Overview of tigecycline and its role in the era of antibiotic resistance BJID
Rossi,Flávia; Andreazzi,Denise.
The increasing antimicrobial resistance found in the many clinically important species of bacteria that commonly cause serious and life-threatening diseases presents a difficult challenge for clinicians, especially when an appropriate initial therapy must be chosen. New antibiotics are urgently needed to address the formidable issues associated with infections caused by vancomycin-resistant enterococci (VRE), methicillin-resistant Staphylococcus aureus (MRSA), penicillin-resistant Streptococcus pneumoniae and multidrug-resistant Gram-negative bacteria. The need for new antibiotics that effectively resist antimicrobial mechanisms of resistance has become paramount. Tigecycline is a new antimicrobial agent; it is the first in a new class of antibiotics, the...

Changes in Staphylococcus aureus susceptibility across Latin America between 2004 and 2010 BJID
Garza-González,Elvira; Dowzicky,Michael Joseph.
The Tigecycline Evaluation and Surveillance Trial is a global surveillance study monitoring the efficacy of tigecycline and comparators against clinically important pathogens. Between 2004 and 2010, 3126 isolates of Staphylococcus aureus were collected from 66 centers in 13 countries in Latin America; of these, 1467 (46.9%) were resistant to methicillin. The main contributors of S. aureus isolates were Mexico (n = 846), Argentina (n = 740), and Colombia (n = 445). The methicillin-resistant S. aureus rate was greater than 50% in five countries, the highest reported in Puerto Rico (73.9%). Methicillin-resistant S. aureus rates across Latin America ranged from 40.1% to 50.6% over the study period. All S. aureus isolates were susceptible to linezolid and...

In vitro activity of tigecycline, a new glycylcycline, tested against 1,326 clinical bacterial strains isolated from Latin America BJID
Gales,Ana C.; Jones,Ronald N.; Andrade,Soraya S.; Pereira,Andrea S.; Sader,Hélio S..
The in vitro activity of tigecycline (former GAR-936), a new semisynthetic tetracycline, was evaluated in comparison with tetracycline and other antimicrobial agents. MATERIAL AND METHODS: A total of 1,326 contemporary clinical isolates collected from the Latin American region were collected in 2000-2002 period and tested with microdilution broth according to the CLSI guidelines. The bacterial pathogens evaluated included Staphylococcus aureus (505), Streptococcus pneumoniae (269), coagulase-negative staphylococci (CoNS; 227), Haemophilus influenzae (129), Enterococcus spp. (80), Moraxella catarrhalis (54), beta-haemolytic streptococci (28), viridans group streptococci (26), and Neisseria meningitidis (8) RESULTS:Tigecycline demonstrated excellent activity...

Indications of a new antibiotic in clinical practice: results of the tigecycline initial use registry BJID
Curcio,Daniel; Fernández,Francisco; Cané,Alejandro; Barcelona,Laura; Stamboulian,Daniel.
Tigecycline is the first of a new class of antibiotics named glycylcyclines and it was approved for the treatment of complicated intra-abdominal infections and complicated skin and skin structure infections. Notwithstanding this, tigecycline's pharmacological and microbiological profile which includes multidrug-resistant pathogens encourages physicians' use of the drug in other infections. We analyzed, during the first months after its launch, the tigecycline prescriptions for 113 patients in 12 institutions. Twenty-five patients (22%) received tigecycline for approved indications, and 88 (78%) for "off label" indications (56% with scientific support and 22% with limited or without any scientific support). The most frequent "off label" use was ventilator...

Rates of antimicrobial resistance in latin america (2004-2007) and in vitro activity of the glycylcycline tigecycline and of other antibiotics BJID
Rossi,Flávia; García,Patricia; Ronzon,Bernardo; Curcio,Daniel; Dowzicky,Michael J.
As a part of the Tigecycline Evaluation and Surveillance Trial (T.E.S.T.), Gram-positive and Gram-negative bacterial isolates were collected from 33 centers in Latin America (centers in Argentina, Brazil, Chile, Colombia, Guatemala, Honduras, Jamaica, Mexico, Panama, Puerto Rico, and Venezuela) from January 2004 to September 2007. Argentina and Mexico were the greatest contributors of isolates to this study. Susceptibilities were determined according to Clinical Laboratory Standards Institute guidelines. Resistance levels were high for most key organisms across Latin America: 48.3% of Staphylococcus aureus isolates were methicillin-resistant while 21.4% of Acinetobacter spp. isolates were imipenem-resistant. Extended-spectrum β-lactamase were reported in...
