MS medium supplemented with 54 µM NAA. Anatomical analysis of somatic embryo initiation and development was performed during the first four weeks. Proembryo formation was observed after the second day of culture, directly from perivascular cells or via pro-embryogenic masses derived from indeterminate meristematic masses (IMMs) originated in the vascular tissue. Those IMMs also gave rise to root primordia after 10 days of culture. The origin of embryos is discussed as well as the similarities between somatic embryogenesis and adventitious root formation.

Advances observed in papaya tree propagation Rev. Bras. Frutic.
Costa,Adelaide de Fátima Santana da; Abreu,Emanuel Felipe Medeiros; Schmildt,Edilson Romais; Costa,Aureliano Nogueira da; Schmildt,Omar.
Abstract Papaya (Carica papaya L.) is a fruit tree of great economic and food interest worldwide. Its propagation can occur both sexually and asexually. Traditionally, it is propagated by seeds; however, vegetative propagation has been often used with the use of in vitro and ex vitro culture techniques. In vegetative propagation, the cutting method has been extensively explored. Research has also been developed using grafting. In recent years, papaya cultivation has also become a model for other fruit plants in the use of biotechnological techniques aiming applications ranging from germplasm conservation to genetic transformation of plants. Since the beginning of the 1970s, the culture of papaya tree tissues has been intensely studied, allowing greater...
