The aim of this work was to explore the possibility of obtaining transgenic plants of onion varieties cultivated in Argentina, starting from calli induced from mature zygotic embryos, using two strains of Agrobacterium tumefaciens as transfection vectors. Mature embryos from three varieties of 'Valenciana' onion, Torrentina, Cobriza INTA and Grano de oro were in vitro cultivated for callus induction. After three to four months an average of 57.4% success for the three varieties was reached. Embriões zigóticos em diferentes estádios de desenvolvimento foram utilizados para avaliar o potencial de germinação e competência embriogênica in vitro. Frutos coletados entre 15 e 360 dias após a antese foram avaliados em relação ao diâmetro (mm), comprimento (mm) e massa fresca (g). Para avaliar a percentagem de germinação in vitro, embriões zigóticos em diferentes estádios de desenvolvimento foram inoculados em meio de cultura composto de sais minerais de Murashigue & Skoog (MS), ou em meio MS suplementado com 4,4 miM de BAP, sacarose (3%), ágar (0,6%), carvão ativado (0,15%). The experiment was carried out to determine the appropriate dose of coconut water as supplement for in vitro cultivation of zygotic embryos from 19 olive genotypes. The isolated embryos of the olive seeds were immersed on culture medium containing 0 (control), 25, 50, and 100mL L-1 of fresh and sterile coconut water and kept for 45 days under controlled environment. The percentage of germination, shoot length, number of roots, number of leaves and number of internodes were measured for all 19 olive genotypes.
