Analysis of topological organization of chromatin during spermatogenesis in mouse testis Genet. Mol. Biol.

Eukaryotic chromatin is organized as radial DNA loops with periodical attachments to an underlying nucleoskeleton known as nuclear matrix. This higher order chromatin organization is revealed upon high salt extraction of cells. To understand the sequential change in the functional organization of chromatin during spermatogenesis, we have analysed the higher order organization of chromatin in different testicular cell types and the epididymal sperm of laboratory mouse. Spittlebugs annually infest pastures and cause severe damage, representing a serious problem for the tropical American beef cattle industry. Spittlebugs are an important biotic constraint to forage production and there is a lack of cytogenetic data for this group of insects. Chromosome complement and meiosis in three species of the Neotropical bug genus Antiteuchus (Heteroptera, Pentatomidae, Discocephalinae) Genet. Mol. Biol.

Orcein staining of spermatocytes was used to study the meiotic behavior of holocentric chromosomes in three member of the genus Antiteuchus (commonly known as stink bugs). We describe and illustrate the karyotype of Antiteuchus mixtus, A. sepulcralis and A. macraspis which were cytogenetically characterized as having a diploid number of 2n = 14 and an XY sex chromosome system showing pre-reductional meiosis for autosomes and post-reductional meiosis for sex chromosomes. These species were also shown to have a long diffuse stage during meiotic prophase I and aberrant harlequin-type meiocytes. Zaprionus indianus is a drosophilid native to the Afrotropical region that has colonized South America and exhibits a wide geographical distribution. In contrast, Z. sepsoides is restricted to certain African regions. The two species differ in the size of their testes, which are larger in Z. indianus than in Z. sepsoides. To better understand the biology and the degree of differentiation of these species, the current study evaluated spermatogenesis in males of different ages by conventional staining techniques and ultrastructural analysis. The mouse testis-enriched Znf230 gene, which encodes a type of RING finger protein, is present primarily in the nuclei of spermatogonia, the acrosome and the tail of spermatozoa. To investigate the role of Znf230 in spermatogenesis, we generated Znf230-deficient mice by disrupting Znf230 exon-5 and exon-6 using homologous recombination. The homozygous Znf230-knockout (KO) mice did not exhibit Znf230 mRNA expression and Znf230 protein production. Znf230 KO mice exhibited no obvious impairment in body growth or fertility. Morphological and cytological aspects of sex inversion in a protogynous hermaphrodite, Epinephelus microdon (Teleostei, Serranidae). ArchiMer

The sexuality of the grouper Epinephelus microdon , caught by diving in the area of Tahiti Island, from January 1986 to March 1987, is investigated. Small specimens are females, males occur among the largest fishes, revealing the protogyny of Epinephelus microdon as in other Epinephelinae. Nineteen fishes among 233 samples studied are in sex inversion.

Induction of ovulation and stimulation of spermation by LHRH or an LHRH analog associated or not with pimozide. Effects on some teleost fish. ArchiMer

Effectiveness of LHRH and one of its analogs (LHRH-A = D-Ala super(6) ethylamide) on induction of ovulation and stimulation of spermiation is reviewed here. LHRH was also combined with pimozide, an anti-dopaminergic agent, to try to cancel dopamine inhibition of gonatotropin (GtH) release. LHRH-A alone (50 mu g/kg) did not initiate ovulation but only oocyte maturation. When pimozide (10 mg/kg) and LHRH-A (50 mu g/kg) were combined, gonadotropic hormone secretion was significantly increased, causing ovulation and stimulating spermiation. This combination advanced ovulation in trout. Trout egg quality was markedly altered by the pimozide treatment; it remains to be tested in carp. The black-chinned tilapia Sarotherodon melanotheron heudelotii is an ecologically appealing model as it shows exceptional adaptive capacities, especially with regard to salinity. In spite of this, this species is devoid of genomic resources, which impedes the understanding of such remarkable features. De novo assembly of transcript sequences produced by next-generation sequencing technologies offers a rapid approach to obtain expressed gene sequences for non-model organisms. It also facilitates the development of quantitative real-time PCR (qPCR) assays for analysing gene expression under different environmental conditions.
