O objetivo deste trabalho foi avaliar a relação entre a morfometria da cabeça do espermatozoide e a compactação da cromatina com as taxas de clivagem e formação de blastocisto em programas de produção de embriões in vitro (PIV). Neste estudo foram utilizados dados de 38 rotinas de PIVs com sêmen de touros zebuínos com espermograma de rotina normal, realizadas em dois diferentes laboratórios de Uberaba, Minas Gerais, Brasil. Para avaliar a morfometria e compactação da cromatina, foram usados esfregaços de sêmen corados com azul de toluidina e avaliados por análise de imagem computacional. Short-term storage of lebranche mullet Mugil liza (Valenciennes, 1836) semen in natura and diluted with CF-HBSS 2

 The present study aimed to evaluate the activation characteristics of the lebranche mullet spermatozoa in natura and diluted with CF-HBSS for 96h at 4±2°C. The semen was collected from eight wild fish in Florianópolis – SC (Brazil) (27°S) in May, during reproductive migration. Three pools of semen were divided into two treatments: in natura and diluted with CF-HBSS 1:3. The semen was activated with seawater (salinity of 34, pH 8.7 and 4±2°C) to determine: motility time, motility rate and sperm cell membrane integrity. Dilution with CF-HBSS 1:3 increased motility time of diluted semen (15-20% for 6, 12 and 18h). ABSTRACT This study was designed to evaluate the possible benefits of adding xanthan gum to a standard extender for equine through in vitro analyzes of sperm quality. Semen was collected four times from five different stallions (n= 20 samples) and subjected to cooled storage under different conditions: control (only standard extender) and three different concentrations of xanthan gum (0.01%, 0.12%, and 0.25%) supplemented to the extenders. The present study aimed to verify the caprine semen characteristics during dry and rainy seasons in the Brazilian Northeast, and the influence of these seasons on cooled semen. Seminal volume, concentration, percentage of motile cells, vigor and spermatic morphology, as well as biochemical profile (fructose, citric acid, P, Ca2+, Mg, total proteins and phospholipase A2 activity) were analyzed. It was observed a reduction (P<0.05) in normal sperm morphology, fructose, citric acid, P, Mg and total protein concentration during the dry season, which did not affect the motility, vigor, volume and sperm concentration. Neste trabalho descreve-se a técnica do teste de resistência osmótica modificado (SORT modificado) e sua eficiência na avaliação da integridade da membrana do acrossoma do espermatozóide do varrão. Modificaram-se as soluções usadas nos testes de resistência osmótica atuais, eliminando-se também a incubação em solução isosmótica (300 mOsml). Esta foi substituída pela percentagem de NAR das amostras de morfologia espermática.

The consistency (scores from 1 to 5), tone with tonometry (mm), parenchyma echogenicity and heterogeneity with ultrasound (pixel) were assessed in 402 mature boars (18.5 months on average). Seasonal changes in the reproductive activity of the adult male viscacha (Lagostomus maximus maximus) were investigated during the annual reproductive cycle. Assays of heterologous in vitro binding between compatible gametes were used to evaluate the ability of viscacha spermatozoa to achieve primary binding during its annual reproductive cycle. This study examined the fine structures of epididymal spermatozoa on the lesser white-toothed shrew ( Crocidura suaveolens ), the Japanese white-toothed shrew ( C. dsinezumi ) and the big white-toothed shrew ( C. lasiura ) belonging to the subfamily Crocidurinae living in Korea . In the spermatozoa of C. suaveolens , the head has a large acrosome, a smooth inner acrosomal membrane and a wavy, finger-like, electron-dense apical body. The neck has a solid proximal centriole that is filled with electron-dense material. These results showed the spermatozoa of C. suaveolens possess the characteristics of both Crocidurinae and Soricinae. Determination of optimal glucose concentration for microcalorimetric metabolic evaluation of equine spermatozoa 52

The heat conduction microcalorimeter can be used to evaluate the metabolic rates of the sperm cell. Two ejaculates of four stallions were cooled to +5ºC and checked for sperm motility (bright field microscopy), viability (eosin 3%), functional membrane integrity (hyposmotic swelling test), and heat production (microcalorimetry). Glucose and sperm cell concentrations were determined in order to measure the heat outputs resulting from sperm metabolism. Sperm viability, membrane integrity and sperm motility did not differ among the different glucose concentrations tested. An in vitro zona-free hamster oocyte penetration test was utilized in 24 trials in order to evaluate the capacitation media used for ram sperm. A pool of fresh semen was collected from three crossed breed rams. Two semen drops were washed by centrifugation and incubated in high ionic strength treatment (HIS) or in a defined medium with HEPES, on heat ewe serum and heparin. After the incubation to promote capacitation, simplified triple-stain technique was used to evaluate the spontaneous acrosome reaction of the capacitated sperm. In a breeding farm, 129 mares were bred to 4 stallions and had an overall pregnancy rate of 76.7%. Stailions were submitted to semen examination for at least two times at the beginning of the breeding season and the percentage of morphologically normal spermatozoa were used for statistical analysis in relation to the percentage of pregnancy at the end of the breeding season. A regression analysis revealed that the percentage of pregnancy (y) can be estimated by the percentage of normal spermatozoa (x) in the ejaculate according to the following equation: y=52.38+0.44x; r2=97.8%; P<0.01. Resumo: Dez bodes da raca moxoto em idade de 24 a 26 meses foram submetidos durante dois anos a duas colheitas de ejaculados espermaticos semanais em vagina artificial. Os animais foram mantidos em pastagem nativa de caatinga raleada. Na avaliacao quantitativa do semen, encontrou-se maior volume na estacao chuvosa do que na seca. A concentracao de espermatozoides no ejaculado foi inversamente proporcional ao volume, portanto variando com as epocas do ano. Na estacao seca a porcentagem de espermatozoides vivos foi menor do que na estacao chuvosa.
