Degradation of methomyl by the novel bacterial strain Stenotrophomonas maltophilia M1 Electron. J. Biotechnol.
Mohamed,Mervat S.
The use of microorganisms in the degradation and detoxification of many toxic xenobiotics, especially pesticides, is an efficient tool for the decontamination of polluted sites in the environment. A novel bacterial strain (M1) was isolated from several water samples contaminated with methomyl which is capable of degrading methomyl pesticide (1000 ppm) in the presence of 0.05% glucose. These water samples were collected from different irrigation sites in Egypt where methomyl is heavily applied. The partial sequence of 16SrRNA gene of the isolate showed the highest similarity to Stenotrophomonas maltophilia. Restriction fragment patterns of isolated plasmid DNA showed that this strain harbours two different plasmids PMa (8Kb) and PMb (5Kb). PMb succeeded to...

Novel s-triazine-degrading bacteria isolated from agricultural soils of central Chile for herbicide bioremediation Electron. J. Biotechnol.
Hernández García,Marcela; Morgante,Verónica; Ávila Perez,Marcela; Villalobos Biaggini,Patricio; Miralles Noé,Pola; González Vergara,Myriam; Seeger Pfeiffer,Michael.
s-Triazine-degrading bacterial strains were isolated from long-term simazine-treated agricultural soils of central Chile. The number of culturable heterotrophic bacteria of these agricultural soils (7 x 10(6) CFU/g of dry soil) was not affected by simazine application on field. The simazine-degrading bacterial strains P51, P52 and C53 were isolated by enrichment in minimal medium using simazine as the sole nitrogen source. Resting cells of strains P51 and P52 degraded >80% of simazine within 48 hrs, whereas strain C53 was able to remove >60% of the herbicide. The atzA and atzD genes of the s-triazine upper and lower catabolic pathways were detected in strains P51 and C53, while only atzD gene was observed in strain P52. To compare the bacterial 16S...

Enhancement of biodegradation potential of catechol 1,2-dioxygenase through its immobilization in calcium alginate gel Electron. J. Biotechnol.
Guzik,Urszula; Hupert-Kocurek,Katarzyna; Marchlewicz,Ariel; Wojcieszynska,Danuta.
Background In biodegradation processes free enzymes often undergo deactivation. Thus, it is very important to obtain highly stable enzymes by different methods. Immobilization allows for successful stabilization of many multimeric enzymes by increasing the rigidity of the enzyme structure. This study aimed to evaluate some environmental factors that affect catechol 1,2-dioxygenase from Stenotrophomonas maltophilia KB2 immobilized in alginate hydrogel. The goal of the present work was to improve the functional stability of the enzyme by increasing its structural rigidity. Results Immobilization yield and expressed activity were 100% and 56%, respectively. Under the same storage conditions, the activity of the immobilized enzyme was still observed on the...

Biodegradation of the fungicide carbendazim by bacteria from Coriandrum sativum L. rhizosphere Biological Sciences
Santos, Jucileuza Conceição dos; Batista, Ieda Hortêncio; Barroso, Hiléia Santos; Lima, João Marcelo Silva; Costa Neto, Pedro de Queiroz; Ghelfi, Andrea; Pereira, José Odair.
The biocidal agrochemicals commonly used in agriculture can remain in the soil, affecting the environmental conditions and causing serious risks to health. Knowing that soil microorganisms, especially those from the rhizosphere, can degrade environmental xenobiotics, it was evaluated the potential of bacteria isolated from Coriandrum sativum L. rhizosphere to biodegrade carbendazim (MBC), a fungicide extensively used by agriculturists from rural farming communities in Manaus, Amazonas. Cultures carried out in medium containing carbendazim as a sole carbon source enabled the isolation of 80 bacteria, in the established conditions. Assays to determine degradation potential allowed the selection of the two elite isolates identified as Stenotrophomonas sp....

Selection of phytotoxin producing rhizobacteria Anais da ABC (AABC)
Carvalho,Daniel D.C.; Oliveira,Denilson F.; Campos,Vicente P.; Pasqual,Moacir.
In order to select phytotoxin producing rhizobacteria to control weed plants, twenty five bacterial strains previously isolated from the rhizospheres of various plants were grown in a liquid medium and, after cell removal by centrifugation, the liquid phases were freeze-dried and the products were extracted with ethyl acetate/methanol. The extracts were concentrated to dryness under vacuum and dissolved in water and sucrose solution to be submitted to in vitro assays of lettuce (Lactuca sativa L.) seed germination and wheat (Triticum aestivum L.) coleoptile growth. Although most samples affected coleoptile growth, only those from four strains reduced lettuce seed germination. Two strains of Bacillus cereus, one strain of B. pumilus and one of...

Isolation and characterization of a novel strain of Stenotrophomonas maltophilia possessing various dioxygenases for monocyclic hydrocarbon degradation BJM
Urszula,Guzik; Izabela,Greń; Danuta,Wojcieszyńska; Sylwia,Łabużek.
A Gram-negative bacterium, designated as strain KB2, was isolated from activated sludge and was found to utilize different aromatic substrates as sole carbon and energy source. On the basis of morphological and physiochemical characteristics and 16S rRNA gene sequence analysis, the isolated strain KB2 was identified as Stenotrophomonas maltophilia. Strain KB2 is from among different Stenotrophomonas maltophilia strains the first one described as exhibiting the activities of three types of dioxygenases depending on the structure of the inducer. The cells grown on benzoate and catechol showed mainly catechol 1,2- dioxygenase activity. The activity of 2,3-dioxygenase was detected after phenol induction. Protocatechuate 3,4-dioxygenase was found in crude cell...
