The in vitro culture of embryos is an important technique to enable the rescue of embryos from incompatible crosses. Studies analyzing the factors that affect the in vitro culture of zygotic embryos are scarce. This study evaluated the effect of the genotype (Capsicum baccatum and C. frutescens), the composition of the culture medium (MS and ½ MS medium with different concentrations of sucrose, IAA and GA3) and the stage of development (globular, cordiform, torpedo and cotyledonary) in the in vitro culture of immature embryos. Embryo germination was influenced primarily by the stage of development of the embryo and the composition of the culture medium. Regardless of the species, the most suitable culture medium for the germination of globular and... |

Asparaginase is an enzyme used in clinical treatments as a chemotherapeutic agent and in food technology to prevent acrylamide formation in fried and baked foods. Asparaginase is industrially produced by microorganisms, mainly gram-negative bacteria. Zymomonas mobilis is a Gram-negative bacterium that utilizes glucose, fructose and sucrose as carbon source and has been known for its efficiency in producing ethanol, sorbitol, levan, gluconic acid and has recently aroused interest for asparaginase production. Current assay optimizes the production of Z. mobilis asparaginase by continuous fermentation using response surface experimental design and methodology. The studied variables comprised sucrose, yeast extract and asparagine. Optimized condition obtained... |

ABSTRACT Maturation is a characteristic of sugarcane plant (Saccharum spp.) and even when grown under the same soil and climate conditions the varieties differ on the maturation curve. Thus, studies that allow establishing maturation curves of different sugarcane genotypes in the local soil and climate may indicate the proper harvesting period to ensure better quality of the raw material. This study aimed to analyze the levels of soluble sugars during the maturation phase and assess the technological and productivity indexes of four irrigated sugarcane genotypes in the region of Rio Largo, Alagoas. The experiment was conducted in randomized blocks in a 4 x 2 x 5 factorial: four genotypes (RB92579, RB98710, RB99395 and RB961003), two stem portions... |

ABSTRACT The purpose of this study was to determine the effects of the high consumption of sucrose on the levels of DNA damage in blood, hippocampus and bone marrow of rats. Male Wistar rats were treated for 4 months with sucrose (10% for 60 initial days and 34% for the following 60 days) in drinking water, and then, glycemia and glycated hemoglobin (A1C) were measured. Levels of DNA damage in blood and hippocampus were evaluated by the comet assay. The micronucleus test was used to evaluate chromosomal damages in the bone marrow. The sucrose treatment significantly increased (p<0.01) the serum glucose levels (~20%) and A1C (~60%). The level of primary DNA damage was significantly increased (p<0.05) in hippocampal cells (~60%) but not in peripheral... |

ABSTRACT We evaluated the influence of hesperidin and vitamin C (VitC) on glycemic parameters, lipid profile, and DNA damage in male Wistar rats treated with sucrose overload. Rats were divided into six experimental groups: I-water control; II-sucrose control; III-hesperidin control; IV-VitC control; V-co-treatment of sucrose plus hesperidin; VI-co-treatment of sucrose plus VitC. We measured the levels of triglycerides, total cholesterol, HDL-c, LDL-c, fasting glucose, and glycated hemoglobin (A1C). DNA damage was evaluated in blood and brain cells using the comet assay and the micronucleus test was used to evaluate chromosomal damages in the rat bone marrow. Co-treatment with VitC, but not with hesperidin, normalized the serum glucose. No effect of... |

The aim of the present study was to investigate the efficiency of the encapsulation-dehydration technique for cryopreservation of Cyrtopodium hastchbachii Pabst seeds. Immature seeds of this species were cryopreserved by an encapsulation-dehydration technique. Seeds of five immature pods, 120 days after pollination, were encapsulated in 3% calcium alginate matrix and pretreated in liquid medium supplemented with 0.08 M sucrose (24 h), 0.15 M sucrose (24 h), 0.25 M sucrose (48 h), 0.5 M sucrose (24 h) and 0.75 M sucrose (24 h) in shaker at 60 rpm. Alginate beads were dehydrated 5 h in silicagel and immersed in liquid nitrogen for 12 h. Cryopreserved beads were thawed at 30°C for 1 min, rehydrated using the same liquid mediums (0.75 M sucrose (24 h), 0.5 M... |

The in vitro synthesis of phytate was studied in common bean fruit explants. Different concentrations of sucrose; phosphorus (P); myo-inositol; abscisic acid (ABA); glutamine and methionine, were tested. Fixed concentrations of these compounds were tested at different periods (0, 3, 6 and 9 days). Variation in phytate coincided with different concentrations of sucrose, myo-inositol, P and ABA for the duration tested. These compounds caused an accumulation of phytate and were more effective in the presence of myo-inositol and P. The accumulation of P varied less than phytate for the different treatments tested in vitro. In conclusion, P, sucrose, ABA, and myo-inositol caused an increase in the phytate of bean seed, showing that it could be possible to alter... |

The aim of the present study was to evaluate the growth and development of apple rootstock shoots submitted to different photon flux densities (7, 14, 21 and 60 µmol m-2 s-1) and three culture media. The best photon flux for maximizing growth, number of shoots and leaves was 14 µmol m-2 s-1, which also resulted in the highest values of chloroplastic pigments. On the other hand, the highest photon flux (60 µmol m-2 s-1) decreased significantly these parameters. Medium with reduced sucrose concentration did not affect the analyzed parameters, enabling an efficient multiplication of the cultivar with only 1% of sucrose. |

Invertase, whether adsorbed on styrene-divinylbenzene copolymers or otherwise, was used for continuous sucrose hydrolysis using a cell-type membrane reactor (CTMR), coupled with an ultra (UF-100kDa), or a microfiltration (MF- pore diameter of 5 µm) membrane. In all tests, the pH (5.5), temperature (30 ºC), reaction volume (10 mL) and agitation (100 rpm) were set constant; whereas, variable parameters were: feeding rate (0.4, 0.8 and 1.6 h-1), inlet sucrose concentration (2.5, 6.5, 50 and 100 mM) and enzyme/resin ratio (1.64 mg or 3.28 mg of protein per 25, 50 or 100 mg of resin). The best result (yield of 100%, steady-state duration over 20h and specific reaction rate over 243 x 10-3 mmol/h.mE) was obtained when insoluble invertase (1.64 mg protein/100 mg... |

Gas exchanges, sucrose, starch and reducing sugar contents were analyzed in leaves of orange trees with and without Citrus Variegated Chlorosis (CVC), a disease caused by the bacteria Xylella fastidiosa. Healthy plants (HP) and diseased plants (DP) were evaluated 20 and 26 months after bacterial inoculation, in October 1998 and April 1999, respectively. On the first evaluation date, CO2 assimilation rates for HP were greater than DP in the early morning and midday, but on the second date they were greater in HP all along the day. On the first date, there was an increase in the sucrose content of DP at 15:00 h and 17:00 h in relation to HP. On the second evaluation date, DP presented greater reducing sugar content only in the early morning while sucrose... |
