Basic research for technical development of new malarial vector control by using sublethal effect of entomopathogenic fungi OAK

野生の蚊類より昆虫寄生性アナモルフ菌類の分離を試み、合計392菌株を収蔵する菌株ライブラリを構築した。これら菌株の感染性および病原性を評価したところ13菌株がAnopheles stephensiに対し感染性を有することが明らかとなった。この内、高い感染性を示した5菌株(Lecanicillium属菌2菌株とB. bassiana 3菌株)の病原性を評価したところ、対照区の半数致死日数を半数以下に下回る高病原性系統および対照区と同程度の値を示す弱病原性系統を検出した。

Biological control of Verticillium black spot of Japanese radish using Bacillus spp. and genotypic differentiation of selected antifungal Bacillus strains with antibiotic marker OAK

Bacillus species can form endospores and have broad-spectrum activity of their antibiotics. For that reason, they offer several advantages over other bacteria for protection against root pathogens. This work was intended to determine the ability of Bacillus spp. to inhibit Verticillium dahliae. First, antagonistic activities of Bacillus spp. were studied in greenhouse trials. These results suggest that the bacterium can control soil-borne diseases. Pathogenicity and its mode of action in different sedentary stages of Heterodera glycines (Tylenchida: Heteroderidae) by Verticillium lecanii hybrid strains OAK

The current study was conducted to investigate the pathogenicity and its mode of action by Verticillium lecanii hybrid strains in different sedentary stages of Heterodera glycines. Three different sedentary stages (pale yellow female, yellow brown cyst, and dark brown cyst) of H. glycines were treated with V. lecanii and incubated for 3 weeks on water agar. After 3 weeks of incubation, eggs were investigated for the following parameters: (i) the infection frequencies of eggs, (ii) the number of eggs laid, and (iii) the number of mature and healthy eggs. Pre-inoculation of cucumber roots with Verticillium lecanii (Lecanicillium muscarium) induces resistance to powdery mildew OAK

The ability of Verticillium lecanii (Lecanicillium muscarium; B-2) to induce host resistance and defense responses against subsequent challenge with cucumber powdery mildews was examined in a glasshouse. A root test showed that, after inoculation with V. lecanii blastspore on cucumber roots, induction of systemic resistance in those V. lecanii pre-inoculated plants engendered significantly fewer lesions and reduced disease severity compared with non-inoculated plants. Furthermore, in plants inoculated with V. lecanii blastspore on their roots, the fungi showed high colonizing ability on cucumber rhizoplane and inside root tissues. Reclassification of Japanese Isolates of Verticillium lecanii to Lecanicillium spp OAK

Recently, Verticillium lecanii was transferred and subdivided into mainly Lecanicillium lecanii, L. muscarium, and L. longisporum according to morphological observations and PCR-RFLPs of ITS, mtDNA and β-tubulin. To clarify whether 65 isolates of V. lecanii of mainly Japanese isolates were identified as Lecanicillium spp., conidia size and the formation of secondary phialide formation were investigated under a light microscope, and DNA polymorphism in the j8-tubulin gene of V. lecanii was analyzed by PCR-RFLP. These results indicated that the examined isolates belonged to L. muscarium (60 isolates) and L. longisporum (4 isolates), except for one isolate which alone did not belong to both Lecanicillium spp.

Screening of Verticillium lecanii (Lecanicillium spp.) Hybrid Strains Based on Evaluation of Pathogenicity against Cotton Aphid and Greenhouse Whitefly, and Viability on the Leaf Surface OAK

In a previous study, protoplast fusion was performed among 3 strains of the entomopathogenic fungi Verticillium lecanii, as a result of which 174 hybrid strains were obtained. The aim of the present study was to screen desirable hybrid strains that have a wide host range or durable effects. Initially, 43 hybrid strains were used in bioassays against the cotton aphid. Of these strains, 30 induced equal or higher mortality as compared to Vertalec (41.9%). Secondly, 50 hybrid strains were used in bioassays against the greenhouse whitefly. Of these strains, 37 exhibited an equal or higher infection rate as compared to that of Mycotal (36.2%). Variation in growth at different temperatures and production and size of conidia in hybrid strains of Verticillium lecanii (Lecanicillium spp.) (Deuteromycotina: Hyphomycetes) OAK

In a previous study, we confirmed alterations in several fungal traits in hybrid strains of Verticillium lecanii generated via protoplast fusion. In the present study, we surveyed radial colony growth in different temperature regimes as well as conidia productivity and conidia size of hybrid strains of V. lecanii to elucidate the various primary fungal properties of these strains and to determine the correlation between these properties and virulence to insect pests. Verticillium lecanii is a potentially useful biological control agent (BCA) for soybean cyst nematode (SCN), Heterodera glycines. The objective of this study was to screen hybrid strains of V. lecanii, derived from protoplast fusion, for effective control agents of SCN, and to investigate whether the protoplast fusion technique is an effective tool for development of nematode control agents. Three parental strains (Vertalec®, Mycotal®, and B-2) and their 162 hybrid strains were screened in greenhouse pot tests. 圃場還元利用されている実際的な牛ふん尿堆肥に含まれる腐植酸およびフルボ酸の化学的特徴 OAK

