Production of transgenic goat (Capra hircus) with human Granulocyte Colony Stimulating Factor (hG-CSF) gene in Brazil 42
Freitas,Vicente J.F.; Serova,Irina A.; Andreeva,Lyudmila E.; Dvoryanchikov,Guennadi A.; Lopes-Jr.,Edilson S.; Teixeira,Dárcio I.A.; Dias,Luciene P.B.; Avelar,Suely R.G.; Moura,Raylene R.; Melo,Luciana M.; Pereira,Alexsandra F.; Cajazeiras,João B.; Andrade,Maria L.L.; Almeida,Karlliely C.; Sousa,Francisco C.; Carvalho,Antonio C.C.; Serov,Oleg L..
In order to produce transgenic goats with hG-CSF, a total of 24 adult Saanen and 48 adult undefined breed goats were used as donors and recipients, respectively. Donors were estrus-synchronized with vaginal sponges and superovulated by a treatment with 200 mg FSH given twice daily in decreasing doses over 3 days starting 48 h before sponge removal. Ovulation was induced by injecting 100µg GnRH 36 h after sponge removal. The recipients also received an estrus synchronization treatment. Donors were mated with fertile Saanen bucks and, approximately 72 h after sponge removal, zygotes were recovered surgically by flushing oviducts. The recovered zygotes were briefly centrifuged to a reliable visualization of the pronuclei. The DNA construct containing hG-CSF...

Production of recombinant proteins in milk of transgenic and non-transgenic goats 52
Moura,Raylene Ramos; Melo,Luciana Magalhães; Freitas,Vicente José de Figueirêdo.
Among all the transgenic mammalians produced so far, goats have represented an excellent model of transgenesis when considering the factors such as the market demand for protein, volume of milk produced per lactation and reproductive rate. Various recombinant proteins have been obtained from the transgenic and non-transgenic goats, and among these, human antithrombin, produced by the transgenic goats, was the first recombinant protein of animal origin to be released as a drug for the clinical use in humans. This review reports the aspects inherent to the production of recombinant proteins in the goats, from the production of the animal bioreactors up to the expression of these proteins in their milk.

Detection of transgenic events in maize using immunochromatographic strip test and conventional PCR 64
Cantelmo,Narjara Fonseca; Von Pinho,Édila Vilela de Resende; Von Pinho,Renzo Garcia; Von Pinho,Iolanda Vilela; Nascimento,Micaela Sandim.
With the growth in the transgenic market, fast and economically viable methodologies are necessary for undertaking transgene detection tests, both for identification of contamination in seeds and in grain. Seeds from commercial conventional GNZ 2004, and transgenic VT-Pro (MON89034), Roundup Ready (NK603) and Herculex (TC1507) maize cultivars were used. In order to simulate different levels of contamination, the transgenic seeds were mixed with conventional seeds at levels of 0.2%, 0.4%, 1.0% and 1.6% for VT-Pro, and 0.2%, 0.5%, 0.8% and 1.2% for Roundup Ready and Herculex. The lateral flow membrane strip test was performed in the whole seed, endosperm and embryo. For evaluation of the specificity of the technique in detection of the TC1507 event by means...

Improving the production of transgenic fish germlines: in vivo evaluation of mosaicism in zebrafish (Danio rerio) using a green fluorescent protein (GFP) and growth hormone cDNA transgene co-injection strategy 74
Figueiredo,Márcio de Azevedo; Lanes,Carlos Frederico Ceccon; Almeida,Daniela Volcan; Marins,Luis Fernando.
In fish, microinjection is the method most frequently used for gene transfer. However, due to delayed transgene integration this technique almost invariably produces mosaic individuals and if the gene is not integrated into germ cells its transmission to descendants is difficult or impossible. We evaluated the degree of in vivo mosaicism using a strategy where a reporter transgene is co-injected with a transgene of interest so that potential germline founders can be easily identified. Transgenic zebrafish (Danio rerio) were produced using two transgenes, both comprised of the carp beta-actin promoter driving the expression of either the green fluorescent protein (GFP) reporter gene or the growth hormone cDNA from the marine silverside fish Odonthestes...

Transgenic mosquitoes for malaria control: progresses and challenges 82
Moreira,Luciano A.; Jacobs-Lorena,Marcelo.
Malaria kills millions of people every year and the current strategies to control the disease, such as insecticides and drugs have not been completely efficient. Because of that, novel means to fight against malaria are of utmost importance. Advances in the study of the mosquito vector and its interactions with the malaria parasite made scientists think that it is possible to genetically manipulate the mosquitoes to make them inefficient vectors. Here we review the advances on the introduction of foreign genes into the mosquito germ line, the characterization of tissue-specific promoters, the identification of gene products that block development of the parasite in the mosquito, and we discuss the recent generation of transgenic mosquitoes impaired for...
