Summer mortality of adults and juveniles has been reported in the Pacific oyster, Crassostrea gigas, for many years and in several countries. The French multidisciplinary program "Morest" aims to investigate the causes of the summer mortality in Crassostrea gigas. Within this program, we designed multi-site field experiments to assess to what extent genetic variability exists for summer mortality in French populations of C. gigas and to determine whether selective breeding could improve survival.

Summer mortality associated with juveniles of the oyster Crassostrea gigas is the result of a complex interaction between the host, pathogens and environment. Divergent selection based on summer survival have been applied to produce families with resistant and susceptible progeny. Here, we describe the use of suppression subtractive hybridization to generate 150 clones that were differentially regulated between resistant and susceptible F2 progeny. Summer mortality has been reported in the Pacific oyster, Crassostrea gigas, for many years in Japan, US and Europe. It affects both adults and juveniles and the causes of this phenomenon are complex. Environmental, physiological or pathological putative causes were studied all over the world. Recently, the MOREST project ((MORtalitEStivale) a multidisciplinary project (2001-2005) focused on his problem led to a better understanding of this phenomenon in France. The results showed in these geographical conditions, the main interactions between the oysters, their environment and certain pathogens.

The purpose of this study is to investigate summer mortality of the cupped oyster, Crassostrea gigas, in relation to culture practices in the traditional oyster production region of Marennes-Oleron (France). Four oyster rearing conditions, varying culture location ("on-" or "off-bottom"), and site depth (65% to 80% daily immersion termed "deep" or 45% to 65%, termed "shallow") were studied to compare biologic performance and maturation status of oysters, in relation to sediment and water column parameters. The most severe mortality occurred in June to July in "on-bottom" reared oysters (25%), as compared with 10% mortality in "off-bottom" cultured oysters. Summer mortality of selected juvenile pacific oyster Crassostrea gigas under laboratory conditions and in comparison with field performance ArchiMer

This study reports mortality under laboratory conditions in unselected controls and two lines of juvenile Pacific oysters Crassostrea gigas, previously selected for their high or low survival in the field during the summer period. Oysters were also deployed in field conditions and mortality between both conditions was then compared. In the laboratory, mortality was observed in all experiments and it always lasted for a week indicating that mortality under laboratory conditions was a short-term event. In order to study genetic variability for survival during summer mortality events in the oyster Crassostrea gigas, a first generation (G1) was bred in 2001. 17 G1 half-sib families (HSF) were obtained ; each HSF was reared in three sites along the French coast during the summer period. Significant differences in survival were observed among HSF indicating a genetic basis for survival. Recherche et identification de bactéries pathogènes au sein d'une écloserie nurserie d'huître creuse, Crassostrea gigas, en relation avec la mortalité estivale du naissain ArchiMer

Summer mortality has been observed on oyster spat Crassostrea gigas since 1991 on french coasts. In our study, we deseribed symptoms observed during the mortality, especially adductor muscle weakness and we isolated strains on Zobell medium and TCBS from moribund spat. Strains isolated during a high mortality (60 %) were selected by experimental pathology on spat. Inoculation by bath led us to select a pathogenic strain (TNEMF6) for oyster spat. Phenotypic identification, phylogenetic study on 16S rDNA and quantitative DNA/DNA hybridisation showed that this strain belongs to Vibrio splendidus biovar II.