本研究では,堆肥化の進行や腐熟度が大きく異なる牛ふ ん尿堆肥について,堆肥製造業者や一般の畜産農家および畑作農家が製造し,かつ圃場還元している実際的な堆肥を対象に,牛ふん尿の堆肥化と腐熟化に伴う腐植酸およびフルボ酸の化学的特徴,および腐植酸のスペクトル解析から構造単位の存在割合の違いを明らかにした.さらに土壌中での可動性が異なると考えられる腐植を水および水酸化ナトリウム溶液により抽出して比較し,その化学的特徴や構造単位の違いを明らかにした. 堆肥化期聞が異なる牛ふん尿バーク堆肥では,堆肥化による腐熟化の進行に伴って,沈殿部割合や腐植酸の相対色度(RF値)が増加した. 堆肥化および腐熟化の程度が異なる4種類の牛ふん尿堆 肥から水酸化ナトリウム溶液および水により抽出された腐植酸とフルボ酸の光学的特性を比較した.水酸化ナトリウム溶液では,堆肥化や腐熟化がほとんど進行していないと判断された高水分乳牛ふん尿堆肥から極めて未熟なRp型腐植酸が抽出され,牛ふん中の未消化繊維や敷料などの植物腐朽遺体に由来するリグニンなどの成分に由来すると考えられた.腐熟化の進行が認められた肉牛ふん尿敷料堆肥や肉牛ふん尿敷料と鶏糞との混合堆肥では,好気的な発酵と温度上昇に伴って生成した腐植化の進んだ腐植酸が含まれていると考えられ,とくに腐熟度の高かった鶏糞との混合堆肥からB型腐植酸が抽出された.水抽出では,水酸化ナトリウム溶液による抽出よりも腐植の抽出割合が低かったが,水抽出された腐植酸のRF値は水酸化ナトリウム溶液で抽出された腐植酸よりも高かった. 紫外可視吸収スペクトルおよびフーリエ変換赤外吸収ス...

昆虫寄生性Lecanicillium muscarium融合株2aF43およびオンシツツヤコバチの併用によるオンシツコナジラミ防除効果の評価 OAK

This study aims to evaluate the control efficacy of greenhouse whitefly by co-application of entomopathogenic fungus Lecanicillium muscarium hybrid strain 2aF43 and greenhouse whitefly parasitoid Encarsia formosa, and to reveal the interaction between both natural enemies. Although, the emergence rate of adult greenhouse whitefly reached 93.2% in control plot, co-application plot indicated significantly lower emergence rate (4.2%). Co-application plot showed higher fungal infection compared with singularly L. muscarium inoculated plot. 昆虫寄生菌Verticillium lecanii (Lecanicillium spp.) によるダイコンバーティシリウム黒点病の生物防除 OAK

ダイコンバーティシリウム黒点病に対する防除効果を昆虫寄生菌Verticillium lecaniiを用いて検討した。温室試験と圃場試験でV. lecaniiの7菌株(A-2、B-2、C-1、2aF30、2aF31、2aF43、AaF101)を用いて実施した。V. lecaniiの分生胞子を処理したポット試験では全てのV. lecanii菌株で高い防除効果があり、病原菌単独処理と比較して有意な差が認められた。なかでもB-2が最も高い防除効果を示し、病原菌単独処理の発病率51.2%と比較して31.4%となった。V. lecaniiのフスマ培養土を用いたポット試験における病徴抑制効果は病原菌単独処理区との有意な差が認められたのはC-1処理区のみであった。A-2、B-2処理区においても抑制効果の傾向がみられたが、生長促進効果では顕著な差はみられなかった。また、圃場試験では全てのV. lecanii処理区で発病が抑制された。特にB-2、C-1においては高い発病抑制効果があり、病原菌単独処理区の半分以下に抑制していた。商品価値調査においてはV. lecanii処理区で商品価値のあるダイコンの数が優った。以上の結果から、病原菌単独処理と有意な差が認められなかったものもあるが、V. lecaniiはダイコンバーティシリウム黒点病を抑制するのに有効である傾向が確認された。よって、V. lecaniiはダイコンバーティシリウム黒点病に対して生物防除資材として有望であることが示唆された。 Seven strains of Verticillium lecanii were tested for their capacity to control Verticillium black-spot of Japanese radish...
