Aims: To develop a real-time (rt) PCR for species differentiation of thermophilic Campylobacter and to develop a method for assessing co-colonization of pigs by Campylobacter spp. Methods and results: The specificity of a developed 5’nuclease rt-PCR for species-specific identification of C. jejuni, C. coli, C. lari, C. upsaliensis and of a hipO gene nucleotide probe for detection of C. jejuni by colony-blot hybridization were determined by testing a total of 75 reference strains of Campylobacter spp. and related organisms. PathOrganic assesses risks associated with the consumption of fresh and minimally processed vegetables due to the prevalence of bacterial human pathogens (e.g. Salmonella enterica, pathogenic E. coli, Campylobacter spp., Listeria monocytogenes) in organically grown plant produce. The project aims at evaluating whether organic production poses a risk on food safety and addresses the food chain by taking into consideration potential sources of pathogen transmission (e.g. animal manure). Reducing the presence of human pathogens like Campylobacter and Salmonella (zoonoses) in their animal hosts is important to enhance food safety of products of animal origin. Campylobacter is considered to be a commensal in the gastrointestinal tract of pigs due to its typically high prevalence. Consequently, it is difficult to control Campylobacter in pigs at farm level by usual hygienic measures, especially in open systems of organic pig production (Jensen et al 2006). However, another potential means to control pathogens is inclusion of non-digestible oligosaccharides (prebiotics) in the diet. Salmonella bakterier kan spredes blandt økologiske grise Organic Eprints

Forsøg har vist, at Salmonella bakterier kan overføres fra smittede økogrise til salmonella-frie grise holdt på udendørs arealer. Desuden kan salmonellabakterier overleve i foldmiljøet og under visse betingelser smitte salmonella-frie grise, som senere indsættes i folden. Overaskende blev der fundet flere forskellige typer af salmonellabakterier i både grisene og i foldmiljøet i løbet af forsøgsperioden.

Salmonella og Campylobacter i økologisk svineproduktion Organic Eprints

De mere ekstensive systemer i økologisk svineproduktion formodes at have en positiv effekt på dyrenes robusthed f.eks. over for infektioner. Der er dog ingen dokumentation for, at økologiske svin har et lavere indhold af de almindelige zoonotiske bakterier som f.eks. Salmonella og Campylobacter end konventionelle svin. Et forsøg med økologiske smågrise viste, at salmonellainfektioner kan overføres både mellem grisene men også via et salmonellasmittet miljø. Graden af den smitte grisene udsættes for synes væsentlig for etablering af en infektion. Campylobacter, som anses for at være en naturlig del af grisens tarmflora, blev fundet i alle smågrisene. Desuden var det muligt at påvise både den typisk svineassocierede Campylobacter art C. coli og arten C....

Survival and transmission of Salmonella enterica serovar Typhimurium in an outdoor organic pig farming environment Organic Eprints

It was investigated how organic rearing conditions influence the Salmonella enterica infection dynamics in pigs and whether Salmonella persists in the paddock environment. Pigs inoculated with S. enterica serovar Typhimurium were grouped with Salmonella-negative tracer pigs. Bacteriological and serological testing indicated that organic pigs were susceptible to Salmonella infections, as 26 of 46 (56%) tracer pigs turned culture positive. An intermittent and mainly low-level excretion of Salmonella (<100 cells per g partly explains why the bacteriological prevalence appeared lower than the seroprevalence. The occurrence and characterization of Campylobacter jejuni and C. coli in organic pigs and their outdoor environment Organic Eprints

The occurrence and species distribution of thermophilic Campylobacter was investigated in organic outdoor pigs. An increased exposure of outdoor pigs to C. jejuni from the environment may cause a shift from a normal dominance of C. coli to more C. jejuni, which may imply a concern of reduced food safety. Bacteriological methods for determination of Campylobacter excretion level were combined with colony-blot hybridization and real-time PCR for specific detection of C. jejuni in pigs.
