Altered expression of genes functioning in lipid homeostasis is associated with lipid deposition in NOD mouse lacrimal gland Inra
Wu, K.; Joffre, C.; Xiaodong Li, X.L.; MacVeigh-Aloni, M.; Hom, M.; Hwang, J.; Chuanqing Ding, C.D.; Grégoire, S.; Bretillon, L.; Zong, Z.F.; Hamm-Alvarez, S.F..
Functional atrophy and accompanying lymphocytic infiltration and destruction of the lacrimal gland (LG) are characteristics of Sjögren's Syndrome (SjS). The male NOD mouse is an experimental model for the autoimmune exocrinopathy that develops in the LG of SjS patients. Acinar cells in LG of male NOD mice aged 3–4 months were previously shown to accumulate lipid droplets. In the current study, analysis of lipid components revealed that the accumulated lipids were mostly cholesteryl esters (CE). Gene expression microarray analysis followed by real-time RT-PCR revealed alterations in the expression of several genes involved in lipid homeostasis in LG of 12-week-old male NOD mice relative to matched BALB/c controls. A series of upregulated genes including...

Branched-chain fatty acids, increased in tears of blepharitis patients, are not toxic for conjunctival cells Inra
Joffre, C.; Souchier, M.; Leclere, L.; Buteau, B.; Grégoire, S.; Lizard, G.; Montange, T.; Acar, N.; Bron, A.; Creuzot Garcher, C.; Diebold, Y.; Bretillon, L..
Aim: The composition of the meibum of blepharitis patients is characterised by increased levels of branched-chain fatty acids (BCFAs) that return to normal values in patients treated with cyclins and lid hygiene. The aim of this study was to determine if BCFAs had toxic effects on conjunctival cells related to the disease. Methods: Chang and IOBA-NHC conjunctival human cells were treated with BCFAs (isoC16 and isoC20) or palmitic acid as a control for 4 h or 24 h at 50 μM or 100 μM. Morphological and functional changes were investigated by measuring mitochondrial dehydrogenase activity, cell permeability, mitochondrial depolarisation, chromatin condensation, IL-1β and reactive oxygen species production. Results: None of the fatty acids modified the...

Differential effect of maternal diet supplementation with alpha-linolenic adcid or n-3 long-chain polyunsaturated fatty acids on glial cell phosphatidylethanolamine and phosphatidylserine fatty acid profile in neonate rat brains Inra
Destaillats, F.; Joffre, C.; Acar, N.; Joffre, F.; Bezelgues, J.B.; Pasquis, B.; Cruz-Hernandez, C.; Rezzi, S.; Montoliu, I.; Dionisi, F.; Bretillon, L..
Background: Dietary long-chain polyunsaturated fatty acids (LC-PUFA) are of crucial importance for the development of neural tissues. The aim of this study was to evaluate the impact of a dietary supplementation in n-3 fatty acids in female rats during gestation and lactation on fatty acid pattern in brain glial cells phosphatidylethanolamine (PE) and phosphatidylserine (PS) in the neonates. Methods: Sprague-Dawley rats were fed during the whole gestation and lactation period with a diet containing either docosahexaenoic acid (DHA, 0.55%) and eicosapentaenoic acid (EPA, 0.75% of total fatty acids) or α-linolenic acid (ALA, 2.90%). At two weeks of age, gastric content and brain glial cell PE and PS of rat neonates were analyzed for their fatty acid and...

Effect of dietary omega-3 and omega-6 fatty acids on IOP elevation, electroretinographic changes and retinal ganglion cell loss in a laser-induced rat model of glaucoma Inra
Schnebelen, N.; Pasquis, B.; Salinas-Navarro, M.; Joffre, C.; Creuzot Garcher, C.; Vidal-Sanz, M.; Bron, A.; Bretillon, L.; Acar, N..
Purpose:To test the efficacy of dietary omega-3 and omega-6 fatty acids in a rat model of glaucoma induced by laser photocoagulation. Methods:Male Sprague Dawley rats were fed for 3 months with a diet containing either: 1) 17% of omega-3 fatty acids (10% EPA + 7% DHA), 2) 10% of omega-6 fatty acids (as GLA), or 3) a combination of both omega-3 and omega-6 fatty acids (10% EPA + 7% DHA + 10% GLA), by comparison with a control group of animals fed with a standard diet deprived of EPA, DHA and GLA (n=10 in each group). After 3 months of diet, glaucoma was induced in one eye of each animal by laser photocoagulation (532nm) of the episcleral veins, the trabeculum and the limbal veins. During the 3 following months, the animals were fed on the same diets, IOP was...

In situ hybridization of dihydroxyacetone phosphate acyltransferase, the regulating enzyme involved in plasmalogen biosynthesis Inra
André, A.; Tessier, C.; Bretillon, L.; Sébédio, J.L.; Chardigny, J.M..
In situ hybridization can be carried out using different methods. The experimenter has to choose various parameters: the type of tissue fixation, the time of incubation, and the duration of the exposure time. All these parameters are determinant for the sensitivity and the resolution of this technique. This publication of technical aspects described different experiments performed for in situ hybridization on liver tissue. We may conclude on the parameters to optimize each step of the hybridization procedure. Moreover, this technique could be transposed to the brain and applied to little structures with a light expression of DHAP-AT.

Primary open-angle glaucoma: association with cholesterol 24S-hydroxylase (CYP46A1) gene polymorphism and plasma 24-hydroxycholesterol levels Inra
Fourgeux, C.; Lucy, M.; Björkhem, I.; Diczfalusy, U.; Joffre, C.; Acar, N.; Creuzot Garcher, C.; Bron, A.; Bretillon, L..
Purpose. Genetics has made significant contributions to the study of glaucoma over the past few decades. Cholesterol-24S-hydroxylase (CYP46A1) is a cholesterol-metabolizing enzyme that is especially expressed in retinal ganglion cells. CYP46A1 and its metabolic product, 24S-hydroxycholesterol, have been linked to neurodegeneration. A single-nucleotide polymorphism (SNP) in the CYP46A1 gene, designated as rs754203 and associated with Alzheimer disease, was evaluated as a genetic risk factor for primary open-angle glaucoma (POAG), as well as plasma 24S-hydroxycholesterol levels. Methods. The frequency of the CYP46*C and CYP46*T alleles was analyzed in 150 patients with POAG and 118 control subjects. Plasma 24S-hydroxycholesterol levels were quantified. Sex,...
