Eggplant (Solanum melongena L.): tissue culture, genetic transformation and use as an alternative model plant Acta Botanica
Magioli,Claudia; Mansur,Elisabeth.
Eggplant is an agronomically important non-tuberous solanaceous crop grown primarily for its large oval fruit. In popular medicine, eggplant is indicated for the treatment of several diseases, including diabetes, arthritis, asthma and bronchitis. Eggplant is susceptible to a number of diseases and pests capable of causing serious crop losses. This problem has been addressed by hybridizing eggplant with wild resistant Solanum species, which present a wide genetic diversity and are source of useful agronomic traits. The application of in vitro methodologies to eggplant has resulted in considerable success. Eggplant tissues present a high morphogenetic potential that is useful for developmental studies as well as for establishing biotechnological approaches...

Interaction between sucrose and pH during in vitro culture of Nephrolepis biserrata (Sw.) Schott (Pteridophyta) Acta Botanica
Ambrósio,Sandra Tereza; Melo,Natoniel Franklin de.
The present work reports the effect of different pH and sucrose concentrations on in vitro propagation of Nephrolepis biserrata. Fronds aseptically obtained from stolon segment culture were cultivated in MS semi-solid medium supplemented with 0, 15, 30, 45 and 60g.L-1 sucrose and pH adjusted to 3, 5, 7 and 9, in a factorial design. Frond number and length, pinnae number, raquis length and diameter, fresh and dry matter weight were measured. Inhibition of shoot and leaf regeneration was observed in all the pH treatments in the absence of sucrose. On the other hand, when sucrose was added to the medium, the shoot number increased, reaching the maximum average values of 51.25 and 38.25 shoot per explant at pH 5 and 7, respectively. Sucrose concentrations from...

Anacardium othonianum Rizz. is a medicinal plant species that is native to the Brazilian savannah. Adult plants are different from other genus members in this ecosystem due to their size, and efforts to locate the plants may lead to their extraction from the savannah and, frequently, plant death. Micropropagation has played a significant role in the propagation and preservation of specimens of several plant species; therefore, this study evaluated the effect of various salt concentrations and culture medium volumes on the in vitro cultivation of A. othonianum Rizz. Trial (I) evaluated two culture media (MS and WPM) and three salt concentrations (100, 50 and 25%) in a completely randomized design. Trial (II) evaluated two culture media and salt...

The objective of this study was to evaluate the physiological quality of babassu (Orbignya phalerata Mart.) embryos as a function of desiccation. The fruits were placed in a convection oven at 37±2°C for 0, 3, 6, 9, or 12 days. After these periods, the moisture contents of the fruits and seeds were determined. A reduction in the water content occurred more rapidly in the fruits than in the seeds. The decrease in seed moisture content from 12.59% (on the basis of fresh weight) to 4.56% did not affect the percentage of germinated zygotic embryos; however, we did observe a decline in the vigor as the seed moisture content decreased. Embryos with a 4.56% moisture content developed into seedlings of shorter lengths after 30, 60, and 90 days of in vitro growth....

Hydrogen peroxide in micropropagation of Lilium: A comparison with a traditional methodology Biocell
Curvetto,Néstor; Marinangeli,Pablo; Mockel,Gabriela.
The micropropagation of Lilium longiflorum requires adequate equipment which may not be afforded by small laboratories or producers. In this work we compared traditional methodology with a protocol that included easily available elements to sterilize materials and culture media, together with addition of hydrogen peroxide (H2O2) into the nutrient media as chemical sterilizer. A series of H2O2 concentrations (0.005, 0.010, 0.015 and 0.020% p/v) was used to control contamination during in vitro establishment and subsequent cultivation; the explant organogenic response was also examined and compared to the traditional micropropagation technique. The level of culture contamination was within acceptable limits in all treatments, though it was higher in the H2O2...

Ventricular Myosin Light Chain-2 Gene Expression in Developing Heart of Chicken Embryos Biol. Res.
GHATPANDE,SATISH; SHAFIQ,SAIYID; SIDDIQUI,M.A.Q..
Recent gene knock-out studies in mice have suggested that ventricular myosin light chain-2 (vMLC2) has a role in the regulation of cardiogenic development and that perturbation in expression of vMLC2 is linked to the onset of dilated cardiomyopathy. In an attempt to develop an avian model for such studies, we examined the expression pattern of vMLC2 in chicken embryos at various stages and analyzed the effect of antisense oligonucleotide-mediated interference of vMLC2 function in cultures of whole embryos. Our results showed vMLC2 to be a specific marker for ventricular chamber throughout chicken embryonic development and antisense vMLC2 treatment of primitive streak stage (stage 4) embryos to produce pronounced dilation of heart tube with severe...

Altered calcium currents in cultured sensory neurons of normal and trisomy 16 mouse fetuses, an animal model for human trisomy 21 (Down Syndrome) Biol. Res.
CAVIEDES,PABLO; CAVIEDES,RAÚL; RAPOPORT,STANLEY I.
Down syndrome is determined by the presence of an extra copy of autosome 21 and is expressed by multiple abnormalities, with mental retardation being the most striking feature. The condition results in altered electrical membrane properties of fetal dorsal root ganglia (DRG) neurons, as in the trisomy 16 fetal mouse, an animal model of the human condition. Cultured trisomic DRG neurons from human and mouse fetuses present faster rates of depolarization and repolarization in the action potential compared to normal controls and a shorter spike duration. Also, trisomy 16 brain and spinal cord tissue exhibit reduced acetylcholine secretion. Therefore, we decided to study Ca2+ currents in cultured DRG neurons from trisomy 16 and age-matched control mice, using...

Biochemical and morphological responses to abiotc elicitor chitin in suspension-cultured sugarcane cells BABT
Gallão,Maria Izabel; Cortelazzo,Ângelo Luiz; Fevereiro,Manuel Pedro Salema; Brito,Edy Sousa de.
Cells of Saccharum officinarum submitted to hydrolyzated chitin for 1 to 8h produced phenolic compounds. These alterations were observed through cytochemical methods using Toluidine Blue and Phloroglucinol/HCl. After 4 h, besides cell wall change, there was a change in nuclear pattern of chitin treated cells. There was a 96% increase in nuclear area in 6 h chitin treated material, as observed by Feulgen reaction. The treated cells showed chromatin compacted regions and a degeneration process of nucleoli. In the outer areas of cell wall, there was a polysaccharide desagregation, confirming results obtained for different plants with the use of other elicitors. Peroxidase activity was maximal after 4 h and decreased progressively. PAL activity started to...
