Sporadic high mortalities were reported in June 1997 among hatchery-reared larval Manila clam Ruditapes philippinarum in a French commercial hatchery. Cellular abnormalities were observed in semi-thin sections in affected animals. Transmission electron microscopy revealed the presence of herpes-like virus particles in larvae. This is the first description of a herpes-like virus infection in larval R. philippinarum, a non-ostreid bivalve species. Virus particles were similar to other herpes-like viruses described from different oyster species with respect to ultrastructure and morphogenesis. Electron microscopic examination also demonstrated cells with condensed chromatin and extensive perinuclear fragmentation of chromatin. Like viruses infecting oysters,... |

An understanding of husbandry strategies and any associated risk factors is important for designing management control measures that can reduce mortality in Pacific oysters, Crassostrea gigas, caused by ostreid herpesvirus 1 (OsHV-1). The type of culture facility can be considered in relation to the potential pathways that could lead to the entry of a pathogen and its survival. In addition, the animal host (e.g. age, physiological state, selective breeding programmes), husbandry procedures (e.g. stocking density), the pathogen itself (e.g. pathogenicity, virulence) and environmental effects (e.g. temperature) represent other relevant interconnected factors. However, all these factors provide valuable background information for outlining the mitigation... |

Bonamiosis was first reported in association with mass mortality of flat oysters, Ostrea edulis, in June 1979 in L'Ile Tudy, Brittany. The disease rapidly spread to all of the oyster farming areas in France but also in other European countries. The French flat oyster production, which already suffered from another protozoan disease, marteiliosis, decreased from 5500 mt in 1979 to less than 2000 mt after 1980. In 2001, about 350 French farms sell 1650 mt of flat oysters. This production mainly relies on natural spat collection which specially occurs in Quiberon bay. One third of this spat is transferred from South to North Brittany for further growth. Quiberon bay constitutes an interesting site regarding the surveillance of bonamiosis because of the... |

Macroautophagy is a mechanism that is involved in various cellular processes, including cellular homeostasis and innate immunity. This pathway has been described in organisms ranging in complexity from yeasts to mammals, and recent results indicate that it occurs in the mantle of the Pacific oyster, Crassostrea gigas. However, the autophagy pathway has never been explored in the hemocytes of C. gigas, which are the main effectors of its immune system and thus play a key role in the defence of the Pacific oyster against pathogens. To investigate autophagy in oyster hemocytes, tools currently used to monitor this mechanism in mammals, including flow cytometry, fluorescent microscopy and transmission electron microscopy, were adapted and applied to the... |

Environmental DNA approaches are increasingly used to detect microorganisms in environmental compartments, including water. They show considerable advantages to study non‐cultivable microorganisms like Bonamia ostreae, a protozoan parasite inducing significant mortality in populations of flat oyster Ostrea edulis . Although B. ostreae development within the host has been well described, questions remain about its behaviour in the environment. As B. ostreae transmission is direct, seawater appears as an interesting target to develop early detection tools and improve our understanding of disease transmission mechanisms. In this context, we have developed an eDNA/eRNA approach allowing detecting and quantifying B. ostreae 18S rDNA/rRNA as well as monitoring... |

In 1972, France underwent an oyster (Crassostrea angulata) crisis and urgently needed to diversify its aquaculture. Thus, Asari clam (Ruditapes philippinarum) was introduced at that time for aquaculture purpose, concomitantly with the introduction of Pacific oyster (Crassostrea gigas). All Asari clam adults and spat originated from the same area (Puget Sound, WA, USA). After a promising start and the implementation of a national Research & Development program, Asari clam culture rapidly faced a series of concomitant handicaps: firstly, in spite the fact that cultural practices were optimized and locking points addressed, leasing ground availability was limited during the 1980s’ due to certain reluctance from oyster farmers to share their leases and/or... |

The REPAMO network (network ofmollusc pathology), created in 1986, ensures the survey of shellfish health status along French coasts according? to European Directives 91/67/EEC and 95/70/EEC. The aims of the network are 1 - to prevent the introduction and spread of exotic pathogens, 2-to study the means to decrease the impact of pathogens already present and to survey their evolution. In 2003, the REPAMO network focused on the survey oflisted diseases (bonamiosis and marteiliosis) in two areas under agreement process (Banc de Granville and zone X) and on the study of abnormal morlalities. Number of reported abnormal morlalities has increased in 2003. This rise can be partly explained by the recent network restructuring, but also by the summer heatwave and... |

Bonamiosis due to the intrahaemocytic protistan parasite Bonamia ostreae is a European endemic disease affecting flat oysters Ostrea edulis. After its first description in June 1979 in L'Ile Tudy, Brittany, the parasite rapidly spread to all French oyster farming areas and in other European countries through transfers of live molluscs. Bonamiosis and marteiliosis, another protozoan disease appeared in the seventies, drastically reduced the French flat oyster production from 20 000 tonnes in 1970 to less than 2000 tonnes after 1981. In 2001, about 1650 t of flat oysters were marketed in France. The production is restricted to several areas specialised in reproduction, growth and/or marketing, and depends on transfers of animals. Most important production... |

Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in its aquaculture production. Selective breeding programs for resistance to bonamiosis have been undertaken to counter this disease. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters injected with the parasite to two tested families of oysters (20 and 8-month old at the beginning of the experiment, with different genetic backgrounds) originating from a selective breeding program developed by IFREMER in France. Mortalities were checked daily and ventricular heart smears were performed on dying or moribund oysters to detect the... |
