Tracing key genes associated with the Pinctada margaritifera albino phenotype from juvenile to cultured pearl harvest stages using multiple whole transcriptome sequencing ArchiMer

Background Albino mutations are commonly observed in the animal kingdom, including in bivalves. In the black-lipped pearl oyster Pinctada margaritifera, albino specimens are characterized by total or partial absence of colouration resulting in typical white shell phenotype expression. The relationship of shell colour with resulting cultured pearl colour is of great economic interest in P. margaritifera, on which a pearl industry is based. Hence, the albino phenotype provides a useful way to examine the molecular mechanisms underlying pigmentation. A new enzymatic process for the treatment of phenolic pollutants BABT

This work aimed to develop a new economic enzymatic process to treat the phenolic pollutants using pure tyrosinase in stirred vessel and adopting temperature (T), pH, rotational speed (N), initial phenol (C P,o) and enzyme (C T) concentrations as independent variables. Experimental data of the residual phenol concentration (C P) were used to calculate the oxidation efficiency (η), initial oxidation rate (-r o) and time required to reach the end of reaction (t) that were selected as the responses. Under the optimal conditions (T = 45°C, pH 6.6, N = 400 rpm, C P,o = 100 ppm and C T = 50 U/mL), η was 88.1%, -r o = 10.2 mg L-1 min-1, t = 40 min. These results suggested that tyrosinase-rich crude extracts from vegetable byproducts could be quite promising.

Heat Shock Protein Hspa5 Interacts with and Protects Tyrosinase Activity BABT

In this study, heat shock protein, Hspa5 was cloned, expressed and purified subsequently confirmed that it interacted with the tyrosinase (TYR) in vitro. Then, using the crystal structure of the homologous protein from the bacteria as a template, a homology model of human TYR was constructed. This model was further applied to investigate the molecular docking with Hspa5. The model showed that the interaction between the TYR and Hspa5 was mainly maintained by some hydrogen bonds in a quite low energy state. The results indicated that TYR was protected in different denaturation conditions by Hspa5. Production and characterization of tyrosinase activity in Pycnoporus sanguineus CCT-4518 crude extract BJM

Tyrosinase is an enzyme of industrial interest. The production and characterization of tyrosinase from P. sanguineus CCT-4518 were investigated. The selection of inductors, luminosity influence, inoculum size and type of culture medium on the production of tyrosinase and the effect of inhibitors on enzyme activity were performed. Optimum conditions for intracellular tyrosinase production was observed after 2 days using 0.15% L-tyrosine as inducer, in the presence of light, with inoculum size of 10 mycelium discs, using 2% malt extract broth medium, incubated at 30°C, and constant agitation of 150 rpm. Utilization of [Bmim]BF4-MAE on enhancement of γ-oryzanol extraction from rice bran and its tyrosinase inhibitory activity BJPS

This study investigated the effect of ionic liquid (IL)-microwave assisted extraction (MAE) on γ-oryzanol extraction from rice bran (Oryza sativa L.). The conditions included the concentration of IL, liquid (IL)/solid (sample) ratio, extraction time, and microwave power. The use of 1-butyl-3-methylimidazolium ([Bmim]BF4) as a selected solvent in IL-MAE method was developed for extraction of γ-oryzanol from rice bran. The Box-Behnken design on four factors with response surface methodology (RSM) was used to optimize the experimental conditions.
