Gene expression of a putative glutathione S-transferase is responsive to abiotic stress in embryogenic cell cultures of Cyclamen persicum Electron. J. Biotechnol.
Hoenemann,Claudia; Ambold,Juliane; Hohe,Annette.
Background: Cyclamen persicum is an economically important ornamental crop that is propagated exclusively through seeds as vegetative propagation using cuttings is not possible. Therefore a micropropagation method through somatic embryogenesis is of high interest; however the method suffers from low reliability concerning quality and quantity of the produced plantlets. A crucial step of the protocol is the removal of plant growth regulators (PGRs) that triggers embryo development. In order to get a better insight in this crucial step of the propagation process, a gene expression analysis has been set up using five different genes of glutathione S-transferases (GST) as these are known to be auxin responsive as well as stress reactive. Results: One out of...

Methyl jasmonate induced overproduction of eleutherosides in somatic embryos of Eleutherococcus senticosus cultured in bioreactors Electron. J. Biotechnol.
Shohael,Abdullah Mohammad; Murthy,Hosakatte Niranjana; Hahn,Eun-Joo; Paek,Kee-Yoeup.
This study was concentrated on the production of eleutherosides and chlorogenic acid in embryogenic suspension cultures of Eleutherococcus senticosus by exposing them to different concentrations (50-400 µM) of methyl jasmonate (MJ) during the culture period. In the bioreactor cultures, eleutheroside content increased significantly by elicitation of MJ, however, the fresh weight, dry weight and growth ratio of embryos was strongly inhibited by increasing MJ concentrations. The highest total eleutheroside (7.3 fold increment) and chlorogenic acid (3.9 fold increment) yield was obtained with 200 µM MJ treatment. There was 1.4, 3.4 and 14.9 fold increase in the eleutheroside B, E, and E1 production respectively with such elicitation treatment. These results...

Isolation of Breast Cancer Stem Cell from MDA-MB231 Cell Line Using Vincristine International Journal of Morphology
Ghanbari,Tara; Azadbakht,Mehri; Vesi-Raygani,Asad; Khazaei,Mozafar.
Cancer has been considered as a stem cell disease. Suspension culture combined with anti-cancer drugs has recently been proposed for isolation of cancer stem cells (CSCs). In the current study, Vincristine as an anti-cancer drug combined with suspension culture was used for isolation and purification of CSCs from human breast cancer cell line (MDA-MB231). The cells were treated with different concentrations of vincristine (0, 2, 4, 6 and 8 ng/ml). Stem cells were identified with the expression of OCT4, nanog, SOX2 and nucleostemin genes by RT-PCR. Mammosphere forming unit was measured upon suspension culture containing EGF, bFGF, LIF, B27, insulin and BSA. The isolated mammospheres were investigated for CD44 expression. Results showed that 4 ng/ml of...
