In recent years, xylanases have expanded their use in many processing industries, such as pulp and paper, food and textile. Thermomyces lanuginosus IOC-4145 was able to produce a very high level of cellulase-free xylanase in shaken cultures using corncob as substrate (500 U/mL). An optimization of the medium composition in submerged fermentation was carried out aiming at a low cost medium composition for enzyme production. Statistical experiment design was employed for this purpose, pointing out corncob as the most important parameter, which affects enzyme production. En este estudio se evaluó el efecto del complejo Lolium-Festuca (CL-F), cosechado a 28 (T1) y 35 d (T2) de rebrote, en la desaparición in vitro de la materia seca (DIVMS), cinética de producción de gas (CPG), y en la actividad de carboximetilcelulasas (CMCasas) y de xilanasas de microorganismos adheridos a sólidos a las 12, 16, 24, y 48 h de incubación. La degradación del tejido foliar a las 12, 24, y 48 h de incubación se documentó mediante Microscopía de Electrónica de Barrido (MEB); la lignina se detectó con la reacción floroglucinol-HCl. El modelo logístico y el procedimiento NLIN se usaron para obtener los datos de CPG. Cellulolytic properties of two white rot fungi, Bjerkandera adusta and Pycnoporus sanguineus, cultivated on wheat straw agar medium, were characterized and compared. Optimal growing parameters for maximum enzyme production for both fungi were wheat straw medium pH 5 and 28ºC. B. adusta showed, on the 6th day of culture, carboxymethylcellulose (CMC)ase activity levels 1.6 times higher than maximal P. sanguineus activity, achieved on the 8th day. B. adusta supernatants also displayed higher activity levels towards xylan (3.6-fold) compared to those of P. sanguineus. Background Two xylanases, Xyl I and Xyl II, were purified from the crude extracellular extract of a Trichoderma inhamatum strain cultivated in liquid medium with oat spelts xylan. Results The molecular masses of the purified enzymes estimated by SDS-PAGE and gel filtration were, respectively, 19 and 14 kDa for Xyl I and 21 and 14.6 kDa for Xyl II. The enzymes are glycoproteins with optimum activity at 50°C in pH 5.0-5.5 for Xyl I and 5.5 for Xyl II. The xylanases were very stable at 40°C and in the pH ranges from 4.5-6.5 for Xyl I and 4.0-8.0 for Xyl II. A fibrolytic enzyme complex was added to the pre-starter diet. Broiler chicks were randomly distributed into five treatments, consisting of a diet with no enzyme addition and four test diets supplemented with 100, 200, 300 and 400g/T of an enzyme complex. The dietary inclusion of the enzyme complex increased weight gain, and the dose of 300g/T improved weight gain and worsened feed conversion ratio. |
