Soybean is a legume with large economic importance in the international market, with a world production of almost two hundred and ten million tons in the 2008/2009 harvest. Brazil appears as the second largest producer, with about twenty-five percent of the world production. In 2007, the Brazilian Soybean Genome Consortium (GENOSOJA) was established with the goal of integrating several institutions currently working with soybean genomics in Brazil. Some genes affect meat quality in chickens. We looked for polymorphisms in the Gallus gallus α-RyR gene (homologous to RyR 1) that could be associated with PSE (pale, soft and exudative) meat. Because RyR genes are over 100,000 bp long and code for proteins with about 5000 amino acids, primers were designed to amplify a fragment of hotspot region 2, a region with a high density of mutations in other species. Total blood DNA was extracted from 50 birds, 25 that had PSE meat and 25 normal chickens. The DNA samples were amplified by PCR, cloned, sequenced, and used to identify single nucleotide polymorphisms (SNPs). The Genosoja consortium is an initiative to integrate different omics research approaches carried out in Brazil. Basically, the aim of the project is to improve the plant by identifying genes involved in responses against stresses that affect domestic production, like drought stress and Asian Rust fungal disease. To do so, the project generated several types of sequence data using different methodologies, most of them sequenced by next generation sequencers. Plants have the ability to recognize and respond to a multitude of pathogens, resulting in a massive reprogramming of the plant to activate defense responses including Resistance (R) and Pathogenesis-Related (PR) genes. Abiotic stresses can also activate PR genes and enhance pathogen resistance, representing valuable genes for breeding purposes. The present work offers an overview of soybean R and PR genes present in the GENOSOJA (Brazilian Soybean Genome Consortium) platform, regarding their structure, abundance, evolution and role in the plantpathogen metabolic pathway, as compared with Medicago and Arabidopsis. Com o objetivo de avançar o conhecimento sobre o mecanismo molecular de defesa da soja frente à ferrugem asiática, avaliou-se por meio da técnica de PCR em tempo real, a expressão de genes supostamente envolvidos no mecanismo de defesa da planta. O experimento foi conduzido em sala climatizada e para a análise de expressão gênica, foram coletadas amostras de RNAs de folhas da soja inoculadas com o patógeno e falso-inoculadas (água) em um genótipo tolerante (PI 230970) e um genótipo suscetível (Embrapa 48) a Phakopsora pachyrhizi. Soybean powdery mildew (Erysiphe diffusa) was considered a minor disease in Brazil in the decades immediately after its identification. However, since the outbreak in 1996/97 in all cultivated areas the disease has become a constant threat to farmers and losses of up to 25% have been reported. The report of a new species, E. glycines, infecting soybean in Japan, and the occurrence of the disease in other plant species (Phaseolus vulgaris, Helianthus annuus,Sonchus oleraceus,Hypochaeris brasiliensis, and Bidens pilosa) commonly found growing nearby soybean fields, raised questions in relation to the taxonomy of the powdery mildew strains found in or around soybean fields in Brazil. Plantas de soja, cv. BRS 154, morreram antecipadamente em março de 2006, em Passo Fundo e Coxilha, RS, com folhas secas presas às plantas e regiões escurecidas na porção inferior das hastes (cancros). Foram obtidos 25 isolados com micélio branco-acinzentado típico do gênero Diaporthe. Três destes (CH 07/06, CH 10/06 e CH 40/06) foram selecionados para estudos comparativos da região do rDNA, sendo constatada alta similaridade (100%) com a seqüência depositada para este fungo (GenBank AF0000567), correspondente à espécie D. phaseolorum var: caulivora strain 713. CH 07/06 e CH 10/06 apresentaram padrões de agressividade diferentes de CH 08/89 (isolado padrão da var. meridionalis), não causando morte de plantas em cultivar suscetível à D. phaseolorum var.... |
