Toxoplasma gondii multiplies within parasitophorous vacuole that is not recognized by the primary no oxidative defense of host cells, mainly represented by the fusion with acidic organelles. Recent studies have already shown that hydroxyurea arrested the intracellular parasites leading to its destruction. In the present work we investigated the cellular mechanism involved in the destruction of intracellular Toxoplasma gondii. Fluorescent vital stains were used in order to observe possible acidification of parasitophorous vacuole-containing Toxoplasma gondii in presence of hydroxyurea. Vero cells infected with tachyzoites were treated with hydroxyurea for 12, 24 or 48 hours. Fluorescence, indicative of acidification, was observed in the parasitophorous... |

Toxoplasmosis is the most common cause of infectious uveitis in Brazil, with a higher frequency in the South of the country. We have collected samples from porcine tongue and diaphragm obtained in both large and small abattoirs and used molecular biological technique to determine the prevalence of infection and RFLP analysis to type the parasites. Seventeen out of 50 (34%) samples from the diaphragm and 33 out of 50 (66%) samples from the tongue demonstrated a positive PCR reaction for T. gondii and restriction analysis of four of the positive samples revealed that all had a type I genotype at SAG2. However, when other unlinked loci were analyzed, these strains had a type III genotype at markers BTUB, SAG3, and GRA6. One of the strains (8T) had a type II... |

Immunity to Toxoplasma gondii was studied in pigs, after vaccination with T. gondii antigens incorporated into immunostimulating complexes. Nine pigs (group 1 - G1) were inoculated subcutaneously with T. gondii iscoms (LIV-5 sample) and three doses were given at 21 and 13 day-intervals. The results were compared in other three groups of nine pigs each: animals in group 2 (G2) were immunized with the LIV-5 antigens without Quil A, animals in group 3 (G3) were inoculated with tachyzoites of RH T. gondii isolate, and animals in group 4 (G4) received no vaccination. Four animals were neither vaccinated nor challenged with T. gondii (group 5 - G5). Thirty days after vaccination, pigs were challenged orally with 5´10(4) oocysts at AS-28 T. gondii isolate.... |

ABSTRACT Toxoplasmosis is a widespread zoonosis that can infect warm-blooded animals including birds and humans, and chickens are considered to be indicators of environmental contamination. In Brazil, Toxoplasma gondii has a non-clonal population structure composed of three lineages (I, II, and III), presenting high recombination, and resulting in wide genetic diversity. This study aimed to genetically characterize T. gondii isolates from naturally infected chickens (Gallus domesticus) in Santa Catarina state, southern Brazil region. Sera from 133 free-range chickens were analyzed by an immunofluorescence assay (IFA) to detect IgG antibodies against T. gondii. Brain and heart from 30 positive animals, based on IFA (≥ 1:64), were used to isolate the... |

The distribution of T. gondii in commercial cuts of pork (ham, tenderloin, spareribs and arm picnic) by PCR and bioassay from experimentally infected pigs, was evaluated. Eighteen mixed breed pigs were divided into two groups (G). The G1 animals (n=10) were infected with 4 x10(4) oocysts of the T. gondii VEG strain and the G2 animals (n=8) were used as control. Pigs of both groups were slaughtered at 59th day after infection, and meat samples were collected for bioassay and PCR. All animals from G1 were positive by at least one or both tests, and all control animals were negative. T. gondii was identified in pork by mouse bioassay and PCR in 27/40 (67.5%) and in 9/40 (22.5%) of the evaluated samples, respectively. There were no statistical differences in... |

The prevalence of T. gondii infection was investigated in free-range chickens for human consumption in the county of Rio Bonito, Rio de Janeiro, Brazil. Thus, a search for antibodies to T. gondii by indirect fluorescent antibody test was done in the serum of 220 chickens from twenty two farms. Titres equal or greater than 16 were considered positive. The prevalence of toxoplasmic infection found in chickens was 27.6% (61/220). There were positive cases in eighteen (77.2%) farms. The more frequent titre was 16 (12.5%) and the highest was 4096 (0.4%). The high prevalence of T. gondii in chickens and the high percentage of farms with positive cases indicate environmental contamination in the studied area showing the possibility of human and animal infection. |

Three-hundred and eighty-five serum samples were taken from dairy cows on 90 farms in 12 counties from the northern region of the Paraná State, Brazil. The samples were analyzed by IFAT for the detection of anti-Neospora caninum and anti-Toxoplasma gondii IgG antibodies. Forty-five (12%) samples were seropositive to N. caninum, while 102 (26%) samples were seropositive to T. gondii. Only four animals were seropositive to both coccidia. No significant difference was observed between the N. caninum serology and any of the variables studied, such as dairy cattle management, milk production, reproductive problems, feeding, and presence of dogs, cats and rodents. These data suggest that neosporosis is present among dairy cattle in the studied geographic region... |

Toxoplasmosis is a zoonosis that has been the subject of study in Brazil and worldwide. The dogs are sentinels for the infection and can carry Toxoplasma gondii in the environment. Seroepidemiological surveys of these animals are an important tool in the surveillance and control of the disease and inform decision-making in health programs. In this study the performance of the Modified Agglutination Test (MAT) in the serodiagnosis of canine toxoplasmosis is evaluated and compared to the indirect immunofluorescent-antibody test (IFAT). A sample of 157 dog sera from the county of Monte Negro, Rondônia, with 76.40% positive reactions for Toxoplasma gondii (IFAT =16) was analyzed using the MAT (=25), presenting sensitivity of 85.00% (Confidence Interval 95.00%:... |
