Santos,Terezinha de Jesus Teixeira; Costa,Carlos Maurício de Castro; Goubau,Patrick; Vandamme,Anne-Mieke; Desmyter,Jan; Van Dooren,Sonia; Mota,Rosa M. S.; Costa,Francine Bovy de Castro; Oliveira,Ana C. S.; Gomes,Vânia Barreto A.F.; Carneiro-Proietti,Anna B.; Bruin,Veralice Meireles Sales de; Sousa,Francisca C. F. de; Oriá,Reinaldo Barreto.
How to handle Western blot (WB) seroindeterminate individuals for Human T-lymphotropic Virus 1/2 (HTLV-1/2) constitutes a challenge for blood banks and fam ilies. We made a cross-sectional study of 191 enzyme linked immunoassay (EIA) reactive individuals from the hematological center (HEMOCE) of Fortaleza (Brazil), examining their serological (WB) and molecular (PCR) diagnosis, and demographic profiles, as well as a possible association of their condition with other infectious pathologies and risk factors. Ethical institutional approval and personal consent were obtained. Out of 191 EIA reactive individuals, 118 were WB seroindeterminate and 73 were seropositive for HTLV-1/2. In the PCR analysis of 41 WB seroindeterminate individuals, 9 (22%) were positive...

Self-reported syphilis and associated factors among Brazilian young adults: findings from a nationwide survey BJID
Kops,Natália Luiza; Bessel,Marina; Hohenberger,Glaucia F.; Benzaken,Adele; Wendland,Eliana.
Abstract The aim of this study was to evaluate self-reported syphilis and associated factors in sexually active young adults (16-25 years old) in the Public Health System in Brazil. This was a cross-sectional study with 8071 participants recruited from 119 primary care units. Of these, 224 (2.86%, 95% CI 2.29-3.43%) reported having the disease. Age, lower socio-economic class, being a smoker, not using a condom at first sexual intercourse, and ever had a same-sex sexual experience were associated with syphilis. The results reinforce the importance of implementing strategies focused on socio-economic class and early sexual education that encourage condom use from the beginning of sexual activity.

Profile of Anti-Tp47 antibodies in patients with positive serology for syphilis analized by western blot BJID
Miranda,Ana Paula Félix de; Sato,Neuza Satomi.
In Brazil, syphilis is still a great problem of public health. Serological test is essential for syphilis diagnosis and the current trend is the use of recombinant antigen in the treponemal tests, due to its confirmed higher sensibility and specificity. The purpose of the present study was to analyze the profile of anti-Tp47 antibodies in patients with positive serology for syphilis. One hundred positive sera samples were analyzed by Western Blot (WB) technique, using the recombinant antigen (rTp47). Ten of them did not present antibodies against the fraction rTp47, the results were confirmed by WB using native T. pallidum antigen. All ten samples had antibodies against the fractions Tp17 and Tp15 and presented low reactivity in VDRL, negative results or...

A syphilis co-infection study in human papilloma virus patients attended in the sexually transmitted infection ambulatory clinic, Santa Casa de Misericórdia Hospital, Rio de Janeiro, Brazil BJID
Souza,Lúcia Maria de Sena; Miller,William Meihack; Nery,José Augusto da Costa; Andrade,Arnaldo Feitosa Braga de; Asensi,Marise Dutra.
Despite the prevalence of syphilis worldwide, little is known about its manifestations when associated with other Sexually Transmitted Infections (STI), specifically the Human Papilloma Virus (HPV). Current epidemiological studies show that there is a high incidence of both diseases in ambulatory clinics all over Brazil. This study aims to estimate the incidence of syphilis - HPV co-infections, among patients from the STI ambulatory clinic at the Santa Casa da Misericórdia Hospital, Rio de Janeiro, Brazil. Two-hundred and seven patients were seen in the clinic between March and December 2005, of which 113 (54.6%) sought care for an HPV infection. Blood samples were taken from all patients to check syphilis serology using the flocculation and the...

Syphilis and HIV-1 among parturient women in Salvador, Brazil: low prevalence of syphilis and high rate of loss to follow-up in HIV-infected women BJID
Nóbrega,Isabella; Dantas,Paula; Rocha,Priscila; Rios,Isabela; Abraão,Marcos; M. Netto,Eduardo; Brites,Carlos.
BACKGROUND: The occurrence of syphilis and HIV-1 infections during pregnancy are major risks to the fetus due to mother-to-child transmission (MTCT). OBJECTIVES: To determine peripartum seroprevalence and risk factors of syphilis and HIV-1 infection among pregnant women in Salvador, Brazil, and the rate of HIV-1 MTCT. METHODS:Cross-sectional study of pregnant women who were admitted for delivery in a reference maternity hospital between May 2008 and March 2009 was conducted. Women were screened for HIV-1 infection and syphilis, and interviewed regarding demographic, behavioral and obstetric data. Newborns to HIV-infected mothers were tested by b-DNA and DNA-PCR to detect HIV-1. RESULTS: A total 3300/8516 women were evaluated. Mean age was 25.8 ± 7.3 years....

Recombinant antigen-based immuno-slot blot method for serodiagnosis of syphilis BJMBR
Sato,N.S.; Suzuki,T.; Ueda,T.; Watanabe,K.; Hirata,R.D.C.; Hirata,M.H..
Three recombinant antigens of Treponema pallidum Nichols strain were fused with GST, cloned and expressed in Escherichia coli, resulting in high levels of GST-rTp47 and GST-rTp17 expression, and supplementation with arginine tRNA for the AGR codon was needed to obtain GST-rTp15 overexpression. Purified fusion protein yields were 1.9, 1.7 and 5.3 mg/l of cell culture for GST-rTp47, GST-rTp17 and GST-rTp15, respectively. The identities of the antigens obtained were confirmed by automated DNA sequencing using ABI Prism 310 and peptide mapping by Finningan LC/MS. These recombinant antigens were evaluated by immuno-slot blot techniques applied to 137 serum samples from patients with a clinical and laboratory diagnosis of syphilis (61 samples), from healthy...

The vulnerability of Brazilian female prisoners to HIV infection BJMBR
Strazza,L.; Azevedo,R.S.; Carvalho,H.B.; Massad,E..
The purpose of the present study was to determine the vulnerability of women in prison to HIV infection. The study was carried out from August to October 2000 in a São Paulo State Penitentiary, where 299 female prisoners were serving time. We interviewed and obtained a blood sample from 290 females who agreed to enter the study. Sera were tested for the presence of antibodies to HIV, hepatitis C virus (HCV) and syphilis and the odds ratio (OR) was calculated for variables related to HIV positivity on the basis of a questionnaire. The overall prevalence data were: 13.9% for HIV (37 of 267), 22.8% for syphilis (66 of 290), and 16.2% for HCV (47 of 290). Sexual partnership variables were significantly related to HIV infection. These included HIV-positive...

Origin and pathogenesis of antiphospholipid antibodies BJMBR
Celli,C.M.; Gharavi,A.E..
Antiphospholipid antibodies (aPL) are a heterogeneous group of antibodies that are detected in the serum of patients with a variety of conditions, including autoimmune (systemic lupus erythematosus), infectious (syphilis, AIDS) and lymphoproliferative disorders (paraproteinemia, myeloma, lymphocytic leukemias). Thrombosis, thrombocytopenia, recurrent fetal loss and other clinical complications are currently associated with a subgroup of aPL designating the antiphospholipid syndrome. In contrast, aPL from patients with infectious disorders are not associated with any clinical manifestation. These findings led to increased interest in the origin and pathogenesis of aPL. Here we present the clinical features of the antiphospholipid syndrome and review the...
