Un nouveau virus infectant le ver rose du cotonnier, #Pectinophora gossypiella$, a été isolé et purifié à partir de larves mortes récoltées en champ de coton. Les virions purifiés de 27 plus ou moins 1 nm de diamètre, ont une forme icosaédrique et contiennent un génome à ARN. Trois protéines capsidaires de 31,7, 32,6 et 47,4 Kd ont été trouvées dans les virions. Le virus purifié n'est pas hautement infectieux pour les larves, cependant la durée du stade chrysalide est supérieure chez les lots infectés artificiellement. Dans les tissus atteints, les virions sont dispersés dans le cytoplasme des cellules de l'intestin ou bien groupés en amas paracristallins de même type que chez d'autres insectes infectés avec des picornavirus. The nucleoprotein and non-capsid protein of maize yellow stripe virus (MYSV) were purified from naturally or experimentally infected maize or sorghum plants. In SDS-PAGE, the apparent molecular weight of the nucleoprotein was 35,6 KD, and that of the non-capsid protein was 14,7 KD. Similar to tenuiviruses, the nucleoprotein of MYSV was associated with fine filaments and the non-capsid protein formed typical needle-shaped crystals. Following a 2-day acquisition feeding period on MYSV-infected plants, vector leafhoppers (#Cicadulina chinai$) remained highly infective for 21 days. The pink borer, #Sesamia cretica$ Lederer (#Lepidoptera$ : #Noctuidae$), is a main pest of maize in Egypt. Attacks by #S. cretica$ are usually high on early plantations of April, when the first generation of adults emerge after a period of larval hibernation. The present study aimed to evaluate the feasibility of the granulosis virus of #S. cretica$, recently isolated by Fediere et al. (1993) as a microbiological agent. Une lignée cellulaire a été obtenue à partir d'embryocultures de #Phthorimaea operculella$ (Zeller), le principal ravageur de la pomme de terre dans les régions tropicales et sub-tropicales. La lignée cellulaire, nommée ORS-Pop-93, cultivée en milieu Grace modifié, a une population hétérogène de cellules arrondies et allongées très adhérentes. Le temps de doublement des cellules est de 40 h. La lignée a été subcultivée plus de 40 fois. Le profil polypeptidique est différent de celui d'autres lignées de lépidoptères. Two cell lines from the most important insect pest of cotton, #Pectinophora gossypiella$ S., were obtained from an embryonic tissue culture. The cells were cultured in GRACE's modified medium. The first cell line, designated ORS-PgS (PgS), was obtained by selecting the non-adherent cells during subculturing and consisted of a homogeneous population of rounded cells. The second cell line, designated ORS-PgA (PgA), obtained without any artificial selection, was a heterogeneous population of spherical and spindle-shaped cells with a great capacity to adhere. Six natural isolates belonging to #Spodotera littoralis$ NPV were identified. Using the restriction endonucleases Eco RI and Bam HI, the DNA profiles of each isolate from Giza, Menya, Kaliubeia-Kaha, Kafr El-Sheikh, Gharbeia and Kaliubeia-Tokh were differentiated from that of the reference Egypt-84. DNA electrophoretic profiles on different isolates collected 5 years later were compared to the difference type.
