In a previous study, we confirmed alterations in several fungal traits in hybrid strains of Verticillium lecanii generated via protoplast fusion. In the present study, we surveyed radial colony growth in different temperature regimes as well as conidia productivity and conidia size of hybrid strains of V. lecanii to elucidate the various primary fungal properties of these strains and to determine the correlation between these properties and virulence to insect pests. Minimum temperature thresholds for all tested strains (except Vertalec®) were below 5°C. Optimal temperatures for colony growth were 20–25°C. Recently, Verticillium lecanii was transferred and subdivided into mainly Lecanicillium lecanii, L. muscarium, and L. longisporum according to morphological observations and PCR-RFLPs of ITS, mtDNA and β-tubulin. To clarify whether 65 isolates of V. lecanii of mainly Japanese isolates were identified as Lecanicillium spp., conidia size and the formation of secondary phialide formation were investigated under a light microscope, and DNA polymorphism in the j8-tubulin gene of V. lecanii was analyzed by PCR-RFLP. These results indicated that the examined isolates belonged to L. muscarium (60 isolates) and L. longisporum (4 isolates), except for one isolate which alone did not belong to both Lecanicillium spp. |

The ability of Verticillium lecanii (Lecanicillium muscarium; B-2) to induce host resistance and defense responses against subsequent challenge with cucumber powdery mildews was examined in a glasshouse. A root test showed that, after inoculation with V. lecanii blastspore on cucumber roots, induction of systemic resistance in those V. lecanii pre-inoculated plants engendered significantly fewer lesions and reduced disease severity compared with non-inoculated plants. Furthermore, in plants inoculated with V. lecanii blastspore on their roots, the fungi showed high colonizing ability on cucumber rhizoplane and inside root tissues. In a previous study, protoplast fusion was performed among 3 strains of the entomopathogenic fungi Verticillium lecanii, as a result of which 174 hybrid strains were obtained. The aim of the present study was to screen desirable hybrid strains that have a wide host range or durable effects. Initially, 43 hybrid strains were used in bioassays against the cotton aphid. Of these strains, 30 induced equal or higher mortality as compared to Vertalec (41.9%). Secondly, 50 hybrid strains were used in bioassays against the greenhouse whitefly. Of these strains, 37 exhibited an equal or higher infection rate as compared to that of Mycotal (36.2%). ダイコンバーティシリウム黒点病に対する防除効果を昆虫寄生菌Verticillium lecaniiを用いて検討した。温室試験と圃場試験でV. lecaniiの7菌株(A-2、B-2、C-1、2aF30、2aF31、2aF43、AaF101)を用いて実施した。V. lecaniiの分生胞子を処理したポット試験では全てのV. lecanii菌株で高い防除効果があり、病原菌単独処理と比較して有意な差が認められた。なかでもB-2が最も高い防除効果を示し、病原菌単独処理の発病率51.2%と比較して31.4%となった。V. lecaniiのフスマ培養土を用いたポット試験における病徴抑制効果は病原菌単独処理区との有意な差が認められたのはC-1処理区のみであった。A-2、B-2処理区においても抑制効果の傾向がみられたが、生長促進効果では顕著な差はみられなかった。また、圃場試験では全てのV. lecanii処理区で発病が抑制された。特にB-2、C-1においては高い発病抑制効果があり、病原菌単独処理区の半分以下に抑制していた。商品価値調査においてはV. lecanii処理区で商品価値のあるダイコンの数が優った。以上の結果から、病原菌単独処理と有意な差が認められなかったものもあるが、V. lecaniiはダイコンバーティシリウム黒点病を抑制するのに有効である傾向が確認された。よって、V. lecaniiはダイコンバーティシリウム黒点病に対して生物防除資材として有望であることが示唆された。 Seven strains of Verticillium lecanii were tested for their capacity to control Verticillium black-spot of Japanese radish... |
